Total Saponin from Root of Actinidia Valvata Dunn Prevents the Metastasis of Human Hepatocellular Carcinoma Cells

Objective：To extract the active component from the root of Actinidia valvata Dunn and to investigate the effects on hepatocellular carcinoma（HCC） cells in ***：Total saponin was extracted from the root of ***（TSAVD）.HCC cells,such as BEL-7402,HepG2,PLC,SMMC-7721,MHCC-97-H, and MHCC-97-L,were treated with TSAVD in 3-（4,5-Dimethylthiazol-2-yl）-2,5-diphenytetrazolium bromide（MTT） ***-7402 and MHCC-97-H cells were also treated respectively with TSAVD at different concentrations for 24 h in wound healing and adhesion assays,and the effects of TSAVD on BEL-7402 and MHCC-97-H cells mobility and adhesion abilities were ***,the effects of TSAVD on invasion and migration of BEL-7402 and MHCC-97-H cells were also investigated by transwell chamber in invasion and migration assays. Results：TSAVD at 1.5 mg/mL inhibited BEL-7402 cell proliferation with inhibition ratios（IRs） of 61.08%,74.12%, 84.55%at 24,48,and 72 h,***,TSAVD inhibited MHCC-97-H proliferation in a concentrationdependent manner from 1.5 to 0.5 mg/mL,with the IR of 36%at 1.5 mg/mL at 24 *** SMMC-7721,PLC, and HepG2,the IR was lower than 30%at 1.5 mg/mL at 24 *** the wound healing assay,mobility abilities of BEL-7402 and MHCC-97-H cells in TSAVD treated groups were significantly weaker than those of the control *** pretreatment for 24 h with TSAVD,adhesion abilities were reduced in both MHCC-97-H and BEL-7402 cells,with IRs of 48.50%±4.86%and 49.85%±5.25%at 200 ｜xg/*** IRs of MHCC-97-H and BEL-7402 cells in the migration assay were 49.13%±2.91%and 79.37%±0.09%at 200μg/mL In the invasion assay,IRs were 69.78%±4.88%and 82.48%±0.25%at 200μg/mL Conclusions：Of all HCC cells,the highest inhibition by TSAVD was seen for BEL-7402 *** could restrain adhesion,invasion,mobility,and migration abilities of BEL-7402 and MHCC-97-H cells in vitro.

Burch colposuspension for stress urinary incontinence: a 14-year prospective follow-up

We aimed to evaluate the long-term effectiveness and safety of Burch colposuspension(BC) for stress urinary incontinence(SUI). In this prospective cohort study, 84 patients with SUI undergoing BC were enrolled from February 2004 to January *** on long-term subjective success and postoperative complications were collected at clinic visits and by telephone *** a mean follow-up period of 14.2 years, 68%(57/84) patients completed the follow-up. A total of 68.4% of patients(39/57) reported absence of SUI symptoms, 73.6%(42/57) were subjectively satisfied according to the Patient Global Impression of Improvement, and 68.4%(39/57) reported subjective success regarding urinary symptoms via the Urinary Distress Inventory Short Form. However, 28.1%(16/57) suffered at least one long-term postoperative complication and incident. Specifically, 1 in25(4.0%) sexually active patients reported dyspareunia, 3 patients(5.3%) had de novo overactive bladder, and 6 patients(10.5%)reported voiding dysfunction. Four patients(7.0%) reported new onset prolapse symptoms, and 3 patients(5.3%) underwent secondary urinary incontinence surgery. Our study indicated that Burch colposuspension is an effective procedure for SUI, and the cure effect was largely maintained for the 14-year follow-up period, with relatively low complication rates. BC should be considered a surgical option for SUI.

Comparison of urinary telomerase,CD44,and NMP22 assays for detection of bladder squamous cell carcinoma

Background:Squamous cell carcinoma(SCC)of the bladder is common in many regions around the *** is very poor,as most cases are diagnosed at an advanced stage due to a lack of affordable and valid screening markers for this type of *** diagnostic accuracy of urinary nuclear matrix protein-22(NMP22),telomerase activity,and CD44 were evaluated in urine samples of patients with bladder *** and methods:We conducted a case-control study comprised of 60 consecutive newly diagnosed bladder SCC patients diagnosed by cystoscopy and histopathological examination,and controls were 60 outpatients with benign urologic conditions and healthy clinic *** samples collected from each subject underwent testing for NMP22,telomerase activity,and *** and correlational statistical analysis of cases and controls were carried out and receiver operating characteristic curve analysis was used to determine optimal cut-off points for the three ***:Area under the curve was calculated at 0.96,0.93,and 0.62 for NMP22,telomerase,and CD44,*** levels of NMP22 and telomerase activity were significantly higher in the SCC group compared to controls(p<0.001).Urine CD44 levels were not significantly higher in the SCC group compared to controls(p=0.111).The overall sensitivity of NMP22,telomerase,and CD44 was 96.7%,87%,and 45%,respectively,while the specificity was 85%,88.6%,and 86.7%,***:Urinary telomerase activity,followed by NMP22 urine levels,showed high diagnostic yield and could hold potential promise as urinary biomarkers for the diagnosis of bladder SCC.

Introduction: Globally, tuberculosis is the leading cause of death from a single infectious agent ahead of HIV/AIDS. Approximately 10 million people contracted TB in 2017, 10% of whom were children aged 0 - 15 years, or about 1 million with 250,000 deaths in 2016 (including children with HIV-associated TB). The signs of TB in children are not always specific and diagnosis remains difficult unlike in adults. According to a study conducted in 2011 in the paediatric department of the CHU-Gabriel Touré, only seventeen cases of all forms of tuberculosis were found, or approximately 0.2% of hospitalised children. The objective of our study was to investigate the epidemiological and clinical aspects of tuberculosis in children. Materials and Method: This was a prospective, cross-sectional and descriptive study that took place from 24 October 2017 to 23 October 2018, or 12 months in children aged 0-15 years. Data were collected from an individual medical record opened for each patient and an individual survey form established for each child. Results: During the study period, 40,434 children were consulted. Tuberculosis was suspected in 91 children, with a frequency of 0.22%. The age range of 1 to 4 years was 36.3% with a median age of 72 months. The sex ratio was 1.8. Chronic cough with 84.6% and malnutrition with 24.17% were the most frequent symptoms. Chest X-ray revealed bilateral pulmonary lesions in 52.7% and mediastinal adenopathy in 12.1%. TST was positive in 10.9% of patients, microscopy in 26.4%, Gene Xpert in 18.7%, and culture in 16.5%. The biological diagnosis of tuberculosis was retained in 48.4% of the patients, the pulmonary form represented 93.2%. The therapeutic regime (2RHZE/4RH) was used in 81.6% of cases and the evolution was favourable in 65.9% of patients. Conclusion: The diagnosis of tuberculosis in children remains difficult in our context. The clinical signs are not always specific, and further studies are needed to further elucidate this disease.

BACKGROUND Hydroxyurea(HU)is a non-alkylating antineoplastic agent that is active in the Sphase of the cell cycle and inhibits the enzyme ribonucleoside *** is currently used to treat leukemia,sickle cell anemia,psoriasis,and chronic myeloproliferative *** HU is easy to use and effective and has high tolerance,there have been numerous reports of cutaneous complications during long-term therapy with *** SUMMARY We report a 67-year-old woman on long-term HU therapy for primary myelofibrosis who developed concurrent skin lesions during *** first skin lesion appeared on the dorsum of her right hand in *** continuous use of HU,her cutaneous changes were *** 3 years ago,she had multiple nodular and keratotic lesions on both hands with sharp margins,branny desquamation,and dotted ***,she developed acutely numerous ulcerative lesions on her hands and *** wound therapy with dressing changes and parenteral antibiotics was applied for management of the *** of the wounds healed after HU *** on both hands were replaced by ***,the wound on her left ankle reached 9 cm×7 cm in size in January *** confirmed welldifferentiated squamous cell carcinoma at the ulcer *** addition,her left foot was severely affected and radical surgery with a below-the-knee amputation was suggested followed by preventive right groin nodal *** In patients receiving continuous HU therapy,close dermatologic follow-up is critical for the early diagnosis and selection of appropriate treatment for cutaneous lesions.

Homeopathic potencies of Arnica montana L. change gene expression in a Tamm-Horsfall protein-1 cell line in vitro model: the role of ethanol as a possible confounder and statistical bias

Marzotto et al. showed that homeopathic preparations of Amica montana L. acted directly on gene expression of Tamm-Horsfall protein- 1 （THP-1） monocyte/macrophage cell lines activated with phorbol 12-mydstate 13- acetate and interleukin-4 （IL-4）. A. montana homeopathic dilutions are used in complementary and alternative medicine to treat inflammation disorders and post-traumatic events as well as for wound repair. The French Pharmacopoeia of these remedies uses 0.3% ethanol in each centesimal dilution. In this paper, we discuss how ethanol-containing A. montana homeopathic centesimal dilutions can change gene expression in IL-4- treated monocyte/macrophage THP-I. We assessed the role of ethanol in the Arnica homeopathic dilutions containing this alcohol by investigating its action on gene expression of THP-1 cell. Evidence would strongly suggest that the presence of ethanol in these remedies might play a fundamental role in the dilutions ability to affect gene expression, particularly for doses from 5c to 15c. Where, rather than playing a major role in the mesoscopic structure of water, the ethanol might have a chemical-physical role in the induction of THP-1 gene expression, apoptosis, and deoxyribonucleic acid function. This evidence generates a debate about the suggestion that the use of a binary-mixed solvent in homeopathic chemistry, used by Hahnemann since 1810, may be fundamental to explain the activity of homeopathy on cell models.

Cables composed of rare-earth barium copper oxide(REBCO)tapes have been extensively used in various superconducting *** recent years,conductor on round core(CORC)cable has drawn the attention of researchers with its outstanding current-carrying capacity and mechanical *** REBCO tapes are wound spirally on the surface of CORC *** extreme loadings,the REBCO tapes with layered composite structures are vulnerable,which can lead to degradation of critical current and even quenching of superconducting *** this paper,we simulate the deformation of CORC cable under external loads,and analyze the damage inside the tape with the cohesive zone model(CZM).Firstly,the fabrication and cabling of CORC are simulated,and the stresses and strains generated in the tape are extracted as the initial condition of the next ***,the tension and bending loads are applied to CORC cable,and the damage distribution inside the tape is *** addition,the effects of some parameters on the damage are discussed during the bending simulations.

Objective:The metastasis of tumor cells is a difficult point in the clinical treatment of ***-mesenchymal transition(EMT) contributes to glioma cells *** previous study confirmed that miRNA-451 is downregulated in gliomas,which can inhibit the PI3K/Akt signaling pathway by directly targeting CAB39 and has a repressive role in glioma by inhibiting cell growth,proliferation and by inducing cell ***,the specific regulatory mechanism is *** this study,we investigated whether mi RNA-451 blocked the process of EMT and metastasis in glioma cells in vivo and in ***:We used RT-PCR and Western blot experiments demonstrated p-AKT(ser-473),N-cadherin,Vimentin,Twist,Snail and Cyclin D1 and E-cadherin expression by over-expression of *** demonstrated glioma cells proliferation,invasion,migration and EMT process by MTT assay,Cell invasion and wound-healing assay and Animal experiments via over-expression of *** :We used RT-PCR and Western blot experiments demonstrated that over-expression of miRNA-451 significantly reduced p-AKT(ser-473),N-cadherin,Vimentin,Twist,Snail and Cyclin D1 expression and increased E-cadherin *** demonstrated that over-expression of miR-451 suppressed glioma cells proliferation,invasion,migration and EMT process by MTT assay,Cell invasion and wound-healing assay and Animal ***:miRNA-451 can inhibit EMT and metastasis in glioma cells through suppression of PI3K/AKT/Snail signaling pathway by targeting CAB39 in vitro and in vivo.

AIM: To investigate the effects of 17β-estradiol via estrogen receptors(ER) or direct administration of ER agonists on human colorectal cancer. METHODS: Lo Vo cells were established from the Bioresource Collection and Research Center and cultured in phenol red-free DMEM(Sigma, United States). To investigate the effects of E2 and/or ER selective agonists on cellular proliferation, Lo Vo colorectal cells were treated with E2 or ER-selective agonists for 24 h and 48 h and subjected to the MTT(Sigma) assay to find the concentration. And investigate the effects of E2 and/or ER selective agonists on cell used western immunoblotting to find out the diversification of signaling pathways. In order to observe motility and migration the wound healing assay and a transwell chamber(Neuro Probe) plate were tased. For a quantitative measure, we counted the number of migrating cells to the wound area post-wounding for 24 h. We further examined the cellular migration-regulating factors urokinase-type plasminogen activator(u-PA), tissue-type plasminogen activator(t-PA) and matrix metalloproteinase(MMP)-9 in human Lo Vo cells so gelatin zymography that we used and gelatinolytic activity was visualized by Coomassie blue staining. And these results are presented as means ± SE, and statistical comparisons were made using Student's ***: The structure was first compared with E2 and ER agonists. We then treated the Lo Vo cells with E2 and ER agonists(10-8 mol/L) for 24 h and 48 h and subsequently measured the cell viability using MTT assay. Our results showed that treatment with 17β-estradiol and/or ER agonists in human Lo Vo colorectal cancer cells activated p53 and then up-regulated p21 and p27 protein levels, subsequently inhibiting the downstream target gene, cyclin D1, which regulates cell proliferation. Taken together, our findings demonstrate the anti-tumorigenesis effects of 17β-estradiol and/or ER agonists and suggest that these compounds may prove to be a potential alte

Unusual complication in patient with Gardner's syndrome: Coexistence of triple gastrointestinal perforation and lower gastrointestinal bleeding: A case report and review of literature

Gardner's syndrome(GS) is a rare syndrome with autosomal dominant inheritance, which is characterized by multiple intestinal polyps, dental anomalies, desmoid tumors, and soft tissue tumors. All gastrointestinal symptoms seen in GS are associated with the underlying familial adenomatosis polyposis and abdominal desmoid tumors, with the most common symptoms being anemia, lower gastrointestinal bleeding, abdominal pain, diarrhea, obstruction, and mucous defecation. To our best knowledge, no case of GS that has presented with gastrointestinal perforation and bleeding has ever been reported in the English language medical literature. A 37-year-old male who had been diagnosed with GS five years earlier was referred to our clinic for lower gastrointestinal bleeding. Despite the absence of a bleeding focus on conventional angiography, the patient was operated on with laparotomy, due to the persistence of both signs and symptoms of mild peritonitis. On the laparotomy, the patient was noted to have areas of perforation in the duodenum, splenic flexura, and midrectum. The third and fourth part of the duodenum, the proximal 15 cm segment of the jejunum, a 10 cm segment of the terminal ileum, the whole colon, and the upper and middle rectum were resected, and duodenojejunal side-to-side anastomosis and terminal ileostomy were performed. The histopathological analysis of the large mass measuring 30 cm × 20 cm was reported as a desmoid tumor. The pathological examination of the tumor foci detected in the colonic specimen revealed poorly differentiated adenosquamous carcinoma.
