Metastatic pheochromocytoma/paraganglioma(MPP)is a rare endocrine tumor that originates from extra-adrenal chromaffin cells such as the paraganglia cells of sympathetic and parasympathetic *** usually causes multiple solid tumors and exhibits strong aggressiveness with poor prognosis,with a reported 5-year survival rate of less than 50%.Cases of brain and retroperitoneal metastases at the initial diagnosis have not yet been *** report a 41-year-old male patient initially diagnosed with MPP in the brain and retroperitoneum who underwent multi-disciplinary collaborative surgery and simultaneous removal of two tumors at our *** pathology revealed infiltrative growth of a skull base *** patient chose to receive the tyrosine kinase inhibitor sunitinib as a targeted treatment.A 3-month follow-up after surgery showed that the patient recovered well without signs of metastasis or *** present multi-disciplinary surgery under similar circumstances for enhanced treatment and postoperative *** patient demonstrates a favorable prognosis during postoperative follow-up,indicating that simultaneous multidisciplinary surgery may offer greater benefits for MPP patients.

Maturity-onset diabetes of the young 10 caused by the c.4G>A (***2Thr)mutation is extremely rare, with only two reported studies to date. Herein, wereport another case that differs from previous cases in *** SUMMARYThe proband developed diabetes at the age of 27 years, despite having a normalbody mass index (BMI). She exhibited partial impairment of islet function, testedpositive for islet antibodies, and required high doses of insulin. Her sister alsocarried the c.4G>A (***2Thr) mutation, and their mother was stronglysuspected to carry the mutated gene. Her sister developed diabetes around 40years of age and required high doses of insulin, while the mother was diagnosedin her 20s and was managed with oral hypoglycemic agents;neither of them *** ***2Thr mutation carriers often experience relatively later onset and normalBMI. Treatment regimens vary between individuals.

To the Editor:Gastric cancer(GC)is the sixth most common type of cancer and the third most common cause of cancer-related deaths worldwide.[1]Due to the lack of typical clinical symptoms in the early stage of GC,most patients have advanced-stage disease at the time of initial treatment and have a poor *** rate of radical resection is *** after radical resection,recurrence or death occurs in 50%to 90%of patients,and the 5-year overall survival(OS)rate is less than 30%.[2]Therefore,various multimodal strategies,which would presumably reduce the risk of cancer recurrence and improve survival,are being explored to eliminate any residual tumors and *** chemotherapy(NAC)followed by surgery is recommended as the standard of care for patients with localized resectable GC.

Background:Ductal carcinoma in situ with microinvasion(DCIS-MI)is defined as ductal carcinoma in situ(DCIS)with a microscopic invasive focus≤1 mm in the longest *** current literature is controversial concerning the clinical prognostic features and management of *** narrative review described recently reported literature regarding the characteristics,treatment,and prognosis of ***:Searching PubMed for relevant articles covering the period of 1982 to 2021 using the following terms by MeSH and free-word:breast cancer,microinvasion,DCIS,DCIS-MI,and invasive ductal carcinoma(IDC).Results:DCIS-MI tends to express more aggressive pathological features such as necrosis,HER2+,ER-or PR-,and high nuclear *** overall prognosis of DCIS-MI is typically good,however,some indicators such as young age,HR-,HER2+and multimicroinvasive lesions,were associated with worse *** there are also conflicting results on the differences between the prognoses of DCIS-MI and DCIS or *** chemotherapy and anti-HER2 therapy still have uncertain benefits and are more likely to be used to treat high-risk patients who are HR-orHER2+to improve the ***:DCIS-MI has more aggressive pathological features,which may suggest its biological behavior is worse than that of DCIS and similar to early *** the overall prognosis of DCIS-MI is good,when making decisions about adjuvant therapy clinicians need to give priority to the hormone receptor status,HER2 expression and axillary lymph node status of patients,because these may affect the prognosis and treatment response.

Interleukin-2(IL-2)is a pleiotropic cytokine that orchestrates bidirectional immune responses via regulatory T cells(Tregs)and effector cells,leading to paradoxical ***,we report a strategy that exploited genetic code expansion-guided incorporation of the latent bioreactive artificial amino acid fluorosulfate-L-tyrosine(FSY)into IL-2 for proximity-enabled covalent binding to IL-2Rαto selectively promote Treg *** found that FSY-bearing IL-2 variants,such as L72-FSY,covalently bound to IL-2Rαvia sulfur-fluoride exchange when in proximity,resulting in persistent recycling of IL-2 and selectively promoting the expansion of Tregs but not effector *** assessment of L72-FSY-expanded Tregs demonstrated that L72-FSY maintained Tregs in a central memory phenotype without driving terminal differentiation,as demonstrated by simultaneously attenuated expression of lymphocyte activation gene-3(LAG-3)and enhanced expression of programmed cell death protein-1(PD-1).Subcutaneous administration of L72-FSY in murine models of pristane-induced lupus and graft-versus-host disease(GvHD)resulted in enhanced and sustained therapeutic efficacy compared with wild-type IL-2 *** efficacy of L72-FSY was further improved by N-terminal PEGylation,which increased its circulatory retention for preferential and sustained *** proximity-enabled covalent binding strategy may accelerate the development of pleiotropic cytokines as a new class of immunomodulatory therapies.

To the Editor: Frailty is defined as a vulnerable state that places older adults at increased risk for adverse health outcomes, and can explain substantial heterogeneity of health status. Previous studies have demonstrated the utility of the frailty phenotype scale and frailty index (FI) in identifying frail older people in China.[1] Several experimental and clinical studies on frailty have been published using the above methods in recent years, but influential factors for discriminating frailty levels in Chinese older adults have not yet been determined. Elucidating this issue is of importance, because early detection of frailty-related factors and subsequent intervention are essential for safeguarding functionality and preventing the occurrence of adverse consequences among older people.
