The BTB/TAZ domain-containing protein CmBT1-mediated CmANR1 ubiquitination negatively regulates root development in chrysanthemum

Roots are fundamental for plants to adapt to variable environmental conditions.The development of a robust root system is orchestrated by numerous genetic determinants and,among them,the MADS-box gene ANR1 has garnered substantial attention.Prior research has demonstrated that,in chrysanthemum,CmANR1positively regulates root system development.Nevertheless,the upstream regulators involved in the CmANR1-mediated regulation of root development remain unidentified.In this study,we successfully identified bric-a-brac,tramtrack and broad(BTB)and transcription adapter putative zinc finger(TAZ)domain protein CmBT1 as the interacting partner of CmANR1 through a yeasttwo-hybrid(Y2H)screening library.Furthermore,we validated this physical interaction through bimolecular fluorescence complementation and pull-down assays.Functional assays revealed that CmBT1 exerted a negative influence on root development in chrysanthemum.In both in vitro and in vivo assays,it was evident that CmBT1mediated the ubiquitination of CmANR1 through the ubiquitin/26S proteasome pathway.This ubiquitination subsequently led to the degradation of the CmANR1 protein and a reduction in the transcription of CmANR1-targeted gene CmPIN2,which was crucial for root development in chrysanthemum.Genetic analysis suggested that CmBT1 modulated root development,at least in part,by regulating the level of CmANR1 protein.Collectively,these findings shed new light on the regulatory role of CmBT1 in degrading CmANR1 through ubiquitination,thereby repressing the expression of its targeted gene and inhibiting root development in chrysanthemum.

Review of construction and demolition waste management tools and frameworks with the classification,causes,and impacts of the waste

This review looks over the current construction and demolition waste management(C&DWM)situations by scrutinizing the definition,classification,components,compositions,generated sources and causes,impacts of generated construction and demolition wastes(C&DWs),waste management hierarchy(WMH),3R principles(Reduce,Reuse,and Recycle),Circular Economy(CE),frameworks,tools,and approaches of C&DWM.After reviewing the literature this study contributes to the literature by the following means:(a)suitable working definitions of C&DW and C&DWM are provided,(b)an expanded WMH for construction and demolition operations is presented,(c)frameworks of C&DWM are identified and listed as follows:frameworks based on WMH,including 3R principles and CE concept,frameworks focusing on the quantification,estimation,and prediction of generated C&DW,frameworks focusing on effective and sustainable C&DWM,frameworks focusing economic,social,and environmental performance assessment,frameworks based on multi-criteria analysis(MCA),frameworks based on post-disaster recovery period,and other miscellaneous frameworks,and(d)four categories of tools utilized in C&DWM are identified and explained,namely,approaches employed in C&DWM,information technology(IT)tools employed in C&DWM,multi-criteria decision analysis(MCDA)tools employed in C&DWM,and C&DWM technologies.Moreover,this study also found that CE,and green rating system(GRS)are widely used approaches,Building Information Modeling(BIM),Radio Frequency Identification(RFID),Geographic Information System,and Big Data are the extensively used IT tools,Analytical Hierarchy Process,FUZZY,TOPSIS(Technique for Order Preference by Similarity to the Ideal Solution),Weighted Summation,Elimination and Choice Expressing the Reality II,Elimination and Choice Expressing the Reality III,Evaluation of Mixed Data,and REGIME(REG)are the widely used MCA tools in C&DWM,and Prefabricated Construction and Modular Construction are broadly used C&DWM technologies.Furthermore,it has been observed that the application of

An approach of the internal friction-dependent temperature changes for conventional and pure biogenic lubricating greases

This work investigated the temperature changes inside the bulk of lubricating greases under controlled high-shear stress conditions(250-500 s-1).For this purpose,a newly developed temperature-measuring cell called Calidus was successfully tested.The temperature changes(ΔT)have been related to the greases'components(thickener,base oil-type,and composition)and the structural degradation of the lubricating greases.Furthermore,a theoretical approach was proposed for calculating the internal temperature change of lubricating greases during shear stress.All greases showed an internal temperature profile characterised by a sudden rise inΔT within the first 4 h from starting the test and subsequentΔT decay until it reaches the steady state value.Furthermore,it was found that greases C1 and C5,formulated with lithium and calcium soap,respectively,with different soap content(16.1 wt%and 9.7 wt%,respectively),but the same base castor oil,showed the highest value of the maximumΔT,c.a.3.2 K,and the most drastic drop ofΔT.These greases showed both the highest specific densities and heat capacities.In addition,they showed the lowest ratio of expended energies(Rtee),which means more structural degradation in the stressed grease.On the contrary,the grease C3,with 13 wt%of Li-soap but the lowest base oil's viscosity,showed the lowest maximumΔT and the temperature profile was characterised by a moderate variation ofΔT along the test.The biogenic grease B3 developed a low-temperature change in the group of pure bio-genic greases close to grease C3.

Wear-driven tool failure is one of the main hurdles in the industry.This issue can be addressed through surface coating with ceramic-reinforced metal matrix composites.However,the maximum ceramic content is limited by cracking.In this work,the tribological behaviour of the functionally graded WC-ceramic-particlereinforced Stellite 6 coatings is studied.To that end,the wear resistance at room temperature and 400°C is investigated.Moreover,the tribological analysis is supported by crack sensitivity and hardness evaluation,which is of utmost importance in the processing of composite materials with ceramic-particle-reinforcement.Results indicate that functionally graded materials can be employed to increase the maximum admissible WC content,hence improving the tribological behaviour,most notably at high temperatures.Additionally,a shift from abrasive to oxidative wear is observed in high-temperature wear testing.

The efficiency and safety evaluation of hemoglobin hydrolysate as a non-heme iron fortifier

Hemoglobin hydrolysate is derived from the enzymatic degradation of hemoglobin.This work aimed to evaluate whether hemoglobin hydrolysate promotes the absorption of non-heme iron and the safety of absorbed iron in mice by analyzing the iron binding content,iron circulation,and liver homeostasis.We found that hemoglobin hydrolysate promoted the absorption of non-heme iron with high efficiency in duodenum by spontaneously binding non-heme iron during digestion,and increased hepatic iron content by up-regulating divalent metal transporter 1,zinc transporter 14,but hepatic iron content only increased at 3 weeks.Duodenal iron entered the blood through ferroportin without restriction at 3 weeks,and excessive iron entered the liver and then affected the hepatocyte membranes permeability and lipid synthesis through oxidative stress.With the prolongation of dietary intervention,the up-regulated hepcidin acted on the ferroportin to restrict excess iron from entering the blood,and then the hepatic homeostasis recovered.In addition,hemoglobin hydrolysate enhanced the hepatic antioxidant capacity.Taken together,hemoglobin hydrolysate has a strong ability to promote the absorption of non-heme iron in vivo,and the absorbed iron is relatively safe due to the regulation of hepcidin.

Application of the CatBoost Model for Stirred Reactor State Monitoring Based on Vibration Signals

Stirred reactors are key equipment in production,and unpredictable failures will result in significant economic losses and safety issues.Therefore,it is necessary to monitor its health state.To achieve this goal,in this study,five states of the stirred reactor were firstly preset:normal,shaft bending,blade eccentricity,bearing wear,and bolt looseness.Vibration signals along x,y and z axes were collected and analyzed in both the time domain and frequency domain.Secondly,93 statistical features were extracted and evaluated by ReliefF,Maximal Information Coefficient(MIC)and XGBoost.The above evaluation results were then fused by D-S evidence theory to extract the final 16 features that are most relevant to the state of the stirred reactor.Finally,the CatBoost algorithm was introduced to establish the stirred reactor health monitoring model.The validation results showed that the model achieves 100%accuracy in detecting the fault/normal state of the stirred reactor and 98%accuracy in diagnosing the type of fault.

Realizing high initial Coulombic efficiency in manganese-based layered oxide cathodes for sodium-ion batteries via P2/O'3 biphasic structure optimization

Abstract Mn-based layered oxides are among the most promising cathode materials for sodium-ion batteries owing to the advantages of abundance,environmenta friendliness,low cost and high specific capacity.P2 and O'3 are two representative structures of Mn-based layered oxides.However,the P2 structure containing insufficien Na generally exhibits low initial charge capacity,while O'3structure with sufficient Na delivers high initial charge capacity but poor cycle stability.This study prepared a multitude of NaxMnO2（x=0.7,0.8,0.9） cathode materials with varying P2/O'3 ratios and further investigated their electrochemical performances.The optimized Na0.8MnO2comprising 69.9 wt%O'3 and 30.1 wt% P2 phase,exhibited relatively balanced specific capacity,Coulombic efficiency and cycle stability.Specifically,it achieved a high specific capacity of 128.9 mAh·g-1with an initia Coulombic efficiency of 98.2%in half-cell configuration The Na0.8MnO2// hard carbon full cell also achieved a high specific capacity of 126.7 mAh·g-1with an initia Coulombic efficiency of 98.9%.Moreover,the capacity fading mechanism was revealed by combining in-situ and ex-situ X-ray diffraction.The findings of this study provide theoretical guidance for further modification design of Mnbased layered cathodes.

Paeoniflorin alleviates depression by inhibiting the activation of NLRP3 inflammasome via promoting mitochondrial autophagy

Depression ranks among the most common neuropsychiatric disorders globally.Current studies examining the roles of inflammation and mitochondrial autophagy in the antidepressant efficacy of paeoniflorin(PF)are sparse.This study aimed to elucidate PF’s antidepressant mechanism by promoting autophagy and inhibiting NLRP3 inflammasome activation using chronic unpredictable mild stimulation(CUMS)-induced C57BL/6 mouse models in vivo and corticosterone(CORT)-induced HT22 cell models in vitro.Results demonstrated that PF enhanced the viability of HT22 cells following CORT exposure,restored mitochondrial membrane poten-tial(MMP),reduced reactive oxygen species accumulation,increased LC3 fluorescence intensity,and suppressed inflammatory cy-tokine secretion and inflammation activation.Additionally,PF ameliorated depressive behaviors induced by CUMS and improved damage in hippocampal neurons.It also reduced the expression of NLRP3,ASC,Caspase-1,IL-1β,and the assembly of the NLRP3 in-flammasome.Moreover,PF upregulated the expression of autophagy-related proteins in the hippocampus,facilitating the clearance of damaged mitochondria and enhancing autophagy.The role of autophagy in PF’s antidepressant effects was further confirmed through the use of the autophagy inhibitor 3-methyladenine(3-MA),which reduced the efficacy of PF.In conclusion,PF effectively improved depressive behaviors in CUMS-induced mice and reduced NLRP3-mediated inflammation both in vivo and in vitro,likely via the in-duction of autophagy.

Recovery and in-situ reduction of precious metals by graphdiyne and graphdiyne oxide for antibacterial use

Based on the in-situ self-reduction and chemical stability,graphdiyne（GDY） and graphdiyne oxide（GDYO） are used as trapping agents to investigate the ability for recovering Au3+,Ag+,and Pd2+under different pH values and interfering ions.Under strong acidity at pH=1,these two agents demonstrate high select recovery towards the three precious metal ions,which could be insitu reduced to nanoparticles（NPs）.In addition,superparamagnetic Fe3O4NPs are deposited on the surfaces of GDY and GDYO.The magnetic responses enable GDYFe3O4to recover precious metals conveniently and efficiently by the aid of an external magnetic field.This study also confirms the antibacterial activity of the as-recovered NPs deposited on GDY and GDYO against Escherichia coli and Staphylococcus aureus(1 × 10～5 colony-forming unit（CFU）·ml-1),and the antibacterial rates are 100%.This strategy of recovering precious metals and subsequently reusing to combat pathogens will be of great significance for environmental remediation and biomedical applications.

Heterogeneous information phase space reconstruction and stability prediction of filling body–surrounding rock combination

Traditional research believes that the filling body can effectively control stress concentration while ignoring the problems of unknown stability and the complex and changeable stress distribution of the filling body–surrounding rock combination under high-stress conditions.Current monitoring data processing methods cannot fully consider the complexity of monitoring objects,the diversity of monitoring methods,and the dynamics of monitoring data.To solve this problem,this paper proposes a phase space reconstruction and stability prediction method to process heterogeneous information of backfill–surrounding rock combinations.The three-dimensional monitoring system of a large-area filling body–surrounding rock combination in Longshou Mine was constructed by using drilling stress,multipoint displacement meter,and inclinometer.Varied information,such as the stress and displacement of the filling body–surrounding rock combination,was continuously obtained.Combined with the average mutual information method and the false nearest neighbor point method,the phase space of the heterogeneous information of the filling body–surrounding rock combination was then constructed.In this paper,the distance between the phase point and its nearest point was used as the index evaluation distance to evaluate the stability of the filling body–surrounding rock combination.The evaluated distances(ED)revealed a high sensitivity to the stability of the filling body–surrounding rock combination.The new method was then applied to calculate the time series of historically ED for 12 measuring points located at Longshou Mine.The moments of mutation in these time series were at least 3 months ahead of the roadway return dates.In the ED prediction experiments,the autoregressive integrated moving average model showed a higher prediction accuracy than the deep learning models(long short-term memory and Transformer).Furthermore,the root-mean-square error distribution of the prediction results peaked at 0.26,thus outperforming the no-prediction method
