Background:Over the past few decades,a threefold increase in obesity and type 2 diabetes(T2D)has placed a heavy burden on the health-care system and *** studies have shown correlations between obesity,T2D,and neurodegenera-tive diseases,including *** is imperative to further understand the relation-ship between obesity,T2D,and cognitive ***:This investigation tested and evaluated the cognitive impact of obesity and T2D induced by high-fat diet(HFD)and the effect of the host genetic background on the severity of cognitive decline caused by obesity and T2D in collaborative cross(CC)*** CC mice are a genetically diverse panel derived from eight inbred ***:Our findings demonstrated significant variations in the recorded phenotypes across different CC lines compared to the reference mouse line,C57BL/***037 line exhibited a substantial increase in body weight on HFD,whereas line CC005 ex-hibited differing responses based on *** tolerance tests revealed significant variations,with some lines like CC005 showing a marked increase in area under the curve(AUC)values on *** weights,including brain,spleen,liver,and kidney,varied significantly among the lines and sexes in response to *** tests using the Morris water maze indicated that cognitive performance was differentially affected by diet and genetic ***:Our study establishes a foundation for future quantitative trait loci map-ping using CC lines and identifying genes underlying the comorbidity of Alzheimer's disease(AD),caused by obesity and *** genetic components may offer new tools for early prediction and prevention.

Ulcerative colitis [UC] is established by a chronic， diffuse inflammation of colonic mucosa with poorly defined etiology， and without sufficient treatment or cure available for remission. In this context， there is a growing recognition and consensus that synbiotic supplement may be promising and impactful strategies towards ameliorating UC and related inflammatory disorders， garnering significant attention from researchers as an alternative therapy. Herein， we innovatively prepared a novel synbiotic combination including Lactobacillus acidophilus， Bifidobacterium infants and prebiotics konjac glucomannan oligosaccharides [KGMO] to explore the potential therapeutic effects in dextran sodium sulfate [DSS] induced UC mouse model. The results demonstrated that the synbiotic effectively improved colitis symptoms in mice by mitigating weight loss， disease activity index [DAI]， colonic pathological damage， and colonic oxidative stress. it also observed that the synbiotic preserve intestinal barrier function integrity， reduces metabolic endotoxemia as well as inhibits the TLR4/NF-κB， NLRP3/Caspase-1， and Nrf2/Keap1 signaling pathway. Furthermore， the synbiotic modulated microbial homeostasis directly by enhancing beneficial microbes and reducing potentially harmful bacteria. In addition， microbiome phenotype prediction and bacterial functional potential prediction analysis demonstrated that the synbiotic supplementation regulated gut microbiota function involving inflammatory injury， metabolism， immune response， and pathopoiesia. Especially， the synbiotic not only restored the balance of Th1/Th2 cells as well as Th17/Treg cells along with specific inflammatory factors expression， but also maintained bile acid homeostasis by regulating FXR/FGF15 signaling pathway. Especially important is that the synbiotic was as effective as mesalazine against UC. Transforming growth factor-beta 1(TGF-β1)has been extensively studied for its pleiotropic effects on central nervous system *** neuroprotective or neurotoxic effects of TGF-β1 in specific brain areas may depend on the pathological process and cell types ***-gated sodium channels(VGSCs)are essential ion channels for the generation of action potentials in neurons,and are involved in various neuroexcitation-related ***,the effects of TGF-β1 on the functional properties of VGSCs and firing properties in cortical neurons remain *** this study,we investigated the effects of TGF-β1 on VGSC function and firing properties in primary cortical neurons from *** found that TGF-β1 increased VGSC current density in a dose-and time-dependent manner,which was attributable to the upregulation of Nav1.3 *** VGSC current density and Nav1.3 expression were significantly abolished by preincubation with inhibitors of mitogen-activated protein kinase kinase(PD98059),p38 mitogen-activated protein kinase(SB203580),and Jun NH2-terminal kinase 1/2 inhibitor(SP600125).Interestingly,TGF-β1 significantly increased the firing threshold of action potentials but did not change their firing rate in cortical *** findings suggest that TGF-β1 can increase Nav1.3 expression through activation of the ERK1/2-JNK-MAPK pathway,which leads to a decrease in the firing threshold of action potentials in cortical neurons under pathological ***,this contributes to the occurrence and progression of neuroexcitatory-related diseases of the central nervous system.

AIM:To isolate and identify the conjunctival microbiota of cataract patients and analyze the associated influencing ***:This study recruited 216 participants(216 eyes)from April 2022 to July *** the condition of no antibiotic use prior to cataract surgery,sterile swabs were used to collect samples from the lower conjunctival *** cultures were then conducted,followed by species identification through 16S rDNA gene *** factors associated with positive or negative bacterial isolation rates were analyzed,including age,gender,meibomian gland dysfunction(MGD),history of hypertension,history of diabetes,history of cancer,history of infectious diseases and the habit of wearing ***:Among the 216 eyes,78 eyes yielded isolates,with an isolation rate of 36.11%,detecting a total of 122 ***-positive rods accounted for 49.18%(60 strains),gram-positive cocci accounted for 45.08%(55 strains),gram-negative bacteria accounted for 4.92%(6 strains),and fungi accounted for 0.82%(1 strain).This study found that the most abundant genera in the conjunctival sac were Corynebacterium(42.62%),Staphylococcus(31.15%),Micrococcus(9.84%),Acinetobacter(4.10%),and Bacillus(3.28%).Furthermore,age(P=0.006),gender(P=0.039),diabetes(P=0.003),history of infectious diseases(P=0.02),and duration of mask replacement(P<0.001)were important factors influencing the positive bacterial culture of the conjunctival *** hypertensive patients exhibited a higher isolation rate of conjunctival bacteria,it did not reach statistical significance,and the history of cancer did not affect the isolation rate of the conjunctival microbial community in cataract patients before ***:Potential changes are observed in the conjunctival microbiota among a sample of middleaged and elderly individuals from Beijing,***,an increased isolation rate of Corynebacterium and Micrococcus is detected,suggesting a possible change in the mic

The exopolysaccharide matrix of diazotrophic cyanobacteria was used to integrate phosphorus(P)and potassium(K)solubilizing bacteria,enhancing the survival of plant growth-promoting rhizobacteria,and ultimately the survival of bacteria in the rhizosphere for better plant growth.A new biofilm-based formulation comprising the diazotrophic cyanobacteria Anabaena AMP2,P-solubilizing Bacillus megaterium *** PB1,and K-solubilizing Rhizobium pusense KRBKKM1 was tested for efficacy in *** growth medium with half-strength BG-11 medium supplemented with 3%glucose showed best for biofilm formation under in vitro *** of the methanolic extract of the cyanobacterial-bacterial biofilm(CBB)showed the activity of antioxidants,such as 2-methoxy phenol and pentadecane,which are proven to improve plant-microbe interactions and plant growth,*** of rice seeds with CBB extract at 100 mL/kg or 200 mL/kg showed significant enhancement in germination rate and seedling ***,a pot culture experiment with the CBB formulations was carried out,and different growth and yield parameters were *** component analysis showed that plant growth,yield,soil dehydrogenase activity,and soil chlorophyll content were positively correlated with rice plants amended with vermiculite-based CBB at 2 kg/hm^(2) followed by a spray with aqueous CBB formulation at 5 mL/L at 15 and 30 d after rice transplanting grown with a 25%reduced level of nitrogen/phosphorus/potassium chemical fertilizers than the recommended ***,Pearson correlation analysis showed that yield was positively correlated with soil dehydrogenase(r=0.92**)and soil chlorophyll content(r=0.96**).We concluded that CBB could be used as a novel biofilm-based bio-inoculant to increase rice productivity and crop fitness as a component in integrated nutrient management and sustainable organic farming strategies with reduced chemical fertilizers.
