Correlates of resistance to gastrointestinal nematode infection in Nigerian West African dwarf sheep

Objective:To investigate correlates of resistance to G1 nematode infection in Nigerian West African dwarf(WAD) ***:Thirty three sheep were randomly assigned to two groups, A(n=27) which were used for experimental infections,and B(n=6) which served as uninfected *** infected animal received weekly escalating infections with infective larvae(60% Haemonchus contortus(***) and 40%Trichostrongylus colubriformis(***) for 4 *** responses of all the infected and control sheep were assessed by faecal egg count(FEC),worm burden(Wb),packed cell volume(PCV),body weight(Bwt).and body condition score(BCS).On the basis of their individual faecal egg output,Lambs in group A with epg≤1 000 on any sampling day were classified as low faecal egg count(LFEC) phenotype(n = 16),those with epg between 1 000 and 10 000 as intermediate(n=5) and lambs with epg > 10 000 as high feacal egg count(HFEC) phenotype(n=6).Results:The difference between the FEC classes was highly significant(P=0.001).The BCS and weight gained at the end of the experiment by the control and LFEC sheep was significandy higher(P≤0.05) than those of the intermediate and HFEC *** was a significant and negative correlation between the parasitological measures and the trio of BCS,PCV and Bwt of ***:The result of the study indicated that the FEC,weight gain,PCV,and BCS are correlates and potential selection crileria of G1 nematode resistant WAD sheep.

Isolation and identification of bioactive antibacterial components in leaf extracts of Vangueria spinosa(Rubiaceae)

Objective:The column chromatographic fraction of ethyl acetate(EA1,EA2,EA3,EA4 and EA5) leaf extracts of Vangueria spinosa(***) were screened for antibacterial activity and phytochemical ***:EA3 fraction was isolated and identified by column chromatography,thin layer chromatography,spectral data analysis and phytochemical screening were used for ***:EA3 fraction was significantly active at 4 to 64 mg/L against Staphylococcus aureus,Escherichia coli,Klebsiella pneumoniae and Pseudomonas aeruginosa with minimum inhibitory concentration of 1.5625 to 3.1250 mg/*** active fraction(EA3) revealed the presence of flavonoid with retention factor value(R_f) of *** active antibacterial agent in the most potent fraction(EA3) was isolated and identified as flavonoid(-)-epicatechin-3-O-β-glucopyranoside by thin layer chromatography(TLC) and phytochemical ***1 and EA2 show inhibitory activity at 4 to 64 mg/L against Staphylococcus aureus only where as fraction EA4 and EA5 do not shows any inhibitory activity within that range of concentration against any ***:The results support the ethnomedicinal use of leaf of *** for the treatment of bacterial diseases.

Effect of artmether,hemin and Fe^(3+) on recombinant lactate dehydrogenase from Schistosoma japonicum

Objective:To explore antischitosome effects of artemether,hemin and Fe on S/***: Enzyme activity of rS/LDH was assayed in the standard reaction system by adding different concentration of reagents(0.00-0.10 mM artemether,0.00-0.02 mM hemin,0.00-0.50 mM Fe^(3+)). Same solvents of the each reagent were used as ***:There was no enzyme activity inhibition observed at 0.10 mM artemther:obivious inhibition for lactate oxidation reaction and pyruvate reduction reaction were detected at 0.002 mM and 0.004 mM of hemin,respectively: comparing with that of the control(P<0.05).The relative enzymatic activity inhibitions for pyruvate reduction reaction and lactate oxidation reaction at 0.02 mM hemin were 93.48%and 100.00%,respectively,comparing with that of the control(P<0.01):both pyruvate reduction and lactate oxidation reaction were inhibited completely at 0.50 mM Fe^(3+),comparing with that of the control(P<0.01).Conclusions:The results implied that SjLDH was not the direct molecular target of *** and Fe are inhibitors of SjLDH.

Acanthus leucostachyus leaf extracts promote excision wound healing in mice

Objective:To evaluate the in vivo wound healing activity of Acanthus leucostachyus leaf extracts using an excision wound model in ***:Mice were divided into two groups of six animals in each group:the control group and the Acanthus leucostachyus extract-treated *** potential was evaluated by determination of physical parameters(contraction rate,epithelialization period,and tensile strength),biochemical parameters(protein,DNA,and hydroxyproline content),the expression of growth factor and pro-inflammatory cytokines,as well as histological and ultrastructural ***:Treatment with Acanthus leucostachyus leaf extracts markedly increased the rate of wound contraction,tensile strength,the concentrations of protein,DNA,and hydroxyproline,and the expression of growth factor,as well as promoted epithelialization,compared to the *** addition,Acanthus leucostachyus leaf extracts significantly reduced the expression of pro-inflammatory *** and ultrastructural studies revealed the presence of thicker epithelial layer and smoother surface topography in the extract-treated group compared to the ***:Acanthus leucostachyus leaf extracts show potent wound-healing activity and can be used as a wound healing agent.

Multiplex PCR assay for discrimination of Centrocestus caninus and Stellantchasmus falcatus

Objective: To develop the multiplex PCR method based on the internal transcribed spacer 2 to discriminate the intestinal trematodes, Centrocestus caninus(C. caninus), and Stellantchasmus falcatus(S. falcatus).Methods: Four species of heterophyid trematodes including C. caninus, S. falcatus,Haplorchis taichui and Haplorchoides sp. were amplified and the specific primer was designed based on the internal transcribed spacer 2 region. Two specific primers were used to validate the optimized PCR conditions: the specificity test and the sensitivity ***: Both of these specific primers confirmed the specificity through multiplex PCR reaction which generated both PCR products(231 and 137 bp) in the mixed DNA template of C. caninus and S. falcatus with no cross-reaction with other heterophyid trematodes. The optimum annealing temperature of both primers was 54–59℃. The sensitivity test used the two-fold serial dilution DNA template, which was concentrated between 10 and 0.312 5 ng/mL. The lowest concentration of the DNA template of this multiplex PCR was 2.5 ng/***: The technique described here proved to be a species-specific technique and was found to be a rapid method for the diagnosis of C. caninus and S. falcatus in terms of the larval and adult stages in intermediate and/or definitive hosts in the endemic area.

Prevalence of ruminants fascioliasis and their economic effects in Kashan,center of Iran

Objective:To determine the prevalence and economic losses of Fasciola *** and costs incurred due to liver condemnation from ruminants in Kashan region,center of Iran from 2012 to ***:In this cross-sectional study,a total number of 251 325 slaughtered sheep,goats and cattle(88939 sheep.151 924 goats and 10462 cattle) were examined via necropsy for fasciolasis in industrial Kashan abattoirs located in center of *** average weight was calculated as 1 000 g for sheep,900 g for goat and 5000 g for cattle in this study *** on market value,the average price of a kilo gram of normal liver at Kashan city during the study period was 8 *** amount of main nutrients and vitamins elimination in liver contaminated to fascioliasis was ***:Overall 3.28%of the livers were found to be *** total number of sheep,goats and cattle slaughtered annually in region study,it was estimated that 7505 livers were infected and total annual economic losses of fascioliasis of studied animals was 41 784 USD(based on market prices in study period).Of this.23 360 USD,30240 USD and 15400 USD were associated with the fascioliasis of sheep,goats and cattle,***:This study indicated that the Fasciola *** remains prevalent and causes considerable economic losses in study region in sheep,goats and cattle and presumably,other areas and hosts of Iran,providing baseline data for the future monitoring of this potentially important parasitic infection in the country.

Cloning, expression and protective immunity evalua-tion of the full-length cDNA encoding succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum

1071-bp fragment was obtained from the Schistosoma japonicum (Chinese strain) adult cDNA library after the 3′ and 5′ ends of the incomplete expression sequence tag (EST) of succinate dehydrogenase iron-sulfur protein of Schistosoma japonicum (SjSDISP) were amplified by the anchored PCR with 2 pairs of primers designed according to the EST of SjSDISP and the sequence of multiclone sites of the library vector. Sequence analysis indicated that the fragment was a full-length cDNA with a complete open reading frame (ORF), encoding 278 amino acid residues. The fragment was cloned into prokary- otic expression vector pQE30, and subsequently sequenced and expressed in Escherichia coli. SDS-PAGE and Western-blot analyses showed that the recombinant protein was about 32 kD and could be recognized by the polyclonal antisera from rabbits immunized with Schistosoma japonicum adult worm antigen. Compared with the FCA controls, mice vaccinated with rSjSDISP (test) or rSjGST (posi- tive control) all revealed high levels of specific antibody and significant reduction in worm burden, liver eggs per gram (LEPG), fecal eggs per gram (FEPG) and intrauterine eggs. These results suggest that SjSDISP may be a novel and partially protective vaccine candidate against schistosomiasis. In contrast to the worm burden reduction rate, the higher degree of egg reduction rate in the test group also sug- gested that SjSDISP vaccine may primarily play a role in anti-embryonation or anti-fecundity immunity.

Chemoprophylaxis in malaria:drugs,evidence of efficacy and costs

This review concentrates on different aspects of malaria chemoprophylaxis,namely drug combinations,resistance,impact of malaria prevention in pregnancy and cost effectiveness. A MEDLINE search was performed for all articles with the key word ’Malaria’ in the title field and ’Prophylaxis’ in any *** search was restricted to articles published in English within the last decade(1999-2009).Data sources included review articles published in core clinical journals,cohort studies,interventional studies,case control studies and cross sectional analyses. The mechanism of action,trial evidence of efficacy,side effects and geographical distribution of resistance is discussed for each prophylactic drug *** of prophylaxis in pregnancy and the cost considerations are discussed under two separate sub topics.

Leishmaniasis is a parasitic infection caused by species leishmaniae,which can produce two types of manifestations:visceral and *** south America cutaneous leishmaniasis is more common than visceral leishmaniasis.A case of primary cutaneous leishmaniasis from Bolivia is presented for its *** patient of our case showed an ulcerated lesion of the knee. Montenegro’s intradermal test was ***-stained touch preparation of the skin biopsy revealed amastigotes inside macrophages,consistent with *** patient was treated with meglumine antimoniate intramuscular(20 mg of Sb+/kg/day) three weeks,with complete cicatrization of the lesion.

The tissue destruction characteristic of syphilis infection may be caused by inflammation due to Treponema pallidum and the ensuing immune responses to the *** membrane proteins are thought to be potent inducers of inflammation during the early stages of ***,the actual membrane proteins that induce inflammatory cytokine production are not known,nor are the molecular mechanisms responsible for triggering and sustaining the inflammatory *** the present study,Tp0751 recombinant protein from *** was found to induce the production of proinflammatory cytokines,including TNF-α,IL-1βand IL-6,in a THP-1 human monocyte cell *** signal transduction pathways involved in the production of these cytokines were then further *** inhibition of TNF-a,IL-1β,or IL-6 production was observed following treatment with the SAPK/JNK specific inhibitor SP600125 or with an ERK inhibitor *** contrast,anti-TLR2 mAb,anti-CD14 mAb,and the p38 inhibitor SB203580 significantly inhibited the production of all three *** addition,pyrrolidine dithiocarbamate (PDTC),a specific inhibitor of NF-κB,profoundly inhibited the production of these ***0751 treatment strongly activated NF-κB,as revealed by Western ***,NF-κB translocation was significantly inhibited by treatment with *** results indicated that TLR2,CD14,MAPKs/p38,and NF-κB might be implicated in the inflammatory reaction caused by *** infection.
