Activity estimation in radioimmunotherapy using magnetic nanoparticles

Objective： Estimation of activity accumulated in tumor and organs is very important in predicting the response of radiopharmaceuticals treatment. In this study, we synthesized ~77Lutetium （177Lu）-trastuzumabiron oxide nanoparticles as a double radiopharmaceutical agent for treatment and better estimation of organ activity in a new way by magnetic resonance imaging （MRI）. Methods： ^177Lu-trastuzumab-iron oxide nanoparticles were synthesized and all the quality control tests such as labeling yield, nanoparticle size determination, stability in buffer and blood serum up to 4 d, immunoreactivity and biodistribution in normal mice were determined. In mice bearing breast tumor, liver and tumor activities were calculated with three methods： single photon emission computed tomography （SPECT）, MRI and organ extraction, which were compared with each other. Results： The good results of quality control tests （labeling yield： 61%±2%, mean nanoparticle hydrodynamic size： 41±15 nm, stability in buffer： 86%±5%, stability in blood serum： 80%±3%, immunoreactivity： 80%±2%） indicated that ^177Lu-trastuzumab-iron oxide nanoparticles could be used as a double radiopharmaceutical agent in mice bearing tumor. Results showed that ^177Lu-trastuzumab-iron oxide nanoparticles with MRI had the ability to measure organ activities more accurate than SPECT. Conclusions： Co-conjugating radiopharmaceutical to MRI contrast agents such as iron oxide nanoparticles may be a good way for better dosimetry in nuclear medicine treatment.

Preparation, characterization and release of methyl viologen from a novel nanoparticle delivery system with double shells of silica and PLGA

The use of nanotechnology in drug delivery is a rapidly expanding field. Biodegradable or nontoxic nanomaterials have the most promising application potentials in nanomedicine. Herein, we report a novel core-shell nanoparticle with double shell coatings (silica and poly(D,L-lactide-co-glycolide) (PLGA)) with the total shell thickness of (8.7 ± 1.3) nm. The outer shell of PLGA is biodegradable and used for controlled and sustained release, and the inner shell of silica is mesoporous for the preservation of the chemical radiation therapeutic of methyl viologen (MV), an oxidant that produces reactive oxygen species during cancer radiation therapy. The dissolution time course data and transmission electron microscopy images showed that the novel nanoparticles (Au@SiO2&PLGA) have been successfully prepared, and silica and PLGA coated well the gold (Au) template surfaces. Nano- capsules (MV@SiO2&PLGA) were obtained after the gold templates were dissolved using sodium cyanide. The sustained release property was characterized through detecting fluorescence quenching time course of fluorescent isothiocyanate after mixing with MV@SiO2&PLGA nanocapsules that encapsulate MV molecules. The sustained release of MV molecules could be extended to approximately four weeks. This novel delivery system has high potential in future application for the delivery of therapeutic drugs, particularly for the treatment of cancer by radiation therapy.

Differential and total cross sections of the γp→π0p and the γd→π0pn reaction were measured for photon energies between 640 and 1 150 MeV. The data have been compared with SAID and MAID model calculations. π0 photoproduction on the deuteron shows a strong suppression of the 2nd resonance which is clearly seen in the γp→π0p reaction.

EFFECTS AND MECHANISM OF STRESS AND OF CRF MICROINJECTION INTO PVN ON GASTRIC EMPTYING

Objective To study the effects of wrap-restraint stress and of CRF or CRF antagonist (a-belieal-CRF9-41 ) microinjection on gastric emptying, and the role of plasma motilin (Mot ). Methods The effects of wrap- restraint stress or CRF and CRF antagonist microinjection into paraventricular nucleus (PVN) on gastric empiring were determined by using dextran blue-2000 as a marker. The plasma level of Mot was determined by radioimmunoassay. Results Wrap-restraint stress inhibited gastric emptying. Microinjection of CRF into PVN inhibited gastric emptying dose-dependently. In normal condition, a-he- lical-CRF9-4l bilaterally microinjected into PVN didn’ t modify gastric emptying; however, a-belieal-CRF9-41 microinjection bilaterally into PVN 15min before wrap-restraint stress could significantly antagonize the effect of stress on gastric emptying. After 30min of stress or CRF microinjection into PVN, plasma levels of Mot significantly decreased, as compered with the normal control group. Conclusion Wrap-restraint stress can inhibit gastric emptying, and microinjection of CRF into PVN has the similar effect. The PVN is one of the speific sites for the action of CRF affecting on gastric emptying. Plasma Mot might partic ipate in the effects of stress and CRF-induced alteration of gastric emptying.

THE CHANGES OF PANCREATIC ACINAR CELL FUNCTION IN ACUTE NECROTIZING PANCREATITIS OF RATS

ObjectiFe To evaluate the changes of pancreatic acinar cell functions in the rats with acutenecrotizing pancreatitis (ANP). methods Seventy SD rats were randomized into two groups: experimental group(n=35) and control group (n=35). To prepare the experimental model, the retrograde injection of 5% sodiumtaurocholate into the pancreatic duct was used for inducing ANP. Radioactive tracing by L -3H-phenylalanineand autoradiography were performed for scoring the differences of changes of amino acid uptake, enzyme-proteinsynthesis and output from acinar cells in rats between both groups. Results No changes were observed in aminoacid uptake and enzyme -protein synthesis in rats with dotted and haemorrhagic necrotizing foci as compared withcontrol group. However, accumulated zymogen granules in the interstitial of acinar cells were seen in theexperimental group. Conclusion It indicates that in experimental ANP rats, the functions of acinar cells in bothamino acid uptake and protein synthesis were essentially normal, but the pathway of enzyme output was affectedinto ectopic secretion through the bottom or lateral cellular membrane of pancreatic acinar cell.

This paper presents the results ofI-Tplasma binding ratio test for assessment of thyroid function, the test was done in 160 cases of whom 44 were normal subjects, 38 hyperthyroid(including 29 before therapy and 9 euthyroids after antithyroid drug treatment,I or thyroidectomy), 9 hypothyroid, 20 pregnant women and 49 women on oral contraceptive. The data of this series indicated that taking the mean value of the normal group ±1.6 S.D.（i.e., 0.83-1.15） as its upper and lower limits, the percentage of agreement of the normal, hyper and hypothyroid groups, are 88.6%, 90.0%, and 77.8% with certain *** percentage of diagnostic agreement of theI uptake rate is compared with that of theI-Tplasma binding ratio both for the normal and hyperthyroid groups. The diagnostic value and limits of the test in assessment of thyroid function were discussed.

Leukocyte-technetium-99m uptake in Crohn's disease:Does it show subclinical disease?

AIM:To evaluate inflammatory activity in patients with Crohn's disease (CD) using technetium-99m-hexamethylpropyleneamine oxime (99mTc-HMPAO) granulocyte ***: Twenty patients (7 male and 13 female) with CD and five healthy volunteers were selected for 99mTc-HMPAO granulocyte scintigraphy. The Crohn's Disease Activity Index (CDAI), blood tests and C-reactive protein (CRP) of each patient were performed 7 d before the scintigraphic images. The leukocytes were labeled according to the International Society of Radiolabeled Blood Elements (ISORBE) consensus protocol and the scintigraphic images, including single photon emission computed tomography, were obtained 30 min and 2 h after injection of the radiolabeled ***:The labeling yield of the leukocytes with the lipophilic complex 99mTc-HMPAO was 55.0%±10%. Six of the 20 patients (30%) presented congruent results for the three parameters investigated (CDAI, Scintigraphic Index and CRP). On the other hand, 14 patients (70%) did not show congruent results. There was no significant correlation between the indices analyzed according to the Spearman test (P>0.05,n=20).CONCLUSION: The results suggest that 99mTc-HMPAO-labeled leukocyte scintigraphy could be important for determining inflammatory activity in CD even in the absence of clinical symptoms.

Effects of Qindan Capsule (芩丹胶囊) on Blood Pressure, Endothelin, Calcitonin Gene-related Peptide and Angiotensin-Ⅱ in Spontaneous Hypertensive Rats

Objective： To observe the hypotensive effects of Qindan Capsule （芩丹胶囊, QC） on spontaneous hypertensive rats （SHR） and its effect on the contents of endothelin （ET）, calcitonin gene-related peptide （CGRP） and angiotensin-Ⅱ （Ang-Ⅱ ） in plasma and vascular tissues, and to investigate the possible mechanism of QC in lowering blood pressure. Methods： Forty SHRs were divided into 5 groups： the high dosage QC group [QCHD, 750 mg/（kg.d） ], the low dosage QC group [QCLD, 150 mg/（kgd） ], the Niuhuang Jiangya Pill group [牛黄降压丸,, NJP, 200 mg/（kg.d）], the Captopril group [ 15 mg/（kgd）]and the model group, 8 in each group. Meanwhile, a normal control group consisting of 8 Wistar-Kyoto （WKY） rats was set up also. All the rats were administered with medicine level of ET, CGRP and Ang-Ⅱ in plasma and Ang-Ⅱ rats after 12 weeks of treatment. Results： The leve through gastrogavage. Systolic blood pressure （SBP）, in tissues of mesenteric artery were detected in all the of SBP after treatment in the QCHD group was lower than that in the model group （ P〈0.01 ）, but with no significant difference as compared with that in the Captopril group and the NJP group （P〉0.05）. After treatment, the plasma level of ET was lower and CGRP higher than those in the model group （both P〈0.05）, and also higher than those in the NJP and Captopril group （both P〈0.05）. As for the content of Ang- Ⅱ , in mesenteric arterial tissues, it was lower in the QCHD group than that in the model group （ P〈0.05）, but in plasma, it showed no significant difference between the two groups （P〉0.05）. Conclusion： QC has a satisfactory hypotensive action on SHR rats, and its mechanism may be associated with the regulation on plasma vasoactive peptide and regional renin-angiotensin system.

Potassium(K)is a major essential element in plant cells,and KUP/HAK/KT-type K+transporters participate in the absorption of K+into roots and in the long-distance transport to above-ground *** Arabidopsis thaliana,KUP9 is involved in the transport of K+and Cs+in *** this study,we investigated KUP9 function in relation to the K+status of the *** expression of KUP9 was upregulated in older leaves on K+-depleted medium,compared to the expression of the other 12 KUP genes in the KUP/HAK/KT family in *** grown on low K+medium,the kup9 mutant had reduced chlorophyll content in seedlings and chlorosis in older rosette ***-specific expression of KUP9 determined by KUP9 promoter:GUS assay depended on the K+status of the plants:In K+sufficient medium,KUP9 was expressed in the leaf blade towards the leaf tip,whereas in K+depleted medium expression was mainly found in the *** accordance with this,K+accumulated in the roots of kup9 *** short-term 43K+tracer measurement showed that 43K was transferred at a lower rate in roots and shoots of kup9,compared to the wild *** data show that KUP9 participates in the distribution of K+in leaves and K+absorption in roots under low K+conditions.
