Schwann cell transplantation is considered one of the most promising cell-based therapy to repair injured spinal cord due to its unique growth-promoting and myelin-forming properties.A the Food and Drug Administration-approved Phase I clinical trial has been conducted to evaluate the safety of transplanted human autologous Schwann cells to treat patients with spinal cord injury.A major challenge for Schwann cell transplantation is that grafted Schwann cells are confined within the lesion cavity,and they do not migrate into the host environment due to the inhibitory barrier formed by injury-induced glial scar,thus limiting axonal reentry into the host spinal *** we introduce a combinatorial strategy by suppressing the inhibitory extracellular environment with injection of lentivirus-mediated transfection of chondroitinase ABC gene at the rostral and caudal borders of the lesion site and simultaneously leveraging the repair capacity of transplanted Schwann cells in adult rats following a mid-thoracic contusive spinal cord *** report that when the glial scar was degraded by chondroitinase ABC at the rostral and caudal lesion borders,Schwann cells migrated for considerable distances in both rostral and caudal *** Schwann cell migration led to enhanced axonal regrowth,including the serotonergic and dopaminergic axons originating from supraspinal regions,and promoted recovery of locomotor and urinary bladder ***,the Schwann cell survival and axonal regrowth persisted up to 6 months after the injury,even when treatment was delayed for 3 months to mimic chronic spinal cord *** findings collectively show promising evidence for a combinatorial strategy with chondroitinase ABC and Schwann cells in promoting remodeling and recovery of function following spinal cord injury.

Based on in vivo research on the effect of the coating of the extracellular matrix composition of pedicle screws on the conduction and induction of bone formation in young sheep, the aim of this study was to investigate the application of coated pedicle screws in sheep with scoliosis whose spines are under constant development. Methods Four groups of pedicle screws were randomly implanted into bilateral L2-L5 pedicles of 2.5- to 3-month-old sheep. A static experiment was performed on one side and a loading test was performed on the other side by implanting connecting rods at the L2-L3 and L4-L5 segments. The changes in the force on the coated screws and the combination of the surface of the coated screws with the surrounding bone in the growth process of young sheep＇s spines with aging were observed. After 3 months, the lumbar vertebrae with the screws were removed and examined by micro-CT, histological, and biomechanical analyses. Results Under nonloading conditions, there is bone formation around the surfaces of coated screws. The bone forming on the surface of collagen/chondroitin sulfate/hydroxyapatite coating of pedicle screws is the most, the one of the collagen / chondroitin sulfate coating and hydroxyapatite coating is followed, and no significant difference between the two groups. In terms of the trabecular bone morphology parameters of the region of interest around the surface of the pedicle screws, such as bone mineral content, bone mineral density, tissue mineral content, tissue bone mineral density, bone volume fraction, and connection density, those associated with collagen/chondroitin sulfate/hydroxyapatite coatings are largest and those unassociated with coatings are smallest. Under nonloading conditions, the pullout strength of the collagen/chondroitin sulfate/hydroxyapatite-coated screws was largest, and that of the uncoated screws was minimal （P 〈0.01）. A RETROSPECTIVE ANALYSIS OF SURGICAL TREATMENT FOR BREAST MALIGNANT TUMORS

Objective: To review the evolution of the current surgical treatment for breast malignant tumors over the past twenty years in the First Hospital of Jilin University (the former Bethune University of Medical Sciences). Methods: 1195 eligible patients with primary breast malignant tumor diagnosed and surgically treated at the First Teaching Hospital from January 1980 and December 2000 were retrospectively analyzed. Results: The peak frequency was in 40–49 years of age (40.00%), the age of the patients with breast malignant tumors trends to become young. The most common pTNM classification was Stage II. The most common histological type was infiltrating ductal carcinoma (398 patients, 33.31%), and simple carcinoma (279 patients, 23.53%). Modified radical mastectomy was the most common operation procedure performed (779 patients, 65.19%), and was increasingly used while radical mastectomy was adopted decreasingly in recent decade. Conclusion: The variation of operation procedures performed on patients with breast malignant tumors reflected the advance of our understanding of the biology of cancer and the progression of new treatment principles.

In vivo Anti-tumor Effect of siRNA Against STAT3 on Transplanted Lewis Lung Cancer in Mice

This study aims at determining the therapeutic effect of knockdown of signal transducers and activators of transcription3（STAT3） gene expression by short interference RNA（siRNA） on transplanted Lewis lung cancer in mice in vivo. pSilencer 2.1-U6 STAT3 siRNA against STAT3（STAT3 siRNA） was synthesized. Lewis lung cancer cells were inoculated subcutaneously into C57BL/6 mice. Seven days after inoculation, the tumor-bearing mice were randomly divided into 3 groups and received intratumoral injection of （1） vehicle（PBS solution）, （2） vector（negative control）, and （3） STAT3 siRNA. Tumor volume and weight were calculated. Tumors and the lungs were excised for 21 days after inoculation. Expressions of STAT3, vascular endothelial growth factor（VEGF）, and matrix metalloproteinase-2（MMP2） were analyzed by RT-PCR, Western blot, and immunohistochemical staining. HE staining and TUNEL assay were used to confirm the apoptosis of tumors. The synthetic STAT3 siRNA effectively suppressed tumor growth, prevented tumor from pulmonary metastasis, and induced tumor apoptosis in vivo compared with vehicle and vectore in controls. It significantly inhibited STAT3 expression to contribute to downregulation of VEGF and MMP2 expression within tumors in vivo. This study demonstrates that STAT3 siRNA can effectively inhibit the expression of STAT3 gene within tumors, leading to suppression of tumor growth, prevention of cancer from pulmonary metastasis, and enhancing apoptosis of the transplanted Lewis lung cancer in mice in vivo.

Ocular manifestations of WaldenstrSm＇s macroglobulinemia have been described in association with the conjunctiva, cornea, uvea, retina, and periocular adnexa. Only rarely have retinal findings, particularly retinal detachments, been described in the presence of monoclonal hypergammaglobulinemias. This report presents a case of retinal detachment in the presence of Waldenstr6m＇s macroglobulinemia after being treated with Chinese medicine.

Matrix metalloproteinase-9（MMP-9） and p53 genes play an essential role in the multi-step process of tumorigenesis in lung cancer. Single nucleotide polymorphisms（SNPs） of MMP-9 and p53 genes are associated with the risk and progression of many cancers. In this study, we evaluated the association of the R279Q polymorphism of MMP-9 or the A1/A2 polymorphism of p53 gcne with the risk of no-small-cell lung cancer（NSCLC） in Han population of Northeast China. We examined the frequency of SNPs in the two kinds of genes of 50 patients with NSCLC and 50 cancer-free controls frequency-matched by age and sex. Polymerase chain reaction-restriction fragment length polymorphism（PCR-RFLP） technique was used to determine the genotypes. The results indicate that the 279RR genotype in MMP-9 gene and the A1/A2 genotype in p53 gene show a significantly increased risk of NSCLC. Therefore, the MMP-9 279RR and p53 A1/A2 genotypes may be used as markers for susceptibility to NSCLC in Han population of Northeast China.

BACKGROUND Abnormal systemic artery to the left lower lobe is a rare congenital abnormality characterized by anomalous communication between the systemic and pulmonary *** to its rarity,there is limited clinical experience with respect to the diagnosis and treatment of this *** SUMMARY We report a 60-year-old man who presented with a history of hemoptysis for 20 ***-enhanced computed tomography of the chest confirmed the diagnosis of abnormal systemic artery to the left lower lobe,and surgical treatment was *** aberrant artery arising from the descending thoracic aorta was ligated,followed by removal of the left lower *** patient showed good recovery and was discharged 6 d after the *** the 1-year follow-up,the patient had recovered completely,and lung CT showed no abnormal *** We present a case of abnormal systemic artery to the left lower lobe that was successfully managed by surgical resection of the aberrant artery and the left lower *** case report adds to the clinical experience of diagnosing and treating this rare entity.
