Bacteria with functional DNA repair systems are expected to have low mutation rates due to strong natural selection for genomic ***,our study of the wild-type Streptococcus pneumoniae D39,a pathogen responsible for many common diseases,revealed a high spontaneous mutation rate of 0.02 per genome per cell division in mutation-accumulation(MA)*** rate is orders of magnitude higher than that of other non-mutator bacteria and is characterized by a high mutation bias in the A/T *** high mutation rate may have resulted from a reduction in the overall efficiency of selection,conferred by the tiny effective population size in *** line with this,*** D39 also exhibited the lowest DNA mismatch-repair(MMR)efficiency among *** with the antibiotic penicillin did not elevate the mutation rate,as penicillin did not induce DNA damage and *** lacks a stress response *** findings suggested that the MA results are applicable to within-host scenarios and provide insights into pathogen evolution.

Solar vapor generation is a promising sustainable technology that uses solar distillation to produce fresh water from seawater and wastewater,helping relieve global water resource ***,inspired by naturally grown integrally molded mulberry leaves with a Janus hydrophilic and hydrophobic structure,a novel,simple,and efficient integrated molding method is proposed to break through the limitations of the traditional split manufacturing strategy and realizes the integrated formation of Janus *** on the spontaneous sedimentation characteristics of MXene in silk fibroin solution and its regulation of mesoscopic structure and hydrophilicity of silk fibroin,layered structures with different compositions and hydrophilicities were obtained in one ***,ethanol and glutaraldehyde were added to construct a physical crystallization-chemical crosslinking dual stabilization structure in silk *** evaporator has the evaporation rate of 3.07 kg·m^(-2)·h^(-1) and the efficiency of 86.8%under 1 sun and maintains high evaporation performance under various extreme test conditions including vigorous washing,repeated compression,and high-intensity ultraviolet(UV)***,the evaporator performs well in practical application scenarios,its evaporation rate in the simulated Dead Sea seawater exceeds 2.13 kg·m^(-2)·h^(-1),and more than 99.9%of the salt,heavy metal ions,oil pollution,and dyes are purified.

The Optimization Design of the Nozzle Section for theWater Jet Propulsion System Applied in Jet Skis

The performance of a water jet propulsion system is related to the inlet duct,rotor,stator,and ***,the flow inlet design must fit the bottom line of the hull,and the design of the inlet duct is often limited by stern *** entire section,from the rotor to the nozzle through the stator,must be designed based on system integration in that the individual performance of these three components will influence each ***,the section from the rotor to the nozzle significantly impacts the performance of a water jet propulsion *** study focused on nozzle design and established referable analysis results to facilitate subsequent integrated studies on the design parameters regarding nozzle *** existing studies concentrate on discussions on rotor design and the tip leakage flow of rotors or have replaced the existing complex computational domain with a simple flow ***,research has yet to implement an integrated,optimal design of the section from the rotor to the *** the above,our program conducted preliminary research on this system integration design issue,discussed the optimal nozzle for this section in-depth,and proposed design suggestions based on the *** program used an existing model as the design *** study referred to the actual trial data as the design conditions for the proposed *** prior references’simple flow field form,this study added a jet ski geometry and free surface to the computational *** the linear hull shape was considered,the inflow in the inlet duct would be closer to the actual *** on the numerical calculation result,this study recommends that the optimal nozzle outlet area should be 37%of the inlet area and that the nozzle contour should be ***,for the pump head,static pressure had a more significant impact than dynamic pressure.

Prokaryotes play a fundamental role in global ocean biogeochemical ***,how the abundance and metabolic activity of ecologically distinct subgroups(i.e.,high nucleic acid(HNA)and low nucleic acid(LNA)cells),and their regulating factors,change in response to changing marine environmental conditions remains poorly ***,we delved into the time-evolving dynamic responses of the HNA and LNA prokaryotic subgroups to declining resource availability and selective grazing by protozoa by conducting a 73-day incubation experiment in a large-volume(117,000 L)macrocosm that facilitates community-level *** found that the metabolic activity of the HNA subgroup was higher than that of the LNA subgroup when the macrocosm was resource replete but that the HNA subgroup declined more rapidly than the LNA subgroup as the resources became increasingly scarce,leading to a steadily increasing contribution of LNA cells to prokaryotic ***,as resources in the macrocosm became limited,protozoan grazing preference shifted from the HNA to the LNA subgroup and the contributions of the LNA subgroup to the carbon flow within the macrocosm *** findings highlight the resilience of LNA cells in resource-limited environments,illuminate the critical role of selective grazing by protozoa in balancing distinct prokaryotic subgroups under changing resource conditions,and demonstrate the complex and adaptive interactions between protozoa and prokaryotes across diverse environmental contexts.

Sea cucumber saponins have attracted more attention in recent years due to biological *** is a popular practice to soak sea cucumber in Baijiu at home and being applied to industrial manufacturing in ***,knowledge of the effect of alcohol on the absorption and metabolism of sea cucumber saponins is *** effects of alcohol on digestion,absorption and metabolism of sea cucumber saponins in BALB/c mice were investigated after gavage and tail intravenous *** results showed that the content of saponins in serum and liver was significantly higher under the influence of alcohol than that in the control group after oral *** promoted the absorption of sea cucumber saponins prototype as well as inhibited the process of saponins being transformed into deglycositic metabolites in the small ***,sea cucumber saponins remained in circulation for a long time and alcohol slowed down the clearance of sea cucumber saponins under the influence of alcohol after intravenous *** confirmed the feasibility of marinating sea cucumber in Baijiu to improve the efficacy of saponins and provides an important theoretical basis for the utilization of sea cucumber and the development of sea cucumber liquor.

Expression and characterization of a novel GH128 laminarinase OUC-BsLam26 harboring both hydrolyzing and transferring activities

Laminarin oligosaccharides (LOSs) with a specific degree of polymerization prepared through the laminarin degradation via laminarinase present more significant nutritional functions and application values. Human intestinal bacteria are promising potential producers of novel carbohydrate-active enzymes with unique properties. Here， a novel glycoside hydrolase family 128 (GH128) laminarinase OUC-BsLam26 from the intestinal bacterium Bacteroides sp. CBA7301 was heterologously expressed and characterized. The recombinant OUC-BsLam26 with a molecular mass of 49.86 kDa exhibits highest activity (6.60 U/mg) at 45 ℃ and pH 6.0， which shows noticeable temperature and pH stability. The purified OUC-BsLam26 could degrade laminarin via an endo-type mode with the generation of laminaripentaose， laminaritetraose， laminaritriose， and laminaribiose， among them， laminaritetraose is the principal product， which accounts for 45.25% of the total products， which is significantly different from the reported GH128 laminarinases. The minimum recognition substrate of OUC-BsLam26 is laminarihexaose. Furthermore， OUC-BsLam26 also could catalyze the transglycosylation process with the production of some novel glycosides. Altogether， the intestinal bacterium Bacteroides sp. CBA7301 contains laminarinase with unique product composition and OUC-BsLam26 is a hopeful bio-catalyst with the potential to produce laminaritetraose and some novel glycosides.

作为微纳米机械谐振器主要的能量耗散源之一,支撑损失主要采用未考虑中面拉伸效应的线性理论进行研究.然而该理论却无法适用于大挠度非线性振动状态谐振器的支撑损失.为此,我们研究了考虑几何非线性和轴向预拉力的两端固支微梁谐振器处于面内弯曲振动模式时的支撑损失,旨在准确模拟大挠度振动下高性能谐振器的支撑损失.本文不仅考虑了支撑结构与谐振器连接区域受中面拉伸产生的正应力作用而引起的支撑损失,也考虑了受剪应力作用而引起的支撑损失,并且理论结果与有限元模拟以及文献中实验结果一致.另外,本文还研究了轴向预拉力对谐振器非线性振动特性的影响,并讨论了谐振器的振幅、长高比以及轴向预拉力对支撑损失的影响.理论结果表明,相比于线性振动状态下谐振器的支撑损失,由谐振器大变形振动产生的中面拉伸效应会引起更大的支撑损失,并且会随非线性振幅的增大而增大.本文提出的非线性支撑损失模型有望对评估非线性振动谐振器的支撑损失以及阐明支撑损失随非线性振幅增大的现象提供理论指导.

Extreme waves have a profound impact on coastal infrastructure;thus,understanding the variation law of risky analysis and disaster prevention in coastal zones is *** paper analyzed the spatiotemporal characteristics of extreme wave heights adjacent to China from 1979 to 2018 based on the ERA5 *** extreme value analysis is undertaken in eight repre-sentative points to investigate the trends in the values of 50-and 100-year wave *** show that the mean value of extreme waves is the largest in the eastern part of Taiwan Island and the smallest in the Bohai Sea from 1979 to *** the extreme wave height in the northeastern part of Taiwan Island shows a significant increase trend in the study *** analysis shows remarkable variations in the values of 50-and 100-year significant wave heights in eight *** the annual mean change,E1,E2,S1,and S2 present an increasing trend,while S3 shows a decreasing *** points for the seasonal mean change demon-strate an increasing trend in spring and winter,while other points show a decreasing trend in summer and ***,the E1 point growth rate is large in autumn,which is related to the change in typhoon intensity and the northward movement of the typhoon path.

In this technologically advancing world,the demand for more energy,oil and gas production is rapidly *** accomplish this,people have inclined more towards completely floating offshore structures,deployed in deep waters.A semi-submersible is selected in the present study,due to its better response characteristics and stability under harsh environmental *** semi-submersible is position restrain with spread mooring lines incorporated with submerged buoy at different locations has been studied.A detailed numerical analysis is carried out using Ansys Aqwa for dynamic response analysis of semi-submersible under the combination of wind,wave,and current forces for 0°,45°,and 90°*** was observed that damping ratios and natural periods had been affected based on the position and number of submerged buoys in the mooring ***,reduction in mooring force after incorporating buoy in the mooring lines was ***,a Matlab code based on the S-N curve approach was generated and employed to investigate the fatigue damage of mooring lines under dynamic variation of mooring *** pegged with submerged buoys,fatigue life of mooring lines is increased under intact and postulated damaged mooring ***,coupling of motion responses of semi-submersible is observed,and unbounded response is not seen in any degrees-offreedom,even during damaged condition of mooring lines.
