Evaluation of the therapeutic significance of Glehniae Radix polysaccharides on the ulcerative colitis through DSS-induced rat model

Glehniae Radix has a wide range of pharmaceutical applications， and research on its main components has mainly focused on coumarins， alkaloids， lignans， and flavonoids， while neglecting the research on polysaccharides. Literature reports and our previous studies have shown that polysaccharides have certain therapeutic significance in immune regulation， antioxidant， anti-inflammatory and other aspects. Herein， the rat model of ulcerative colitis (UC) was established to evaluate the anti-inflammatory efficacy of the prepared Glehniae Radix polysaccharide (GLP) from the perspectives of inflammatory factors， intestinal tissue morphology， and microflora changes. The polysaccharides are mainly composed of galacturonic acid， rhamnose， glucose， galactose， and arabinose in molar ratios of 1.4:9.2:33.3:2.5:2.9， and GLP could downregulate the expression pro-inflammatory factors (interleukin-6， tumor necrosis factor-α， and interferon-γ) and significantly upregulate the expression of anti-inflammatory factor (interleukin-10). In addition， Glehniae Radix aqueous extract (GLA)， GLP with low dosage (GLPL) and GLP with high dosage (GLPH) could increase the number of goblet cells， enhance the integrity of crypt structure， and reverse the status of inflammatory infiltrating cells. Moreover， GLA and GLPH could upregulate Lactobacillus and Lachnoclostridium in UC rats， and appropriately downregulate Lachnospiraceae＿NK4A136＿group， thereby optimizing the proportion of bacterial flora and improving the intestinal microbial environment. Our findings not only be valuable as theoretical materials for the further clinical applications of GLP， but the identified biomarkers and metabolic pathways also provide new clues for the diagnosis of UC.

Cysteine proteases continue to provide validated targets for treatment of human *** neurodegenerative disorders,multiple cysteine proteases provide targets for enzyme inhibitors,notably caspases,calpains,and *** reactive,active-site cysteine provides specificity for many inhibitor designs over other families of proteases,such as aspartate and serine;however,a)inhibitor strategies often use covalent enzyme modification,and b)obtaining selectivity within families of cysteine proteases and their isozymes is *** review provides a general update on strategies for cysteine protease inhibitor design and a focus on cathepsin B and calpain 1 as drug targets for neurodegenerative disorders;the latter focus providing an interesting query for the contemporary assumptions that irreversible,covalent protein modification and low selectivity are anathema to therapeutic safety and efficacy.

Biotransformation of 4-Hydroxybenzen Derivatives by Hairy Root Cultures of Polygonum multiflorum Thunb

The biotransformation of four 4-hydroxybenzen derivatives （1,4-benzenediol （compound 1）, 4-hydroxybenzaldehyde （compound 2）, 4-hydroxybenzyl alcohol （compound 3） and 4-hydroxybenzoic acid （compound 4）） by the hairy root cultures of Polygonum multiflorum Thunb. as a new biocatalyst was investigated. It was found that the substrates were transformed to their corresponding glucosides, 4-hydroxyphenyl β-D-glucopyranoside （arbutin, compound la）, 4-hydroxymethylphenyl β-D-glucopyranoside （gastrodin, compounds 2a, 3a） and 4-carboxyphenyl α-D- glucopyranoside （compound 4a）, respectively. In the meantime, the hairy roots of P. multiflorum were able to stereoselectively and regioselectively glucosylate phenolic hydroxyl groups of compounds 1-4, but the cultures could not glucosylate the aldehyde group of compound 2 or the benzyllc hydroxyl group of compound 3, and no glucosyl esterification of carboxyl groups of compound 4 was detected. On the other hand, the result also showed that the hairy roots of P. multiflorum were able to reduce the 4-hydroxybenzaldehyde to its corresponding alcohol. This is the first report that substrate 4 has been converted into its α-D-glucopyranoside by a plant biotransformα- tion system.

Venous thromboembolism(VTE)is a typical cardiovascular disease,which is phenotypically characterized by painful leg swelling,chest pain,shortness of breath,and even death[1].VTE renders a high morbidity and mortality as it is the main cause of death of hospitalized *** treatment of VTE mostly adopts immediate anticoagulation agents via a risk assessment of ***,traditional Chinese medicine(TCM)has unique medicinal priority on reducing swelling,stopping bleeding and promoting blood circulation,which therefore can eliminate the obstruction of patient’s channels so as to exert analgesic and anti-inflammatory bioactivities[2].However,well-controlled and-designed clinical trials of the selected TCM formulae are widely performed to support the clinical application of traditional Chinese medicine for the prevention of VTE in a real world.

Multitarget therapeutic strategies for Alzheimer's disease

Neurodegenerative diseases such as Alzheimer's, Huntington's and Parkinson's diseases have multifaceted nature because of the different factors contributing to their progression. The complex nature of neurodegenerative diseases has developed a pressing need to design multitarget-directed ligands to address the complementary pathways involved in these diseases. The major enzyme targets for development of therapeutics for Alzheimer's disease are cholinesterase and β-secretase enzymes. In this review, we discuss recent advances in profiling single target inhibitors based on these enzymes to multitarget-directed ligands as potential therapeutics for this devastating disease. In addition, therapeutics based on iron chelation strategy are discussed as well.

Phytochemical study of Illicium angustisepalum and its biological activities

Sixteen compounds, including two new natural products(1 and 2), were obtained from the twigs of Illicium angustisepalum. The structures were elucidated based on NMR, MS, IR data and optical rotation values. Compounds 4,5,6 and 8 displayed moderate antibacterial activities against clinical isolates; compounds 4,5,8,9 and 15 protected neural cells against oxidative stress; and compounds 10 and 14 exhibited anti-acetylcholinesterase activity.
