Modeling the modulation of beta oscillations in the basal ganglia by dual-target optogenetic stimulation

Optogenetic techniques provide precise control over the activity of specific neurons within the nucleus,offering more accurate regulatory effects compared to deep brain *** heterogeneity of the globus pallidus externa(GPe)has garnered wide attention,wherein significant differences in pathological changes emphasize its potential as a stimulation target with distinct mechanisms.A basal ganglia-thalamus(BG-Th)network model incorporating heterogeneous GPe is developed to explore potential optogenetic stimulation targets for treating Parkinson's disease(PD).Initially,the modulation mechanisms of single-target optogenetic stimulation on the abnormal rhythmic oscillations of BG nuclei are *** of D1 medium spine neuron(MSN),calcium-binding protein parvalbumin(PV)GPe,and inhibition of globus pallidus interna(GPi)can effectively suppress synchronous bursting activity in GPi,while excitation of GPi promotes high-frequency discharge to disrupt beta ***,dual-target optogenetic stimulation strategies are devised to reduce energy *** show that targets with similar mechanisms exhibit additive effects,whereas targets with opposing mechanisms lead to *** underlying effective mechanisms of dual-target strategies are:enhancing the inhibitory input to GPi thus inhibiting the activity of GPi,or disrupting beta oscillations by restoring high-frequency discharges in *** strategy composed of exciting D1 MSN and inhibiting GPi requires the minimum total light intensity among single-target and dual-target strategies in our ***,simultaneously enhancing PV GPe and inhibiting D2 MSN achieves the greatest reduction in total energy consumption(40.8%reduction),compared to only enhancing PV *** findings unveil effective circuit mechanisms of optogenetic stimulation and provide novel insights for designing precise regulatory strategies for PD.

Tensor Decomposition-assisted Multiview Subgroup Analysis

To learn the subgroup structure generated by multidimensional interaction, we propose a novel multiview subgroup integration technique based on tensor decomposition. Compared to the traditional subgroup analysis that can only handle single-view heterogeneity, our proposed method achieves a greater level of homogeneity within the subgroups, leading to enhanced interpretability and predictive power. For computational readiness of the proposed method, we build an algorithm that incorporates pairwise shrinkage-encouraging penalties and ADMM techniques. Theoretically, we establish the asymptotic consistency and normality of the proposed estimators. Extensive simulation studies and real data analysis demonstrate that our proposal outperforms other methods in terms of prediction accuracy and grouping consistency. In addition, the analysis based on the proposed method indicates that intergenerational care significantly increases the risk of chronic diseases associated with diet and fatigue in all provinces while only reducing the risk of emotion-related chronic diseases in the eastern coastal and central regions of China.

Segmented Linear Regression Trees

Tree-based models have been widely applied in both academic and industrial settings due to the natural interpretability, good predictive accuracy, and high scalability. In this paper, we focus on improving the single-tree method and propose the segmented linear regression trees(SLRT) model that replaces the traditional constant leaf model with linear ones. From the parametric view, SLRT can be employed as a recursive change point detect procedure for segmented linear regression(SLR) models,which is much more efficient and flexible than the traditional grid search method. Along this way,we propose to use the conditional Kendall's τ correlation coefficient to select the underlying change points. From the non-parametric view, we propose an efficient greedy splitting method that selects the splits by analyzing the association between residuals and each candidate split variable. Further, with the SLRT as a single-tree predictor, we propose a linear random forest approach that aggregates the SLRTs by a weighted average. Both simulation and empirical studies showed significant improvements than the CART trees and even the random forest.

Modeling the boundary layer flow of ternary hybrid nanofluids is important for understanding and optimizing their thermal performance,particularly in applications where enhanced heat transfer and fluid dynamics are *** study numerically investigates the boundary layer flow of alumina-copper-silver/water nanofluid over a permeable stretching/shrinking sheet,incorporating both first and second-order velocity *** mathematical model is solved in MATLAB facilitated by the bvp4c function that employs the finite difference scheme and Lobatto IIIa *** solver successfully generates dual solutions for the model,and further analysis is conducted to assess their *** findings reported that only one of the solutions is *** the shrinking sheet case,increasing the first-order velocity slip delays boundary layer separation and enhances heat transfer,while,when the sheet is stretched,the second-order velocity slip accelerates separation and improves heat *** layer separation is most likely to occur when the sheet is shrinking;however,this can be controlled by adjusting the velocity slip with the inclusion of boundary layer suction.

The dyadic representation of any singular integral operator,as an average of dyadic model operators,has found many *** for many purposes it is enough to have such a representation for a“suitable class”of test functions,we show that,under quite general assumptions(essentially minimal ones to make sense of the formula),the representation is actually valid for all pairs(f,g)∈L^(p)(R^(d))×L^(p')(R^(d)),not just test functions.

Developing the Allometric Equations for Estimating Above-Ground Biomass of Vachellia tortilis Using Destructive Sampling in Oman

Biomass models to estimate carbon stocks in arid environment are very limited. This study employed destructive sampling to develop a new biomass model for Vachellia tortilis, a widely known species in the Sultanate of Oman. Twenty trees with a diameter at stump height (DSH) ranging from 18.5 cm to 150 cm were selected based on DSH and height variations for destructive sampling in As Saleel Natural Park Reserve (SNPR) in Al Sharqiyah governorate, South of Oman. Each tree was excavated and cut into three parts: Stems, Branches, twigs, and leaves. The total fresh weight of each tree was obtained in the field using a 300 balance. Sub-samples (250 - 300 grams) were taken from each part of the tree and transferred to the laboratory for dry weight determination. Linear multiple regression analysis was done using SPSS software between the three variables, DSH, H, CA (x) and the total dry biomass (y). Five models were tested for the best-fit model based on R-Square and Mean Square Error (MSE). Model 5 was the best-fit model, including the LOG of DSH and the LOG of CA (R2 = 0.97, MSE = 0.114). The models developed in this research fill a critical gap in estimating the AGB of terrestrial native species in Oman and other countries with similar ecological and climate conditions.

The authors consider the problem of reaching consensus over a communication network via asynchronous interaction between pairs of agents. A well-known method is the linear gossip algorithm due to Tsitsiklis(1984). Extension of this, allowing the selection of a strictly stationary sequence of communicating pairs, was given in Picci and Taylor(2013). Extension of the linear gossip algorithm to directed communication networks, retaining the linear dynamics, was proposed by Cai and Ishii(2012),later extended by Silvestre, et al.(2018). A definite novelty of these algorithms is that L2-convergence with exponential rate can be established. The authors attend the above issues, extending the result of Picci and Taylor(2013) motivated by features of algorithms for directed networks. The authors present and discuss the algorithm of Silvestre, et al.(2018), together with systematic simulation results based on 5M randomly chosen parameter settings. The core of the proposed mathematical technology is a set of simple observations, presented with a tutorial aspect, by which the authors can conveniently establish various results on the almost sure convergence of products of strictly stationary sequences of matrices to a rank-1 matrix.

Objective: To examine the effectiveness of Chinese medicine(CM) Lianhua Qingwen Granule(LHQW) and Jingyin Gubiao Prescription(JYGB) in asymptomatic or mild patients with Omicron infection in the shelter hospital. Methods: This single-center retrospective cohort study was conducted in the largest shelter hospital in Shanghai, China, from April 10, 2022 to May 30, 2022. A total of 56,244 asymptomatic and mild Omicron cases were included and divided into 4 groups, i.e., non-administration group(23,702 cases), LHQW group(11,576 cases), JYGB group(12,112 cases), and dual combination of LHQW and JYGB group(8,854 cases). The length of stay(LOS) in the hospital was used to assess the effectiveness of LHQW and JYGB treatment on Omicron infection. Results: Patients aged 41–60 years, with nadir cycle threshold(CT) value of N gene <25, or those fully vaccinated preferred to receive CM therapy. Before or after propensity score matching(PSM), the multiple linear regression showed that LHQW and JYGB treatment were independent influence factors of LOS(both P<0.001). After PSM, there were significant differences in LOS between the LHQW/JYGB combination and the other groups(P<0.01). The results of factorial design ANOVA proved that the LHQW/JYGB combination therapy synergistically shortened LOS(P=0.032). Conclusions: Patients with a nadir CT value <25were more likely to accept CM. The LHQW/JYGB combination therapy could shorten the LOS of Omicron-infected individuals in an isolated environment.
