Electroacupuncture enhances the mitophagy of granulosa cells in premature ovarian insufficiency model mice by inactivating the hippo-yes-associated protein/transcriptional co-activator with postsynaptic density protein,drosophila disc large tumor suppresso

OBJECTIVE: To investigate the potential mechanism of electroacupuncture（EA） in alleviating premature ovarian insufficiency（POI） and to provide a theoretical basis for EA treatment of POI. METHODS: For this purpose, a POI mice model was developed by injecting 12 mg/kg busulfan and 120 mg/kg cyclophosphamide intraperitoneally to induce POI. It was then proceeded by EA intervention at Guanyuan（CV4） acupoint on the second day following modeling. Similarly, apoptosis in ovarian granulosa cells was detected by terminal deoxynucleotidyl transferase d UTP nick end labeling staining, while enzyme-linked immunosorbent assay was employed for measuring serum folliclestimulating hormone（FSH）, luteinizing hormone（LH）, estrogen（E2）, and anti-müllerian hormone（AMH） levels. Moreover, transmission electron microscopy（TEM） was employed for examining mitochondrial morphology, while autophagy and hippo-yes-associated protein/transcriptional co-activator with postsynaptic density protein, drosophila disc large tumor suppressor, and zonula occludens-1 protein binding motif（YAP/TAZ） pathway related protein levels in ovarian tissue were detected via Western blotting. RESULTS: Analysis of serum levels of various hormones indicated that serum FSH and LH were reduced in EA compared to the POI group, while E2and AMH levels were found to be elevated in EA compared to the POI group. The EA was found to inhibit apoptosis in granulosa cells in POI model mice, in addition to improved mitochondrial damage and significantly improved mitophagy. Pathway analysis revealed that EA was involved in activating the hippo-YAP/TAZ pathway, followed by reversing EA effects on granulosa cell apoptosis and mitophagy with the use of verteporfin, an autophagy and YAP-T-cell factor/enhancer of split and activator of transcription domain family member interaction inhibitor. Tianxiangdan(TXD) alleviates myocardial ischemia reperfusion-induced ferroptosis through the activation of estrogen receptor alpha(ERα)

Tianxiangdan（TXD）, a traditional chinese herbal remedy, demonstrates efficacy in mitigating myocardial ischemia-reperfusion（I/R）-induced damage. This study employed network pharmacology to evaluate the therapeutic targets and mechanisms of TXD in treating I/R. Highperformance liquid chromatography-mass spectrometry（HPLC-MS） identified 86 compounds in TXD. Network pharmacological analysis predicted potential target genes and their modes of action. Cardiac function, ischaemic ST changes, lactate dehydrogenase（LDH）,malondialdehyde（MDA）, superoxide dismutase（SOD） activity, myocardial fiber, and infarct size were assessed using in vivo and in vitro I/R injury models. Estrogen receptor alpha（ERα）protein expression and estradiol（E2） levels were measured to confirm TXD's impact on estrogen levels and ERα expression. To examine if TXD reduces I/R injury through ERα, an AZD group(300 nmol·L-1AZD9496 and 15% TXD serum) was compared to a TXD group（15% TXD serum）. The study hypothesized that TXD upregulates the ERα-mediated iron metamorphosis pathway. I/R injury-induced ferroptosis was identified using a Fer-1 group(1.0 μmol·L-1Fer-1 and 15% TXD serum) to elucidate the potential association between ferroptosis and ERαproteins. A DCFH-DA probe detected reactive oxygen species（ROS） and Fe2+, while Western blotting assessed target protein expression. Both in vitro and in vivo experiments demonstrated that TXD attenuated I/R injury by reducing elevated ST-segment levels, improving cardiac injury biomarkers（LDH, MDA, and SOD）, alleviating pathological features, and preventing I/R-induced loss of cell viability in vitro. The effects and mechanisms of TXD on I/R injury-associated ferroptosis were investigated using I/R-induced H9c2 cells. AIM:To analyze whether alterations of voxel mirror homology connectivity(VMHC)values,as determined by resting-state functional magnetic resonance imaging(rsfMRI),occur in cerebral regions of patients with hypertensive retinopathy(HR)and to determine the relationship between VMHC values and clinical characteristics in patients with ***:Twenty-one patients with HR and 21 agematched healthy controls(HCs)were assessed by rsfMRI *** functional connectivity between the hemispheres of the cerebrum was assessed by measuring VMHC,with the ability of VMHC to distinguish between the HR and HC groups assessed using receiver operating characteristic(ROC)curve *** in the demographic and clinical characteristics of the HR and HC groups were analyzed by independent sample *** relationship between average VMHC in several brain areas of HR patients and clinical features was determined using Pearson correlation ***:Mean VMHC values of the bilateral cuneus gyrus(BA19),bilateral middle orbitofrontal gyrus(BA47),bilateral middle temporal gyrus(BA39)and bilateral superior medial frontal gyrus(BA9)were lower in the HR than in the HC ***:VMHC values can predict the development of early HR,prevent the transformation of hypertensive microangiopathy,and provide useful information explaining the changes in neural mechanism associated with HR.

Plants have mechanisms to transport secondary metabolites from where they are biosynthesized to the sites where they function,or to sites such as the vacuole for ***,current research has mainly focused on metabolite biosynthesis and regulation,and little is known about their ***,a class diterpenoid with medicinal properties,is biosynthesized in the periderm of Salvia miltiorrhiza ***,we discovered that tanshinone can be transported out of peridermal cells and secreted into the soil environment and that the ABC transporter SmABCG1 is involved in the efflux of tanshinoneⅡA and tanshinoneⅠ.The SmABCG1 gene is adjacent to the diterpene biosynthesis gene cluster in the *** *** temporal–spatial expression pattern of SmABCG1 is consistent with tanshinone accumulation ***1 is located on the plasma membrane and preferentially accumulates in the peridermal cells of *** *** expression in Xenopus laevis oocytes demonstrated that SmABCG1 can export tanshinoneⅡA and tanshinoneⅠ.CRISPR/Cas9-mediated mutagenesis of SmABCG1 in *** hairy roots resulted in a significant decrease in tanshinone contents in both hairy roots and the culture medium,whereas overexpression of this gene resulted in increased tanshinone ***76AH3 transcript levels increased in hairy roots overexpressing SmABCG1 and decreased in knockout lines,suggesting that SmABCG1 may affect the expression of CYP76AH3,indirectly regulating tanshinone ***,tanshinoneⅡA showed cytotoxicity to Arabidopsis *** findings offer new perspectives on plant diterpenoid transport and provide a new genetic tool for metabolic engineering and synthetic biology research.
