Various nanoparticle-based drug delivery systems for the treatment of neurological disorders have been widely ***,their inability to cross the blood–brain barrier hampers the clinical translation of these therapeutic *** are nanoparticles composed of lipid bilayers,which can effectively encapsulate drugs and improve drug delivery across the blood–brain barrier and into brain tissue through their targeting and ***,they can potentially treat traumatic and nontraumatic central nervous system *** this review,we outlined the common properties and preparation methods of liposomes,including thin-film hydration,reverse-phase evaporation,solvent injection techniques,detergent removal methods,and microfluidics ***,we comprehensively discussed the current applications of liposomes in central nervous system diseases,such as Alzheimer's disease,Parkinson's disease,Huntington's disease,amyotrophic lateral sclerosis,traumatic brain injury,spinal cord injury,and brain *** studies related to liposomes are still in the laboratory stage and have not yet entered clinical ***,their application as drug delivery systems in clinical practice faces challenges such as drug stability,targeting efficiency,and ***,we proposed development strategies related to liposomes to further promote their development in neurological disease research.

Regulated cell death is a form of cell death that is actively controlled by *** studies have shown that regulated cell death plays a key role after spinal cord *** and ferroptosis are newly discovered types of regulated cell deaths that have been shown to exacerbate inflammation and lead to cell death in damaged spinal ***,a complex form of cell death that is interconnected with various regulated cell death mechanisms,has garnered significant attention in the study of spinal cord *** injury triggers not only cell death but also cellular survival *** signaling pathways play pivotal roles in influencing the processes of both deterioration and repair in spinal cord injury by regulating pyroptosis,ferroptosis,and ***,this review aims to comprehensively examine the mechanisms underlying regulated cell deaths,the signaling pathways that modulate these mechanisms,and the potential therapeutic targets for spinal cord *** analysis suggests that targeting the common regulatory signaling pathways of different regulated cell deaths could be a promising strategy to promote cell survival and enhance the repair of spinal cord ***,a holistic approach that incorporates multiple regulated cell deaths and their regulatory pathways presents a promising multi-target therapeutic strategy for the management of spinal cord injury.

Pigmented Villonodular Synovitis of the Spine: Two Case Reports and a Literature Review

Introduction: Pigmented villonodular synovitis (PVNS) of the spine is a rare condition, with only a limited number of cases documented in the medical literature. In this study, we review imaging findings of two cases of PVNS arising in the spine. Case 1: A 52-year-old male presented with right thigh pain of unclear etiology. His condition subsequently deteriorated and he was referred to our hospital. MRI revealed a spinal tumor with high intensity on T1-weighted imaging (T1WI) and iso-intensity on T2WI in the right L2/3 intervertebral foramen. The tumor exhibited heterogeneous enhancement on contrast-enhanced MRI. Computed tomography (CT) myelography showed the presence of an epidural tumor within the right L2/3 intervertebral foramen, situated externally to the spinal canal and compressing the dura mater. Notable scalloping was observed in the posterior margin of the L2 vertebral body on CT images. Case 2: A 78-year-old male presented with a complaint of muscle weakness of the left upper limb. His gait gradually deteriorated, which led to suspicion of cervical myelopathy. A tumor was observed on plain MRI and exhibited low to iso-intensity on T1WI and high intensity on T2WI. The tumor was in the left C7/T1 intervertebral segment and the spinal cord was compressed from the left side. CT showed destruction of the medial aspect of the left C7 vertebral arch. Conclusion: In spinal PVNS, the signal intensity on MRI is dependent on the timing of hemorrhage within the tumor, and there are no distinctive features. However, if an extradural tumor is suspected and CT shows bone erosion, scalloping or osteolytic changes, PVNS should be considered as a differential diagnosis.

Clinicopathological and molecular features of HR+/HER2- breast cancer patients with distinct endocrine resistance patterns

Objective: Recurrence continues to be a pivotal challenge among hormone receptor-positive（HR+）/human epidermal growth factor receptor 2-negative（HER2-） breast cancers. In the international consensus guidelines,HR+/HER2-breast cancer relapse patterns are divided into three distinct types: primary resistant, secondary resistant, and endocrine sensitive. However, owing to the lack of cohorts with treatment and follow-up data, the heterogeneity among different recurrence patterns remains uncharted. Current treatments still lack ***: This analysis included data from a large-scale multiomics study of a HR+/HER2-breast cancer cohort（n=314）. Through the analysis of transcriptomics（n=312）, proteomics（n=124）, whole-exome sequencing（n=290）,metabolomics（n=217）, and digital pathology（n=228） data, we explored distinctive molecular features and identified putative therapeutic targets for patients experiencing ***: We explored distinct clinicopathological characteristics, biological heterogeneity, and potential therapeutic strategies for recurrence. Based on a shared relapse signature, we stratified patients into high-and lowrecurrence-risk groups. Patients with different relapse patterns presented unique molecular features in primary tumors. Specifically, receptor tyrosine kinase（RTK） pathway activation in the primary resistant group suggested the utility of RTK inhibitors, whereas mammalian target of rapamycin（mTOR） and cell cycle pathway activation in the secondary resistant group highlighted the potential of mTOR and CDK4/6 inhibitors. Hepatic arterial infusion chemotherapy (HAIC) is an advanced targeted therapeuticapproach for hepatocellular carcinoma (HCC), the most common type ofprimary liver cancer. HAIC has demonstrated significant potential in managingadvanced HCC, particularly in regions with high prevalence rates. Despite itspromise, several challenges and areas for future research remain. Clinical studieshave substantiated the efficacy of HAIC in enhancing survival outcomes forpatients with advanced hepatic carcinoma. Notably, combination therapiesinvolving immune checkpoint inhibitors, such as lenvatinib and programmeddeath-1 inhibitors, have shown substantial improvements in median overallsurvival and progression-free survival compared to systemic *** combination therapies have also exhibited superior response rates anddisease control, with manageable and often less severe adverse events relative tosystemic treatments. This article is based on the review by Zhou et al and aims todiscuss the current status and future directions in the treatment of HCC, emphasizingthe role of HAIC and its integration with novel therapeutic agents.
