Bioactive materials from berberine-treated human bone marrow mesenchymal stem cells accelerate tooth extraction socket healing through the jaw vascular unit

Delayed tooth extraction socket(TES) healing can cause failure of subsequent oral implantation and increase socioeconomic burden on patients. Excessive amounts of M1 macrophages, apoptotic neutrophils(ANs), and neutrophil extracellular traps(NETs) impair alveolar bone regeneration during TES healing. In the present study, we first discovered that conditioned medium(CM) collected from berberine-treated human bone marrow mesenchymal stem cells(BBR-HB-CM) accelerated TES healing. BBR-HB-CM contained bioactive materials that promoted the polarization of macrophages from M1 to M2, impeded the formation of ANs and NETs, and modulated M2 macrophage efferocytosis in vivo and in vitro. Mechanistically, BBR-HB-CM promoted bone formation by inhibiting macrophage-myofibroblast transition and reprogrammed macrophage polarization through p85/AKT/mTOR pathway-dependent autophagy. The 3-methyladenine abolished the therapeutic effects of BBR-HB-CM. Further studies revealed that BBR-HB-CM accelerated TES healing in rats with type 2 diabetes mellitus. Overall, our results demonstrated that BBR-HB-CM had high potential to promote rapid TES healing.

Fast electron-hole recombination issues during titanium dioxide(TiO_(2))photocatalysis limit its application in preventing bacterial infection during bone defect *** this study,TiO_(2)@reduced graphene oxide(rGO)composites were synthesized using a hydrothermal method in which rGO,which possesses very high electrical conductivity,promotes the separation of photoelectron-hole pairs of TiO_(2),thus improving the efficiency of photocatalytic production of reactive oxygen species(ROS).Subsequently,TiO_(2)@rGO composites were introduced into poly-L-lactic acid(PLLA)to prepare bone scaffolds with photocatalytic antibacterial function via selective laser *** results showed that TiO_(2)grew on the surface of rGO and formed a covalent bond connection(Ti-O-C)with rGO.A decreased electrochemical impedance of TiO_(2)@rGO composites was observed,and the transient photocurrent intensity increased from 0.05 to 0.5μA/cm^(2).Analysis of electron spin resonance found that the photocatalytic products of TiO_(2)were·OH and·O^(2-),two kinds of ROS capable of killing bacteria via disrupting the structure of the bacterial membrane in *** experiments showed that the PLLA/TiO_(2)@rGO scaffolds had good antibacterial properties against Escherichia coli and Staphylococcus ***,we report that these scaffolds exhibited both enhanced mechanical properties due to the addition of TiO_(2)@rGO as a reinforcement material and good biocompatibility during cell proliferation.

Histopathological examination is still the gold standard for diagnoses of oral-maxillofacial lesions,but it is invasive and *** coherence tomography(OCT)provides a kind of noninvasive,label-free,real-time and high-resolution imaging *** this study,in order to assess the feasibility of OCT in oral clinical application,fresh excised tissue specimens from 59 patients undergoing oral-maxillofacial surgery were imaged in detail by using a benchtop sweptsource OCT *** is shown that different lesions or tissues can be obviously distinguished based on their different microstructural features in OCT images,and the features are similar to those of their corresponding histopathological *** is proven that OCT has great feasibility and potential as a diagnostic aid for surgeons in oral medicine.

Identification of oral squamous cell carcinoma in optical coherence tomography images based on texture features

Surgical excision is an effective treatment for oral squamous cell carcinoma(OSCC),but exact intraoperative differentiation OSCC from the normal tissue is the first *** a noninvasive imaging technique,optical coherence tomography(OCT)has the nearly same resolution as the histopathological examination,whose images contain rich information to assist surgeons to make clinical *** extracted kinds of texture features from OCT images obtained by a home-made swept-source OCT system in this paper,and studied the identification of OSCC based on different combinations of texture features and machine learning *** was demonstrated that different combinations had different accuracies,among which the combination of texture features,gray level co-occurrence matrix(GLCM),Laws'texture measnres(LM),and center symmetric auto-correlation(CSAC),and SVM as the classifier,had the optimal comprehensive identification effect,whose accuracy was 94.1%.It was proven that it is feasible to distinguish OSCC based on texture features in OCT images,and it has great potential in helping surgeons make rapid and accurate decisions in oral clinical practice.

oral squamous cell carcinoma is one of the most common malignant tumours in the oral and maxillofacial regions and is highly malignant and prone to recur despite the development of various effective treatments,including surgery and ***,it is difficult to ensure the complete elimination of tumour cells,and maxillofacial bone defects caused by surgery are hard to heal by *** addition,chemoradiotherapy can bring serious ***,it is imperative to develop a postoperative therapy to kill residual squamous cancer cells and repair bone defects without any ***,we prepared a 3D scaffold by a 3D printing technique and freeze-drying method,which contained collagen,silk and hydroxyapatite(CSH)and was functionalized with MXene nanosheets(M-CSH).The considerable photothermal effect with long-term stability can significantly kill squamous CAL-27 cancer cells in vitro and inhibit tumour growth in vivo,increasing the probability of the M-CSH scaffold being applied in the photothermal therapy of oral squamous cell ***,the cell proliferation-and osteogenic-related protein expression of mouse embryonic osteogenic precursors(MC3T3-E1)indicated excellent biocompatibility and osteogenic activity of M-CSH *** good compression modulus(52.8362.25 kPa)and in vivo bone formation performance made it possible to be used as reconstructive materials for bone *** scaffold is likely promising in future tissue engineering,especially for the multifunctional treatment of maxillofacial tumours.

Low Temperature Degradation of Alumina-toughened Zirconia in Artificial Saliva

The long-term aging behavior of an alumina-toughened zirconia （ATZ） ceramic in artificial saliva hydrothermal atmosphere was determined and compared with that of tetragonal zirconia polycrystals doped with 3mo1% Y203 （3Y-TZP）. The specimens of 3Y-TZP and ATZ were aged in distilled water and artificial salivary respectively at 134 ~C. Monoclinic phase content was evaluated by X-ray diffraction （XRD） analysis. Specimen strength was determined by a biaxial bending test. Surface screening was done using an atomic force microscope （AFM）. No statistically significant influence of any treatment on strength was demonstrated for either material. However, XRD measurements revealed ATZ＇s anti-aging properties were much better than TZP after the aging treatment. Therefore, ATZ can be applied to the field of dentistry. Artificial saliva in ceramic surface sediments may be useful for stren^heninl ceramics.

Xenogenic organ transplantation has been considered the most promising strategy in providing possible substitutes with the physiological function of the failing organs as well as solving the problem of insufficient donor ***,the xenograft,suffered from immune rejection and ischemia-reperfusion injury(IRI),causes massive reactive oxygen species(ROS)expression and the subsequent cell apoptosis,leading to the xenograft *** previous study found a positive role of PPAR-γ in antiinflammation through its immunomodulation effects,which inspires us to apply PPAR-γ agonist rosiglitazone(RSG)to address survival issue of xenograft with the potential to eliminate the excessive *** this study,xenogenic bioroot was constructed by wrapping the dental follicle cells(DFC)with porcine extracellular matrix(p ECM).The hydrogen peroxide(H_(2)O_(2))-induced DFC was pretreated with RSG to observe its protection on the damaged biological *** staining and transmission electron microscope were used to detect the intracellular ROS *** rat orthotopic transplantation model and superoxide dismutase 1(SOD1)knockout mice subcutaneous transplantation model were applied to explore the regenerative outcome of the *** showed that RSG pretreatment significantly reduced the adverse effects of H2O2on DFC with decreased intracellular ROS expression and alleviated mitochondrial *** vivo results confirmed RSG administration substantially enhanced the host’s antioxidant capacity with reduced osteoclasts formation and increased periodontal ligament-like tissue regeneration efficiency,maximumly maintaining the xenograft *** considered that RSG preconditioning could preserve the biological properties of the transplanted stem cells under oxidative stress(OS)microenvironment and promote organ regeneration by attenuating the inflammatory reaction and OS injury.

Characterization and evaluation of a femtosecond laser-induced osseointegration and an anti-inflammatory structure generated on a titanium alloy

Cell–material interactions during early osseointegration of the bone–implant interface are critical and involve crosstalk between osteoblasts and *** surface properties of titanium implants also play a critical role in cell–material *** this study,femtosecond laser treatment and sandblasting were used to alter the surface morphology,roughness and wettability of a titanium *** and osteoclasts were then cultured on the resulting titanium alloy *** disk groups were tested:a polished titanium alloy(pTi)control;a hydrophilic micro-dislocation titanium alloy(sandblasted Ti(STi));a hydrophobic nano-mastoid Ti alloy(femtosecond laser-treated Ti(FTi));and a hydrophilic hierarchical hybrid micro-/nanostructured Ti alloy[femtosecond laser-treated and sandblasted Ti(FSTi)].The titanium surface treated by the femtosecond laser and sandblasting showed higher biomineralization activity and lower cytotoxicity in simulated body fluid and lactate dehydrogenase *** to the control surface,the multifunctional titanium surface induced a better cellular response in terms of proliferation,differentiation,mineralization and collagen *** investigation of macrophage polarization revealed that increased anti-inflammatory factor secretion and decreased proinflammatory factor secretion occurred in the early response of *** on the above results,the synergistic effect of the surface properties produced an excellent cellular response at the bone–implant interface,which was mainly reflected by the promotion of early ossteointegration andmacrophage polarization.

The complexity of oral ulcerations poses considerable diagnostic and therapeutic challenges to oral *** expert consensus was conducted to summarize the diagnostic work-up for difficult and complicated oral ulcers,based on factors such as detailed clinical medical history inquiry,histopathological examination,and ulceration-related systemic diseases *** only it can provide a standardized procedure of oral ulceration,but also it can improve the diagnostic efficiency,in order to avoid misdiagnosis and missed diagnosis.

Systemic immune monitoring is a crucial clinical tool for disease early diagnosis,prognosis and treatment planning by quantitative analysis of immune ***,conventional immune monitoring using flow cytometry faces huge challenges in large-scale sample testing,especially in mass health screenings,because of time-consuming,technical-sensitive and high-cost ***,the lack of high-performance detection platforms hinders the development of high-throughput immune monitoring *** address this bottleneck,we constructed a generally applicable DNA framework signal amplification platform(DSAP)based on post-systematic evolution of ligands by exponential enrichment and DNA tetrahedral framework-structured probe design to achieve high-sensitive detection for diverse immune cells,including CD4+,CD8+T-lymphocytes,and monocytes(down to 1/100μl).Based on this advanced detection platform,we present a novel high-throughput immune-cell phenotyping system,DSAP,achieving 30-min one-step immune-cell phenotyping without cell washing and subset analysis and showing comparable accuracy with flow cytometry while significantly reducing detection time and *** a proof-of-concept,DSAP demonstrates excellent diagnostic accuracy in immunodeficiency staging for 107 HIV patients(AUC>0.97)within 30 min,which can be applied in HIV infection monitoring and ***,we initially introduced promising DSAP to achieve high-throughput immune monitoring and open robust routes for point-of-care device development.
