Diagnosis of coronary artery disease using targeted post-mortem computed tomography coronary angiography:a case report

Stromal vascular fraction combined with silicone rubber chamber improves sciatic nerve regeneration in diabetes

Purpose： To study the effects of transplantation of characterized uncultured stromal vascular fraction （SVF） on sciatic nerve regeneration. Methods： A 10-mm sciatic nerve defect was bridged using a silicone conduit filled with SVF. In control group, silicone conduit was filled with phosphate-buffered saline alone. In sham-operated group, the sciatic nerve was only exposed and manipulated. The regenerated nerve fibers were studied 8 and 12 weeks after surgery. Results： Behavioral and functional studies confirmed faster recovery of regenerated axons in SVF transplanted animals than in control group （p 〈 0.05）. Gastrocnemius muscle mass in SVF transplanted animal was found to be significantly more than that in control group. Morphometric indices of the re- generated fibers showed the number and diameter of the myelinated fibers to be significantly higher in SVF transplanted animals than in control group. In immunohistochemistry, the location of reactions to S- 100 in SVF transplanted animals was clearly more positive than that in control group. Conclusion： SVF transplantation combined with silicone conduit could be considered as a readily accessible source of stromal cells that improves functional recovery of sciatic nerve. It may have clinical implications for the surgical management of acute diabetic patients after facial nerve transection.

Toxico-Pathological Studies of <i>Foeniculum vulgare</i>Plant in Mice

Background and objective: Foeniculum vulgare-Mill (Umlliferae), is widely used in the Arabian Peninsula for treating various human and animal diseases. There is need to insure the safety of this plant as a remedy that could be used for a long time. This study aimed to determine the LD50 of the aerial parts of Foeniculum vulgare in mice, as well as to investigate its toxic pathological effects. Methodology: A total of eighty (80) albino mice of both sexes, weighing 25 - 30 g, were used in the present study. Ten (10) mice were used for the determination of LD50 of Foeniculum vulgare in mice. Fifty (50) mice were used in the acute toxicity, and twenty (20) mice were used to study the subacute toxicity of the aerial parts of Foeniculum vulgare juice. Results: the present results revealed that the LD50 of Foeniculum vulgare juice in mice was 9.772 mg/kg/body weight (BW). On the other hand, mice received 0.9 mg/kg/BW of Foeniculum vulgare juice intraperitoneally showed slightly closed eyes intermittent convulsions itching of the nose and increased respiratory rate and abdominal movements. Histopathological examination of the liver showed hepatocytic degeneration and necrosis, congestion with perivascular mononuclear cell infiltrations and peribiliary mononuclear cell aggregations. Whereas the spleen of mice showed depletion of lymphocytes and mononuclear as well as multinuclear cells. The duodenum showed sub-epithelial congested blood vessels, numerous areas of lymphocytic infiltrations and vascular and hydropic degeneration of epithelial cells lining the duodenal Brunner’s glands. Moreover, the kidney showed degenerated tubular epithelial cells, congestion with excess of mononuclear cells, necrotic and desquamated epithelial cells of the proximal convoluted tubules. The lung of mice showed congestion, emphysema with variable degrees of mononuclear cell infiltrations and peribronchial congested blood capillaries and mononuclear cell infiltration. Methanolic extract of Clausena excavata promotes wound healing via antiinflammatory and anti-apoptotic activities

Objective: To investigate the anti-inflammatory properties of methanolic extract of Clausena excavata in lipopolysaccharide(LPS)-activated macrophages(J774 A.1) and the effect on skin wound in a rat model through determining cytokine levels and gene expressions. Methods: The effects of methanolic extract of Clausena excavata on in vitro viability and TNF-α, IL-6, IL-10, and nitric oxide release by LPS-activated J774 A.1 cells were determined. In addition, relative expressions of BAX, BCL-2 and COX-2 genes were examined in healed wounds of rats. Results: The methanolic extract of Clausena excavata was not toxic to J774 A.1 cells at the highest dose of 400 μg/m L. It decreased levels of TNF-α and IL-6, while increasing IL-10 level in LPSactivated J774 A.1 cells and in the healed wounds of rats. The methanolic extract of Clausena excavata also inhibited nitric oxide production in LPS-activated J774 A.1 cells. The BAX and COX-2 genes were downregulated while the BCL-2 gene was upregulated in the healed wound of rats. Conclusions: The methanolic extract of Clausena excavata promotes wound healing via its anti-inflammatory and anti-apoptotic activities.

Role of Neuropeptide Y and Peroxisome Proliferator-activated Receptor γ Coactivator-1α in Stress Cardiomyopathy

Death following situations of intense emotional stress has been linked to the cardiac pathology described as stress cardiomyopathy, whose pathomechanism is still not clear. In this study, we sought to determine, via an animal model, whether the transcriptional coactivator peroxisome proliferator-activated receptor γ coactivator-1alpha (PGC-1α) and the amino peptide neuropeptide Y (NPY) play a role in the pathogenesis of this cardiac entity. Male Sprague-Dawley rats in the experimental group were subjected to immobilization in a plexy glass box for 1 h, which was followed by low voltage elec-tric foot shock for about 1h at 10s intervals in a cage fitted with metallic rods. After 25 days the rats were sacrificed and sections of their hearts were processed. Hematoxylin-eosin staining of cardiac tissues revealed the characteristic cardiac lesions of stress cardiomyopathy such as contraction band necrosis, inflammatory cell infiltration and fibrosis. The semi-quantitative RT-PCR analysis for PGC-1α mRNA expression showed significant overexpression of PGC1-α in the stress-subjected rats (P<0.05). Fluorescence immunohistochemistry revealed a higher production of NPY in the stress-subjected rats as compared to the control rats (P=0.0027). Thus, we are led to conclude that following periods of intense stress, an increased expression of PGC1-α in the heart and an overflow of NPY may lead to stress car-diomyopathy and even death in susceptible victims. Moreover, these markers can be used to identify stress cardiomyopathy as the cause of sudden death in specific cases.

Effect of local administration of cyclosporine A on pe- ripheral nerve regeneration in a rat sciatic nerve transec- tion model

Objective： To assess the effect of cyclosporine A （CsA） loaded in chitosan conduit on bridg- ing the sciatic nerve defects in a rat model. Methods： A 10 mm sciatic nerve defect was bridged using a chitosan conduit filled with 10 μl carrier-drug dilu- tion （10 pg/L CsA）. In control group, the conduit was filled with the same volume of carrier dilution alone. The regene- rated fibers were studied 4, 8 and 12 weeks after surgery. Results： The functional study confirmed faster recov- ery of the regenerated axons in treatment group than control group （P〈0.05）. There was statistically significant differ- ence of the gastrocnemius muscle weight ratios between treatment and control groups （P〈0.05）. Morphometric indi-ces of regenerated fibers showed that the number and diam- eter of the myelinated fibers in CsA-treated animals were significantly higher than those in control group. In immunohistochemistry, the location of reactions to S-100 in CsA group was clearly more positive than control group. Conclusion： CsA loaded in a chitosan conduit results in improvement of functional recovery and quantitative mor- phometric indices of sciatic nerve. It is easily available with- out any complications compared with its systemic administration.

The forensic pathological analysis of sport-related sudden cardiac death in Southern China

Antioxidant and Anti-Inflammatory Effects of Nano-Selenium against Cypermethrin-Induced Liver Toxicity

Cypermethrin (Cym) is a synthetic class II pyrethroid that is widely used and has a big risk to health. Cypermethrin produces oxidative stress and enhances inflammatory damage of liver. The present study was designed to investigate the ameliorating effects of NSe against Cym-induced hepatotoxicity in rats. For this purpose, twenty four male rats were divided into three groups. Group (I) was gavaged with Cym (control group), group (II) gavaged daily with Cym (1 mg/kg body weight), and group (III) gavaged with Cym + NSe (2.5 mg kg body weight/day, three times a week) for 21 days. Cypermethrin increased serum liver enzymes, oxidative stress and inflammatory markers. Administration of NSe significantly reduced the increased serum liver enzymes and inflammatory parameters and restored the antioxidant capacity in liver. Our results suggest that Nse exhibits promising hepato-protective effects against Cym-induced oxidative damage and inflammation.
