Nerve regeneration following traumatic peripheral nerve injuries and neuropathies is a complex process modulated by diverse factors and intricate molecular *** studies have focused on factors that stimulate axonal outgrowth and myelin ***,recent studies have highlighted the pivotal role of autophagy in peripheral nerve regeneration,particularly in the context of traumatic ***,autophagy-targeting modulation has emerged as a promising therapeutic approach to enhancing peripheral nerve *** current understanding suggests that activating autophagy facilitates the rapid clearance of damaged axons and myelin sheaths,thereby enhancing neuronal survival and mitigating injury-induced oxidative stress and *** actions collectively contribute to creating a favorable microenvironment for structural and functional nerve regeneration.A range of autophagyinducing drugs and interventions have demonstrated beneficial effects in alleviating peripheral neuropathy and promoting nerve regeneration in preclinical models of traumatic peripheral nerve *** review delves into the regulation of autophagy in cell types involved in peripheral nerve regeneration,summarizing the potential drugs and interventions that can be harnessed to promote this *** hope that our review will offer novel insights and perspectives on the exploitation of autophagy pathways in the treatment of peripheral nerve injuries and neuropathies.

Induced neural stem cells regulate microglial activation through Akt-mediated upregulation of CXCR4 and Crry in a mouse model of closed head injury

Microglial activation that occurs rapidly after closed head injury may play important and complex roles in neuroinflammation-associated neuronal damage and *** previously reported that induced neural stem cells can modulate the behavior of activated microglia via CXCL12/CXCR4 signaling,influencing their activation such that they can promote neurological ***,the mechanism of CXCR4 upregulation in induced neural stem cells remains *** this study,we found that nuclear factor-κB activation induced by closed head injury mouse serum in microglia promoted CXCL12 and tumor necrosis factor-αexpression but suppressed insulin-like growth factor-1 ***,recombinant complement receptor 2-conjugated Crry(CR2-Crry)reduced the effects of closed head injury mouse serum-induced nuclear factor-κB activation in microglia and the levels of activated microglia,CXCL12,and tumor necrosis factor-α.Additionally,we observed that,in response to stimulation(including stimulation by CXCL12 secreted by activated microglia),CXCR4 and Crry levels can be upregulated in induced neural stem cells via the interplay among CXCL12/CXCR4,Crry,and Akt signaling to modulate microglial *** agreement with these in vitro experimental results,we found that Akt activation enhanced the immunoregulatory effects of induced neural stem cell grafts on microglial activation,leading to the promotion of neurological recovery via insulin-like growth factor-1 secretion and the neuroprotective effects of induced neural stem cell grafts through CXCR4 and Crry upregulation in the injured cortices of closed head injury ***,these beneficial effects of Akt activation in induced neural stem cells were positively correlated with the therapeutic effects of induced neural stem cells on neuronal injury,cerebral edema,and neurological disorders post–closed head *** conclusion,our findings reveal that Akt activation may enhance the immunoregulatory effects of induced neura

Vaccinations are essential for preventing and treating disease,especially cancer nanovaccines,which have gained considerable interest recently for their strong anti-tumor immune *** can prompt the immune system to generate antibodies and activate various immune cells,leading to a response against tumor tissues and reducing the negative effects and recurrence risks of traditional chemotherapy and *** enhance the flexibility and targeting of vaccines,nanovaccines utilize nanotechnology to encapsulate or carry antigens at the nanoscale level,enabling more controlled and precise drug delivery to enhance immune *** nanovaccines function by encapsulating tumor-specific antigens or tumor-associated antigens within *** small size of these nanomaterials allows for precise targeting of T cells,dendritic cells,or cancer cells,thereby eliciting a more potent anti-tumor *** this paper,we focus on the classification of carriers for cancer nanovaccines,the roles of different target cells,and clinically tested cancer nanovaccines,discussing strategies for effectively inducing cytotoxic T lymphocytes responses and optimizing antigen presentation,while also looking ahead to the translational challenges of moving from animal experiments to clinical trials.

The incidence and predictors of high-degree atrioventricular block in patients with bicuspid aortic valve receiving selfexpandable transcatheter aortic valve implantation

BACKGROUND The high-degree atrioventricular block(HAVB)in patients with bicuspid aortic valve(BAV)treated with transcatheter aortic valve implantation(TAVI)remains *** study aims to explore this poorly understood subject of mech-anisms and predictors for HAVB in BAV self-expandable TAVI *** We retrospectively included 181 BAV patients for *** computed tomography data,the curvature of ascending aorta(AAo)was quantified by the angle(AAo angle)between annulus and the cross-section at 35 mm above annulus(where the stent interacts with AAo the most).The valvular anatomy and leaflet calcification were also *** The 30-day HAVB rate was 16.0%(median time to HAVB was three days).Type-1 morphology was found in 79 pat-ients(43.6%)(left-and right-coronary cusps fusion comprised 79.7%).Besides implantation below membrane septum,large AAo angle[odds ratio(OR)=1.08,P=0.016]and type-1 morphology(OR=4.97,P=0.001)were found as the independent predictors for *** with baseline right bundle branch block,these predictors showed strong predictability for HAVB with area under the cure of 0.84(sensitivity=62.1%,specificity=92.8%).Bent AAo and calcified raphe had a synergistic effect in facilita-ting high implantation,though the former is associated with at-risk deployment(device implanted above annulus+prothesis pop-out,versus straight AAo:9.9%vs.2.2%,P=0.031).CONCLUSIONS AAo curvature and type-1 morphology are novel predictors for HAVB in BAV patients following self-expandable *** patients with bent AAo or calcified raphe,a progressive approach to implant the device above the lower edge of membrane septum is favored,though should be done cautiously to avoid pop-out.

Relationship of body fat and left ventricular hypertrophy with the risk of all-cause death in patients with coronary artery disease

BACKGROUND Left ventricular hypertrophy(LVH)is prevalent in obese ***,both of LVH and obesity is as-sociated with subclinical LV *** study aims to investigate the interplay between body fat and LVH in relation to all-cause death in patients with coronary artery disease(CAD).METHODS In this retrospective cohort study,a total of 2243 patients with angiographically proven CAD were *** fat and LV mass were calculated using established *** were grouped according to body fat percentage and pres-ence or absence of ***-proportional hazard models were used to observe the interaction effect of body fat and LVH on all-cause *** Of 2243 patients enrolled,560(25%)had a higher body fat percentage,and 1045(46.6%)had *** a median follow-up of 2.2 years,the cumulative mortality rate was 8.2%in the group with higher body fat and LVH,2.5%in those with lower body fat and no LVH,5.4%in those with higher body fat and no LVH,and 7.8%in those with lower body fat and LVH(log-rank P<0.001).There was a statistically significant interaction between body fat percentage and LVH(P interaction was 0.003).After correcting for confounding factors,patients with higher body fat and LVH had the highest risk of all-cause death(HR=3.49,95%CI:1.40-8.69,P=0.007)compared with those with lower body fat and no LVH;in contrast,patients with higher body fat and no LVH had no statistically significant difference in risk of death compared with those with lower body fat and no LVH(HR=2.03,95%CI:0.70-5.92,P=0.195).CONCLUSION A higher body fat percentage was associated with a different risk of all-cause death in patients with CAD,strat-ified by coexistence of LVH or *** body fat was significantly associated with a greater risk of mortality among patients with LVH but not among those without LVH.

Acupuncture Treatment for Post-Stroke Dysphagia: An Update Meta-Analysis of Randomized Controlled Trials

Objective： To explore the effectiveness and safety of acupuncture in patients with post-stroke dysphagia by an update meta-analysis. Methods： Potentially eligible RCTs aimed to evaluate the effects of acupuncture vs. non-acupuncture treatments, such as rehabilitation training or routine medication on the swallowing difficulty after stroke were searched from PubMed, Cochrane Library, china National Knowledge Infrastructure, and other database from the earliest record to June 2016. Patient demographics, regimens for acupuncture, type of controls, methods of randomization, and measurements of the clinical symptoms of dysphagia were retrieved. The relative risk （RR） and 95% confidence interval （CI） of effective rate of dysphagia was calculated after intervention performed following admission. Subgroup analyses and a meta- regression analysis were performed to describe the heterogeneity. Results： Twenty-nine RCTs comprising 2,190 patients were included. The included studies had a medium quality grade based on the Consolidated Standards of Reporting Trials （CONSORT） and Standards for Reporting Interventions in Clinical Trials of Acupuncture （STRICTA） checklist. Acupuncture therapy provided a higher effective rate compared with non- acupuncture treatments [RR=1.33, 95% confidence interval （CI）, 1.25 to 1.43]. Subgroup and meta-regression analyses suggested that acupuncture intensity and measurement method were main sources of heterogeneity and resulted in a significant difference for pooled effect size. No severe adverse events were documented in these RCTs. Conclusions： Our meta-analysis provides a new evidence supporting the efficacy and safety of acupuncture in treatment to post-stroke dysphagia in short-term compared with rehabilitation or medication. More high-quality and large-scale research studies are needed.

Effect of statins type on incident prostate cancer risk： a meta-analysis and systematic review

The aim of this study is to investigate the effect of statins type or even when grouping statins by hydrophilic or hydrophobic nature on prostate cancer risk. A literature search was performed without language restrictions using the databases of PubMed （1984.1-2015.3）, MEDLINE （1984.1-2015.3）, and EMBASE （1990.1-2015.3）. Two independent reviewers appraised eligible studies and extracted data. Weighted averages were reported as relative risk （RR） with 95% confidence intervals （CI）. Statistic heterogeneity scores were assessed with the standard Cochran＇s Q-test and F statistic, Publication bias was detected using the Begg＇s and Egger＇s tests. All statistical analyses were conducted by STATA version 10. Finally, fourteen studies were included in the meta-analysis. Both hydrophilic and hydrophobic statins showed no association with incidence of prostate cancer （RR = 1.00, 95% Ch 0.82-1.17; RR = 0.90, 95% CI： 0.73-1.08, respectively）. Meanwhile, the risk of prostate cancer was not reduced in simvastatin （RR = 0.89, 95% CI- 0.72-1.05）, pravastatin （RR = 1.02, 95% Ch 0.94-1.11）, atorvastatin （RR = 0.89, 95% CI： 0.76-1.02）, fluvastatin （RR = 0.99, 95% Ch 0.97-1.01）, or Iovastatin users （RR = 0.94, 95% CI： 0.79-1.08）. The funnel plot showed that there was no publication bias. The results showed that statins had a neutral effect on prostate cancer risk; hydrophilic and hydrophobic statins as well as any subtype of statins did not affect the risk of prostate cancer.
