Molecular phylogenetic relationships of China Seas groupers based on cytochrome b gene fragment sequences

The classification and evolutionary relationships are important issues in the study of the groupers. Cytochrome b gene fragment of twenty-eight grouper species within six genera of subfamily Epinephelinae was amplified using PCR techniques and the sequences were analyzed to derive the phylogenetic relationships of the groupers from the China Seas. Genetic information indexes, including Kimura-2 parameter genetic distance and Ts/Tv ratios, were generated by using a variety of biology softwares. With Niphon spinosus, Pagrus major and Pagrus auriga as the designated outgroups, phylogenetic trees, which invoke additional homologous sequences of other Epinephelus fishes from GenBank, were constructed based on the neighbor-joining (NJ), maximum-parsimony (MP), maximum-likelihood (ML) and minimum-evolution (ME) methods. Several conclusions were drawn from the DNA sequences analysis: (1) genus Plectropomus, which was early diverged, is the most primitive group in the subfamily Epinephelinae; (2) genus Variola is more closely related to genus Cephalopolis than the other four genera; (3) genus Cephalopolis is a monophyletic group and more primitive than genus Epinephelus; (4) Promicrops lanceolatus and Cromileptes altivelis should be included in genus Epinephelus; (5) there exist two sister groups in genus Epinephelus.

Measurements were made on the contents of protein, lipid, glycogen (PLG) and water, and on caloric values and amino acids, in muscle of three mudskippers Periophthalmis cantonensis, Scarteiaos viridis and Boleophthalmus pectinirostris collected from Haicang, Xiamen. The essential amino acids (EAA) for these fishes were also studied with radioisotopic trace method. The results showed: (1) The content of each component in tested fish muscles differed slightly, and protein was the most important component making up from 6.685% to 9.891% of the wet weight (about 44.21%-50.45% of dry weight); (2) Energy calculated from the sum of protein, lipid and glycogen in wet muscle was low (<4.3kJ/g) in these fishes, especially in B. pectinirostris (<3.1 kJ/g); the ratios of energy to protein content (E/P) also were low (<39.873 -45.535kJ/g); (3) Seventeen amino acids were determined in these three fishes. The content of the same amino acid (among the seventeen) tested in different species and sexes varied slightly. The amounts of methionine, phenylalanine lysine, arginme, histidine, threonine isoleucine and leucine which are indispensable for the needs of human beings and animals were relative-ly high, accounting for 47.35%-48.06% of the total amino acid content. (4) Leucine, isoleucine, arginme, lysine, phenylalanine, tryptophan, methionine, histidine, threonine, and valine, are essential in the diet of the three mudskippers as the radioisotopic trace method using D-[U-^(14) C]-glucose showed little or no radioactivity was incorporated into these ten amino acids.

Study of effects of environmental factors on biochemical compositions of four species of marine single-celled algae

Photosynthetic rates (PR ) of four species of marine single- celled algae were determined by means of 14C-tracer method under the experimental modelling conditions. The effects of environmental factors, such as light, temperature and nutrients, on the biochemical compositions of the algae were studied. The results indicate that the suitable light intensities for the growth of Phaeodactylum tricorntum, Dunaliella spp, Skeletonema costatum and Isochrysis galbana are in the range from 5. 8 X 103 to 15 x 103 lx, showing an increasing trend of photosynthetic rates with the heightened light intensities. The irradiance response of Dunaliella spp. and Isochrysis galbana is significant. The contents of carbohydrate increase, but those of protein decrease, and the contents of lipids change very little. The optimum temperatures for the growth of the four species of algae are 14, 26, 21 and 26t respectively, characterized by the maxima of PR and the contents of carbohydrate, protein and lipid as well as their variations. The apparent activation energies in the photosynthesis process of Phaeodactylum tricornutum, Dunaliella spp., Skeletonema costatum and Isochrysis galbana are 23. 2, 38. 5, 22. 4 and 61. 7 kJ/mol respectively, and the temperature coefficients for the four algae are 1. 74, 1. 74, 1. 38 and 1. 69 respectively. The peaks of PR appeared in some culture media (N/P = 16 ) of Phaeodactylum tricornutum and Skeletonema costatum, and in other media (N/P = 28) of Dunaliella spp. and Isochurysis galbana. The contents and their variations of carbohydrate, protein and lipid of the four species of algae are all maximum when N/P is equal to 16 in culture media.

An investigation was carried out in the Huanghai Sea and the East China Sea to study the quantitative relationship between the abundance of flagellates and the density of suspended particles in the summer of 2001. The results show that the abundance of flagellates varies from 44-12 600 cell/cm^3, and flagellates sometimes constitutes a significant part of suspended particles. The size-spectra of suspended particles can be divided into four categories： flat spectrum, humped spectrum, plankton spectrum and mixed spectrum. In general, the abundance of flagellates varies in proportion to the density of suspended particles. However, their quantitative relations reveal different characteristics in the seawater samples of different types of particle-size spectrum. This is only a preliminary study of the quantitative relationship between flagellates and suspended particles, which might lead to a potential convenient approach to the estimation of flagellate abundance in the sea.

The diversity and distribution of Pronoctiluca, a marine dinoflagellate of enigmatic systematic position, was studied in the vicinity of the Kuroshio and Oyashio Currents, the Philippines, Celebes, Sulu and South China Seas, western and central equatorial and southeast Pacific Ocean. The abundance of Pronoctiluca was high- er, with a wide vertical distribution, in eutrophic temperate regions, whereas it was nearly absent in surface waters of oligotrophic tropical regions. Most of the specimens corresponded to P. spinifera. Pronoctiluca pelagica, covered by hyaline layers and with no flagella, is considered as an encysted form. The bipartition was only observed in P. acuta-P, spinifera forms, that together with the occurrence of intermediate forms between P. spinifera and P. pelagica suggest that they may correspond to developmental stages of a single species. Pronoctiluca is essential to understanding the evolutionary history of the alveolates.

A histological and ultrastructural study was performed on the development of the head kidney, thymus, and spleen in Pseudosciaena crocea aging in range from just hatching to the 60th day after hatching （DAH）. Head kidney was first present on the 3rd DAH. Primordial haemopoietic stem cells were first observed in the head kidney which rapidly differentiate into different cellular types. Progenitor spleen was present on the 4th DAH, located close to the gut, which soon became rich in blood capillaries, red blood cells and thrombocytes. The thymus was obvious on the 4th DAH, and was located on either side of the upper comer of the opercular cavity, closely under the membrance of the opercuhr cavity. The thymus was the last lymphoid organ to appear but showed a quick development. This organ seemed to originate from haemopoietic stem cells migrating from the head region of the kidney. The thymus consisted of outer thymocytic and inner epithelioid zones. There was no obvious demarcation between them, but both zones were visible. Small lymphocytes appeared, or lymphoid organs become lymphoid, in the sequence thymus, head kidney and spleen. Only a small number of lymphocytes appeared in the later stages. Non-specific systems may play an important role in the immunocompetence mechanisms of P. crocea during its early development stage.

The genetic diversity of both wild and reared Pseudosciaena crocea (Richardson) collected from Guan Jing Yang in Ningde, China in May 1999 was investigated by random amplified polymorphic DNA (RAPD) in the present study. The polymorphism and mean difference of the wild population as revealed by RAPD were 18.9% and 0.0960 respectively, and those of the reared stocks were relatively lower, with 16.7% in polymorphism and 0.0747 in mean difference. The genetic distance between the two stocks was 0.0041. From the comprehensive investigation, the main reasons for the loss of genetic diversity were probably overfishing, small number of parents as broodstocks and the debatable artificial ranching. Results from this study also showed that the large yellow croaker populations distributed along Fujian coastal waters including Guan Jing Yang still potentially wide genetic variability. It is suggested that genetic management and prevention should be scientifically conducted in order to maintain and improve the genetic diversity of the P. crocea population .

Study of colloidal phosphorus variation in estuary with salinity

The variation of colloidal phosphorus with salinity in estuary was studied in this paper, which was compared with those of particle and truly dissolved phosphorus with the purpose of finding out the similarities and differences between their behaviors in the estuary. Distribution patterns of phosphorus in particle, colloidal and truly dissolved phase at different salinities and their relationships with suspended particulate matter were also studied to understand the transformation and transporta- tion of colloidal phosphorus in estuarine area. The result showed that the concentrations of total colloidal phosphorus, organic colloidal phosphorus and inorganic colloidall phosphate all descended from river-end to sea-end, illustrating their terrigenous source. *** of organic to total colloidal phosphorus decreased with salinity＇s increment, indicating that organic phosphorus was influenced by scavenging process of colloid more significantly as compared with inorganic phosphate. Both of the phosphorus variation tendencies and the proportion between organic and inorganic phosphorus in colloidal phase was similar with that in truly dissolved phase while different from that in particle phase.

Effect of Sea Level Variation on Tidal Characteristic Values for the East China Sea

Tidal waves in the East China Sea are simulated numerically with POM(Princeton Ocean Model) model for normal mean sea level, 30 cm higher, 60 cm higher, and 100 cm higher, respectively, and the simulated result is compared with the harmonic analysis result of hourly sea level data from 19 tide gauges for more than 19 years. It is indicated that the long-term mean sea level variation affects notably tidal waves in this region. Generally, the tidal amplitude increases when the mean sea level rises, but this relationship may be inverse for some sea areas. The maximal variation of tidal amplitude takes place in the zones near the Fujian coast and the Zhejiang coast, rather than the shallowest Bohai Sea. The maximum increase of M2 amplitude can exceed about 15 cm corresponding to the 60 cm rise of the mean sea level along the Fujian coast. The other regions with large variations of tidal amplitude are those along the Jiangsu coast, the south-east coast of Shandong, and the south-east coast of Dalian. The propagation of tidal waves is also related to mean sea level variation, and the tidal phase-lag decreases generally when the mean sea level rises. Almost all the regions where the tidal phase-lag increases with rising mean sea level are close to amphidromic points, meanwhile the spatial area of such regions is very small. Because the influence of mean sea level variation upon tidal waves is spatially marked, such spatial effect should be considered in calculation of the tidal characteristic value and engineering water level. In the region where the amplitudes of the major tidal constituents increase, the probable maximum high water level becomes higher, the probable maximum low water level becomes lower, and both design water level andcheck water level increase obviously. For example, the design water level at Xiamen increases by 13.5 cm due to the variation of tidal waves when the mean sea level rises 60 cm, the total increase of design water level being 73.5 cm.
