Objective:To study the clinical effect of under the diaphragm by stasis soup combinated with point thread-embedding on infertility patients with obesity-type polycystic ovary syndrome (PCOS) and evaluate the the rate of ovulation and ***:Selecting 100 infertility patients with obesity-type PCOS, then they were randomly divided into control group (n=80) and observation group (n=80). The control group patients were given metformin+ under the diaphragm by stasis soup, the observation group were given metformin + exenatide + point thread-embedding, they were treated 3 months. Before and after treatment, we observed the changes of body mass index (BMI), waist-to-hipratio (WHR), fasting plasma glucose (FBG), 2-hour postprandial blood glucose (2hPBG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C), triglycerides (TG), homeostasis model assessment of insulin resistance (HOMA-IR), fasting plasma insulin (FINS), follicule-stimulating hormone (FSH), luteinizing hormone (LH), LH/FSH, estrogen (E2), and testosterone (T). Prognostic value of germline BRCA1 or BRCA2(gBRCA1/2) mutations in epithelia ovarian cancer(EOC) remain controversial,especially for their roles in long-term *** previously reported the largest study of gBRCA1/2 mutation prevalence in Chinese EOC *** aim of this study is to further illustrate correlation of residual disease(RD) and survival in BRCA-associated EOC in *** the current cohort of 615 EOC patients from a Chinese EOC genome-wide association study,overall,we did not find any significant difference between gBRCA1/2 mutation carriers and non-carriers in both progressionfree survival(PFS) and overall survival(OS)(19.3 vs.18.1 months and 77.2 vs.73.2 months,P=0.528 and 0.147,respectively).Same results were found in subgroups of patients with complete cytoreduction(RD=0),but mutation carriers had a better PFS and OS than non-carriers in the subgroup of incomplete cytoreduction(RD>0)(18.5 vs.15.1 months and 68.5 vs.54.3 months,P=0.046 and 0.038,respectively).Our findings provided the evidence that ovarian cancer survival did not differ between gBRCA1/2 mutation carriers and non-carriers,which likely attributed to more than 37%of the patients with no gross residual disease.

Hyperglycemia fluctuation is associated with diabetes mellitus(DM) complications when compared to persistent *** studi es have shown that paeoniflorin(PF),through its anti-apoptosis,anti-inflammati on,and anti-thrombotic properties,effectively protects against cardiovascular an d cerebrovascular ***,the mechanism underlying the protection fr om PF against vascular injuries induced by hyperglycemia fluctuations remains poorly ***,we investigated the potential protective role of PF o n human umbilical vein endothelial cells(HUVECs) subjected to intermittent gl ucose levels in vitro and in DM rats with fluctuating hyperglycemia in vivo.A remarkable increased apoptosis associated with elevated inflammation,increase d oxidative stress,and high protein level of PKCβ1 were induced in HUVECs by intermittently changing glucose for 8 days,and PF recovered those detrime ntal ***333531,a potent PKCβ1 inhibitor and metformin manifested si milar ***,in DM rats with fluctuating hyperglycemia,PF prote cted against vascular damage as what have been observed in *** toget her,PF attenuates the vascular injury induced by fluctuant hyperglycemia throu gh oxidative stress inhibition,inflammatory reaction reduction,and PKCβ1 prot ein level repression,suggesting its perspective clinic usage.

Background: The histone code is an established epigenetic regulator of early embryonic development in *** lysine residue K9 of histone H3(H3 K9) is a prime target of SIRT1, a member of NAD+-dependent histone deacetylase family of enzymes targeting both histone and non-histone substrates. At present, little is known about SIRT1-modulation of H3 K9 in zygotic pronuclei and its association with the success of preimplantation embryo development. Therefore, we evaluated the effect of SIRT1 activity on H3 K9 methylation and acetylation in porcine zygotes and the significance of H3 K9 modifications for early embryonic ***: Our results show that SIRT1 activators resveratrol and BML-278 increased H3 K9 methylation and suppressed H3 K9 acetylation in both the paternal and maternal pronucleus. Inversely, SIRT1 inhibitors nicotinamide and sirtinol suppressed methylation and increased acetylation of pronuclear H3 K9. Evaluation of early embryonic development confirmed positive effect of selective SIRT1 activation on blastocyst formation rate(5.2 ± 2.9% versus 32.9 ± 8.1% in vehicle control and BML-278 group, respectively; P ≤ 0.05). Stimulation of SIRT1 activity coincided with fluorometric signal intensity of ooplasmic ubiquitin ligase MDM2, a known substrate of SIRT1 and known limiting factor of epigenome ***: We conclude that SIRT1 modulates zygotic histone code, obviously through direct deacetylation and via non-histone targets resulting in increased H3 K9 me3. These changes in zygotes lead to more successful pre-implantation embryonic development and, indeed, the specific SIRT1 activation due to BML-278 is beneficial for in vitro embryo production and blastocyst achievement.
