Mesenchymal stem cells and the neuronal microenvironment in the area of spinal cord injury

Cell-based technologies are used as a therapeutic strategy in spinal cord injury(SCI). Mesenchymal stem cells(MSCs), which secrete various neurotrophic factors and cytokines, have immunomodulatory, anti-apoptotic and anti-inflammatory effects, modulate reactivity/phenotype of astrocytes and the microglia, thereby promoting neuroregeneration seem to be the most promising. The therapeutic effect of MSCs is due to a paracrine mechanism of their action, therefore the survival of MSCs and their secretory phenotype is of particular importance. Nevertheless, these data are not always reported in efficacy studies of MSC therapy in SCI. Here, we provide a review with summaries of preclinical trials data evaluating the efficacy of MSCs in animal models of SCI. Based on the data collected, we have tried(1) to establish the behavior of MSCs after transplantation in SCI with an evaluation of cell survival, migration potential, distribution in the area of injured and intact tissue and possible differentiation;(2) to determine the effects MSCs on neuronal microenvironment and correlate them with the efficacy of functional recovery in SCI;(3) to ascertain the conditions under which MSCs demonstrate their best survival and greatest efficacy.

Ultrasound guided testicular fine needle aspiration in buck (Capra hircus)-An animal model

Objective:To establish ultrasound guided testicular fine needle aspiration (TFNA) as well as to assess the effectiveness of uni-directional (UD) and multi-directional (MD) TFNA in buck according to testicular cells, echotexture and gross changes of testicle, age of buck was considered.Methods:A total of 120 samples were collected with both directions (UD, n=60) and (MD,n=60) suction from testis of 10 apparently healthy bucks. All slides were stained with May-Grünwald-Giemsa and examined under light microscope with 1000× magnifications to count spermatogenic cells, spermatozoa and sertoli cells. The percentage of spermatozoa and sertoli cells were expressed as spermatic index and sertoli cell index.Results:Results revealed no difference in the presence of various spermatogenic and sertoli cells in cell cluster of slides made either unidirectional TFNA or multidirectional TFNA. Early spermatids were the most numerous, followed by late spermatids, primary spermatocytes, spermatogonia. Sertoli cell index was higher in TFNA smears of young bucks prepared from (7-13 mo) of age and spermatic index was higher in adult bucks (14-24 mo) of age. No echogenic change was observed in the echotexture of testisafter TFNA.Conclusion: It seems that TFNA has no serious ill effect on the buck testis when uni-direction aspiration is performed. Moreover, the possibility to standardize this method might provide a greater impulse to the clinical diagnostics of male animal infertility.

Every year there are new 1,600 cases of breast cancer in Bosnia and Herzegovina. The most effective method for early detection of breast cancer is mammography. To examine risks and benefits of this diagnostic method it is necessary to determine patient doses. Mainly, almost all published data about patient doses refer to two specific age groups: 40-49 and 50-64 years of age. Very little data about patient doses applied during a routine mammography for patients from 65-80 years of age are available. During the conducted research, one calculated doses for the complete mammographic examination of 42 patients from this age group. The calculated patient doses are related to the whole spectrum of technical, physical, clinical and diagnostic parameters which compose a complete mammographic examination.

Dear Editor,Great progress has been made in the clinical use of photodynamic therapy(PDT)for the treatment of patients with superficial tumors[1].However,cancer recurrence and metastasis have limited the application of PDT in the treatment of solid tumors and advanced cancers.In this context,combining PDT with other complementary immunotherapy regimens may overcome these limitations of PDT[2].

The main therapeutic options for colorectal cancer are surgical resection and adjuvant chemotherapy in non-metastatic disease.However,the evaluation of the overall adjuvant chemotherapy benefit in patients with a high risk of recurrence is challenging.Radiological images can represent a source of data that can be analyzed by using automated computer-based techniques,working on numerical information coded within Digital Imaging and Communications in medicine files:This image numerical analysis has been named“radiomics”.Radiomics allows the extraction of quantitative features from radiological images,mainly invisible to the naked eye,that can be further analyzed by artificial intelligence algorithms.Radiomics is expanding in oncology to either understand tumor biology or for the development of imaging biomarkers for diagnosis,staging,and prognosis,prediction of treatment response and diseases monitoring and surveillance.Several efforts have been made to develop radiomics signatures for colorectal cancer patient using computed tomography(CT)images with different aims:The preoperative prediction of lymph node metastasis,detecting BRAF and RAS gene mutations.Moreover,the use of delta-radiomics allows the analysis of variations of the radiomics parameters extracted from CT scans performed at different timepoints.Most published studies concerning radiomics and magnetic resonance imaging(MRI)mainly focused on the response of advanced tumors that under-went neoadjuvant therapy.Nodes status is the main determinant of adjuvant chemotherapy.Therefore,several radiomics model based on MRI,especially on T2-weighted images and ADC maps,for the preoperative prediction of nodes metastasis in rectal cancer has been developed.Current studies mostly focused on the applications of radiomics in positron emission tomogra-phy/CT for the prediction of survival after curative surgical resection and assessment of response following neoadjuvant chemoradiotherapy.Since colorectal liver metastases develop in about 25%of patients with colorecta

Transcatheter hepatic artery chemoembolization(TACE)is a universal treatment for patients with hepatocellular carcinoma(HCC)that inhibits tumor growth by cutting off the blood supply and provides chemotherapeutics locally to the tumor.The strategy of combining TACE formulation with image-guided ablation holds tremendous potential,but patient tolerance and undesired toxicity/immunosuppression remains a challenge.The application of nanotechnology in TACE opens new doors for the treatment of HCC.Strikingly,nanomaterials or nano-drugs dispersed in the TACE formulation can effectively improve the delivery efficiency of drugs by achieving both controlled and continuous release.In addition,the utilization of multifunctional nanoparticles can provide guidance and monitoring for various advanced imaging methods for TACE treatment,and can realize the combination therapy of thermal ablation,microwave ablation,in situ radiotherapy,and other therapies,greatly expanding the therapeutic strategies available for HCC treatment.Here,the current exploration of nanotechnology in TACE of HCC is briefly summarized and the challenges of TACE with nanoformulations for clinical translation are comprehensively discussed.

Epstein-Barr virus-associated diseases are important global health concerns.As a group I carcinogen,EBV accounts for 1.5%of human malignances,including both epithelial-and lymphatic-originated tumors.Moreover,EBV plays an etiological and pathogenic role in a number of non-neoplastic diseases,and is even involved in multiple autoimmune diseases(SADs).In this review,we summarize and discuss some recent exciting discoveries in EBV research area,which including DNA methylation alterations,metabolic reprogramming,the changes of mitochondria and ubiquitin-proteasome system(UPS),oxidative stress and EBV lytic reactivation,variations in non-coding RNA(ncRNA),radiochemotherapy and immunotherapy.Understanding and learning from this advancement will further confrm the far-reaching and future value of therapeutic strategies in EBV-associated diseases.

Background Foreign bodies(FBs)lodged in the intestine or causing intestinal complications are uncommon in clinical practice but may pose diagnostic difficulties and prove life-threatening.This study aimed to evaluate the risk factors for severe complications and surgery to aid clinicians in the diagnosis and management of intestinal FBs.Methods We performed a retrospective analysis of patients in whom FBs were lodged in the intestine or caused complications from 2010 to 2020 in the First Affiliated Hospital of Wenzhou Medical University(Zhejiang,China).The characteristics of the patients and FBs,symptoms,imaging findings,diagnostics,treatment strategies,and clinical outcomes were analysed.Furthermore,the risk factors for complications and surgery were investigated.Results In total,180 patients were included in our study.Most patients(76.1%)were unable to provide a history of ingestion.Bezoars were the most common FBs(35.6%).The FBs were mainly located in the duodenum(32.8%)and the ileum(27.8%).Surgical removal of FBs was successful in 89(49.4%)patients and endoscopic removal in 54(30.0%)patients.Eleven with perforations were treated conservatively.FBs located in the jejunum or ileum were more likely to cause severe complications than those located in the duodenum.FBs located in the jejunum,ileum,or sigmoid colon were more likely to undergo surgery,and severe complications were an independent risk factor for surgery.Conclusion Intestinal FBs,often localized in angulation,are likely to be misdiagnosed because most patients do not provide a history of FB ingestion.Surgery and endoscopic therapy are the most commonly used treatment modalities.Surgery is not mandatory in clinically stable patients with small and contained perforations.FBs located in the jejunum or ileum are risk factors for both complications and surgery.

Latent membrane protein 1(LMP1)is a major Epstein–Barr virus(EBV)-encoded oncoprotein involved in latency infection that regulates mitochondrial functions to facilitate cell survival.Recently,mitochondrial fission has been demonstrated as a crucial mechanism in oncovirus-mediated carcinogenesis.Mitochondrial dynamin-related protein 1(Drp1)-mediated mitochondrial fission has an impact on the chemoresistance of cancers.However,the mechanism by which oncogenic stress promotes mitochondrial fission,potentially contributing to tumorigenesis,is not entirely understood.The role of Drp1 in the oncogenesis and prognosis of EBV-LMP1-positive nasopharyngeal carcinoma(NPC)was determined in our study.We show that EBV-LMP1 exhibits a new function in remodeling mitochondrial morphology by activating Drp1.A high level of p-Drp1(Ser616)or a low level of p-Drp1(Ser637)correlates with poor overall survival and disease-free survival.Furthermore,the protein level of p-Drp1(Ser616)is related to the clinical stage(TNM stage)of NPC.Targeting Drp1 impairs mitochondrial function and induces cell death in LMP1-positive NPC cells.In addition,EBV-LMP1 regulates Drp1 through two oncogenic signaling axes,AMPK and cyclin B1/Cdk1,which promote cell survival and cisplatin resistance in NPC.Our findings provide novel insight into the role of EBV-LMP1-driven mitochondrial fission in regulating Drp1 phosphorylation at serine 616 and serine 637.Disruption of Drp1 could be a promising therapeutic strategy for LMP1-positive NPC.
