Construction and Characterization of a Novel Bacmid AcBac-Syn Based on a Synthesized Baculovirus Genome

Baculoviruses are large DNA viruses which have been widely used as expression vectors and biological *** recombination and Bac-to-Bac system have been the main methods for manipulating the baculovirus ***, we generated a synthetic baculovirus Ac MNPV-WIV-Syn1 which fully resembled its parental virus Autographa californica multiple nucleopolyhedrovirus(Ac MNPV). Here, we report the modification of Ac MNPV-WIV-Syn1 into a novel bacmid, Ac Bac-Syn, which can be used as a backbone for Bac-to-Bac system. To achieve this, a vector contained a Lac Z:att Tn7 and egfp cassette was constructed, and recombined with a linearized Ac MNPV-WIV-Syn1 genome by transformation-associated recombination in yeast to generate bacmid Ac Bac-Syn. The bacmid was then transfected to insect cells and the rescued virus showed similar biological characteristics to the wild-type virus in terms of the kinetics of budded virus production, the morphology of occlusion bodies, and the oral infectivity in insect larvae. For demonstration, a red fluorescent protein gene Dsred was transposed into the att Tn7 site by conventional Bac-to-Bac method, and the transfection and infection assays showed that Ac Bac-Syn can be readily used for foreign gene insertion and *** Bac-Syn has several advantages over the conventional Ac MNPV bacmids, such as it contains an egfp reporter gene which facilitates visualization of virus propagation and titration;its DNA copy numbers could be induced to a higher level in E. coli;and the retaining of the native polyhedrin gene in the genome making it an attractive system for studying the functions of gene related to occlusion body assembly and oral infection.

Antibacterial and Antioxidant Activities of ZnO Nanoparticles Synthesized Using Extracts of Allium sativum,Rosmarinus officinalis and Ocimum basilicum

Phospholipon 90H(P90H)-based PEGylated microscopic lipospheres delivery system for gentamicin:an antibiotic evaluation

Objective:To formulate gentamicin liposphere by solvent-melting method using lipids and polyethylene glycol 4000(PEG-4000) for oral ***:Gentamicin lipospheres were prepared by melt-emulsification using 30%w/w Phospholipon(?) 90H in Beeswax as the lipid matrix containing *** lipospheres were characterized by evaluating on encapsulation efficiency,loading capacity,change in pH and the release profile. Antimicrobial activities were evaluated against Escherichia coli,Pseudomonas aeruginosa. Salmonella paratyphii and Staphylococcus aureus using the agar diffusion ***: Photomicrographs revealed spherical particles within a micrometer range with minimal growth after 1 *** release of gentamicin in vitro varied widely with the PEC-4000 contents. Moreover,significant(P>0.05) amount of gentamicin was released in vivo from the formulation. The encapsulation and loading capacity were all high,indicating the ability of the lipids to take up the *** antimicrobial activities were very high especially against Pseudomonas compare to other test *** strongly suggested that the formulation retain its bioactive ***:This study strongly suggest that the issue of gentamicin stability and poor absorption in oral formulation could be adequately addressed by tactical engineering of lipid drug delivery systems such as lipospheres.

Segmental Kinematic Coupling of the Human Spinal Column during Locomotion

As one of the most important daily motor activities, human locomotion has been investigated intensively in recent decades. The locomotor functions and mechanics of human lower limbs have become relatively well understood. However, so far our understanding of the motions and functional contributions of the human spine during locomotion is still very poor and simultaneous in-vivo limb and spinal column motion data are scarce. The objective of this study is to investigate the delicate in-vivo kinematic coupling between different functional regions of the human spinal column during locomotion as a stepping stone to explore the locomotor function of the human spine complex. A novel infrared reflective marker cluster system was constrncted using stereophotogrammetry techniques to record the 3D in-vivo geometric shape of the spinal column and the segmental position and orientation of each functional spinal region simultaneously. Gait measurements of normal walking were conducted. The preliminary results show that the spinal column shape changes periodically in the frontal plane during locomotion. The segmental motions of different spinal functional regions appear to be strongly coupled, indicating some synergistic strategy may be employed by the human spinal column to facilitate locomotion. In contrast to traditional medical imaging-based methods, the proposed technique can be used to investigate the dynamic characteristics of the spinal column, hence providing more insight into the functional biomechanics of the human spine.

Circulating autoantibodies to endogenous erythropoietin are associated with chronic hepatitis C virus infection-related anemia

BACKGROUND： Chronic hepatitis C virus （HCV） infection is associated with autoimmune phenomena and is often complicated by anemia. Circulating autoantibodies to endogenous erythropoietin （anti-EPO） have been detected in patients with chronic viral infections and were correlated to anemia. The present study aimed to determine anti-EPO prevalence in pa- tients with chronic HCV infection and investigate its possible association with anemia. METHODS： Ninety-three consecutive patients （62 males and 31 females） with chronic HCV infection, who had never re- ceived antiviral therapy or recombinant EPO, were enrolled in the study. Circulating anti-EPO were detected in the serum by using an ELISA assay. Quantitative determination of serum EPO levels was done by radioimmunoassay. HCV RNA viral load t and genotype sequencing were also performed. RESULTS： Circulating anti-EPO were detected in 10.8% of HCV-infected patients and the prevalence of anti-EPO was significantly higher in patients with anemia （19.4% vs 5.3%, P=0.040） compared to that in those without anemia. Compared to anti-EPO negative cases, anti-EPO positive patients had higher frequency of anemia （70.0% vs 34.9%, P=0.030）, lower EPO concentrations （median 16.35 vs 30.65 mU/mL, P=0.005）, and higher HCV RNA viral load （median 891.5x103 vs 367.5x 103 IU/mL, P=0.016）. In multivariate regression anal- ysis the presence of anti-EPO remained an independent predictor of anemia （adjusted OR： 14.303, 95% CI： 1.417-36.580, P=0.024）. EPO response to anemia was less prominent among anti-EPO positive patients （P=0.001）. CONCLUSIONS： Circulating anti-EPO are detected in a significant proportion of treatment-naive HCV-infected patients and are independently associated with anemia, suggesting a further implication of autoimmunity in the pathophysiology of HCV-related anemia.

Isolation and species identification of enterococci from clinical specimen with their antimicrobial susceptibility pattern in a tertiary care hospital,Bangladesh

Objective:To investigate the species prevalence of Enterococcus with their antimicrobial resistance pattern from patients of Dhaka Medical College ***:Samples were cultured and Enterococcus species were identified by conventional biochemical tests as well as PCR by using species specific primers for Enterococcus faecalis(***)and Enterococcus faecium(***).For isolation of vancomycin resistant enterococci,minimum inhibitory concentration of vancomycin and PCR was done to detect vanA and vanB ***:A total of 16 enterococci were isolated from 300 urine and 200 wound swab samples(15 from urine and 1 from wound swab)from July 2011 to June *** were the third most common organism(8.47%)from urine after Escherichia coli(63.28%)and Enterobacter(11.87%).Out of 16 enterococci,10(62.5%)were ***,4(25%)were *** and 2(12.5%)were other *** the enterococci(100%)were sensitive to vancomycin and *** of the strains were resistant to ciprofloxacin and azithromycin(87.5%),gentamycin(81.25%),ceftriaxone(75%),amoxiclav(31.25%)and imipenem(25%).*** was more resistant than *** to azithromycin(100%),ciprofloxacin(100%),amoxiclav(75%)and imipenem(50%).No vancomycin resistant enterococci were identified and the range of minimum inhibitory concentration for vancomycin was 1-4μg/*** of the enterococci were positive for vanA and vanB ***:The presence of multidrug resistant enterococci should be considered as danger alarm for serious enterococcal infections and further study in large scale is needed.

Interleukins and Acute Phase Proteins of Bovine Sera during Natural Helminth Burden in Ibadan Nigeria

Inflammatory reactions in the gastrointestinal tract play an important role in the pathogenesis of gastroenteritis in bovine helminthosis. This study determined the serum concentrations of cytokines induced acute phase proteins (Serum Amyloid A (SAA), Haptoglobin (Hp) and C-reactive protein (CRP)) and the levels of immunoreactive interleukins (IL-1, IL-2, IL-3, IL-4, IL-5 and IL-6) in cattle naturally infected with helminths. We sampled a total of 480 slaughtered cattle of both sexes in a major abattoir in Ibadan, Nigeria. Sedimentation, floatation and modified McMaster techniques were employed to determine the degrees of helminth infections. Animals with eggs per gram (epg) less than 200 were adjudged to be apparently healthy. The serum concentrations of interleukins and acute phase proteins were determined using the Technicon AutoAnalyzer Model AA II. It was found that mean SAA (μg/l) and CRP (%) levels were significantly (P 0.05). Therefore, some acute phase proteins are involved in the pathophysiology of bovine helminthosis and are closely related to the inflammatory activation of the disease. In lieu of these findings, it is suggested that systemic markers of inflammation can identify subjects at high risk of natural bovine helminthosis and that IL-6 and SAA may be used as indicators for bovine helminthosis.
