In the present study, four different proteases (pepsin, papain, bromelain and ficin) were screened with a murine monoclonal antibody OC859 , in order to verify whether different digestion procedures could improve yield and stability of the F(ab’)2 or Fab fragments. The yields of F(ab’)2 or Fab fragments from digestion with pepsin, papain , bromelain and ficin were respectively 20. 3+/-2. 0%, 50. 5+/-5. 0%,74. 4+/-2. 7% and 82. 8 +/-10. 2% of the theoretical maximum. Immunoreactivity in a noncompetitive solid-phase radioimmunoassay (SPRIA) of the fragments generated by the four proteases were respectively 10+/-5%, 36+/-5%, 60+/-6% and 75+/-6%of the intact OC859 IgG. These results suggested that the fragmentation of OC859 with ficin gave a higher yield of superior immunoreactive fragments.

Objective: To study the clinical effects and side effects of Estramustine phosphate (EMP) on the treatment of endometrial carcinoma. Methods: Fifty-eight patients with endometrial carcinoma diagnosed in our hospital from Oct. 1996 to Feb. 1998 were randomly divided into 3 groups and clinically observed. EMP group (n=21): after oral EMP 280 mg, bid, for 21 days, surgical operation followed in one week. Radiotherapy (RT) group (n=19): surgical operation was preformed after intra-cavity irradiation with half of the standard dosage. Control group (n=18): surgical operation alone. Histopathological changes in the samples from the removed uterus were observed. Estrogen receptor/progestin receptor (ER/PR) and nuclear proliferate antigen (ki-67) index of the endometrial carcinoma tissues of the EMP group were tested by immunohistochemical methods. Results: The microscopic changes induced by irradiation were much heavier than those induced by chemotherapy. In EMP group, 5/21 cases were found with no tumor lesion in the postoperation samples, all of those 5 cases being with ER strong positive (++) and 4/5 cases well differentiated tumor before chemotherapy. In RT group, the tumor lesion was disappeared in 6/19 cases, and 5 cases of which being with the moderate differentiation. No significant difference was shown between those two groups. No any histopathological changes were seen in control group. Immunohistochemical tests revealed a significant decrease in ER staining after EMP treatment and a decrease in ki-67 index, especially for the ER positive tumors, ki-67 index reduced significantly from 49.5% before medication to 35.1% after medication (P<0.05). Only 5 cases in EMP group reported slight nausea and vomit at the beginning of taking medicine. No changes in body weight, blood pressure, WBC count, or liver and kidney functions were seen at all. However, some patients experienced symptoms and signs such as darkening of areola and perineum, an increase in vaginal discharge

OBJECTIVE: To observe the effect of herb-partitioned moxibustion(HPM) for primary ***: Six hundred and forty patients were randomized assigned(1∶1) to HPM group and control group. Duration of treatment was 3 months with 3 month follow-up. The primary outcome was pain relief measured by visual analogue scale(VAS). The second outcomes were Cox Menstrual Symptom Scale(CMSS), menstrual pain duration and frequency of analgesics usage. The exploratory outcome included quality of life,RESULTS: After the 3-month treatment and follow-ups, the pain intensity measured by VAS was significantly reduced in both groups compared with baseline(P < 0.05), and it was significantly decreased in HPM group than that of control group(P < 0.001). The higher proportion of participants in the HPM group had a decrease of at least 50% in VAS at the end of treatment, as compared with the control group(P < 0.001). At the 3 rd and 6 th month, the menstrual pain duration, CMSS score and frequency of analgesics usage in HPM group were significantly lower than those of control group(P < 0.05). After 3 month treatment and follow-ups,the scores of physical, psychological, social and environmental domains were significantly increased than baseline in both groups(P < 0.05),and the sores of physical, psychological and environmental domains were significantly higher in HPM group than those of control group(P < 0.05).CONCLUSION: Herb-partitioned moxibustion reduced menstrual pain and improved quality of life,these were sustained for up to 3 months after treatment. Further research is needed to understand long term effect and the mechanism of the intervention.

Objective:To observe the anti-tumor effect of matrine combined with cisplatin on U14 rat models of cervical ***:A total of 80 female Kunming rats were used to establish U14 rat models of cervical cancer and then divided into groups Ⅰ,Ⅱ,Ⅲ and Ⅳ,with 20 rats in *** Group Ⅰ,the control group,injection of normal saline was given around the *** Group Ⅱ,injection of 2 mg/kg cisplatin was given around the *** Group Ⅲ,injection of 75 mg/kg matrine was given around the tumors while the combined injection of matrine and cisplatin was given for Group Ⅳ with the same doses as Groups Ⅱand Ⅲ.The animals were sacrificed 10 d after the injection and tumors were taken out for the comparisons of tumor weights after injection and calculation of anti-tumor rates,while thymus and spleen were taken for thymus index and spleen *** in eyeball was collected for determination of changes in serum creatinine and urea nitrogen *** of tumor issue were prepared and morphological changes in tumor tissue cells were observed by using immunohistochemistry ***:After injection,the thymus index and spleen index in Groups Ⅲ and Ⅳ were significantly higher than those in Groups Ⅰ and Ⅱ(P<0.05)while the two indexes in Group Ⅱ were significantly lower than Group Ⅰ(P<0.05).The tumor weights in Groups Ⅱ and Ⅳ were significantly smaller than those in Groups Ⅰ and Ⅲ(P<0.05) with significantly higher anti-tumor rates than Groups Ⅰ and Ⅲ(P<0.05).The serum creatinine and urea nitrogen levels in Groups Ⅲ and Ⅳ were significantly lower than Group Ⅱ(P<0.05) and the two indicators in Group Ⅲ were significantly lower than those in Group Ⅳ(P<0.05).The observation under the histological microscope showed densely arranged tumor cells in Group Ⅰ,growing as a crumby structure and diffuse appearance,with hyperchromatic and large nuclei,and abundant *** the case of Group Ⅱ,it showed less tumor cells,with extensive degenerative necrosis,sparse arrangement

To investigate the relationship between inducible nitric oxide synthase (iNOS) and the early spontaneous abortion. , in situ hybridization and immunohistochemistry were used to detect the expression of iNOS in trophoblasts in the early pregnancy with and without spontaneous abortion (group Ⅰ and group Ⅱ). By light microscopy and computer color magic image analysis system (CMIAS), light density (D) and the positive cell number per statistic square (N/S) in situ hybridiza- tion were used to analyze the positive cell index, while total positive cells (N) and the positive unit (Pu) were used in immunohistochemistry. By in situ hybridization, D and N/S in trophoblasts were 0.35±0. 028, 0. 07±0. 011 respectively in group Ⅰ and 0. 18±0.016,0. 015±0. 003 in group Ⅱ . In terms of immunohistochemical staining, N and Pu were 0. 058±0.007, 11. 94±2. 01 in group Ⅰand 0.013±0.009, 1.08±0. 35 in group Ⅱ in trophoblasts. Significant differences existed between two groups. It is concluded that the higher nitric oxide produced by the higher expression of iNOS in tro- phoblasts might play an important role in the early spontaneous abortion.

BACKGROUND Coronavirus disease 2019(COVID-19)has spread rapidly,resulting in a pandemic in January *** studies have focused on the natural history and consequences of acute liver injury(ALI)in mild or asymptomatic COVID-19 patients,manifested by elevated aminotransferase *** is usually expected for severe COVID-19 ***,we present a COVID-19 case with mild respiratory symptoms and significantly elevated alanine aminotransferase(ALT)and aspartate aminotransferase(AST)*** SUMMARY A 60-year-old woman without medical history or chronic illness received three COVID-19 vaccinations since the start of the *** patient was infected with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)and presented with mild symptoms on July 12th,***-recovery,she underwent an examination at our hospital on August 30th,*** and ALT levels in the liver function test were 207 U/L(normal value<39,5.3-fold increase)and 570 U/L(normal value<52,10.9-fold increase),*** patient was diagnosed with ALI,and no treatment was *** following week,blood tests showed a reduction in both levels(ALT 124 U/L,AST 318 U/L).Two weeks later,AST and ALT levels had decreased to near the expected upper limits(ALT 40 U/L,AST 76 U/L).CONCLUSION Clinicians should pay attention to liver function testing during COVID-19 recovery regardless of the disease’s severity.

Dysregulation of the TGF-β Postreceptor Signaling Pathway in Cell Lines Derived from Primary or Metastatic Ovarian Cancer

Transforming growth factor beta (TGF β) may cause cell cycle arrest, terminal differentiation, or apoptosis in most normal epithelial cells, whereas most malignant cell lines are resistant to TGF β. Mechanisms of resistance to TGF β caused by modulation of cell cycle regulators and/or inactivation of components of the TGF β signaling transduction pathway such as C myc and Smad4 are not well understood. To investigate the potential association between loss of sensitivity to TGF β and expression status of transforming growth factor receptor Ⅱ (TβRⅡ), Smad4, CDC25A and C myc in 14 cell lines derived from ovarian cancer, the expression levels of these genes were detected by semi quantitative RT PCR. Normal ovarian surface tissues were used as controls. The expression of TβRⅡ was detectable in all of 14 cell lines. The expression of Smad4 was decreased in 10 cell lines and 9 cell lines overexpressed CDC25A, as compared to normal controls. CDC25A gene was overexpressed with 88 % (8/9) in tumorigenic cell lines as determined by xenografts in nude mice, and only in 20 % (1/5) of non tumorigenic cell lines ( P <0.05). C myc was not overexpressed in any of these cell lines. The loss of sensitivity to TGF β of cell lines derived from ovarian cancers may be related to a decreased expression of Smad4, which mediates TGF β induced growth inhibition, and/or an overexpression of CDC25A. This overexpression of CDC25A correlates with increased tumorigenicity of ovarian cancer cell lines. The loss of sensitivity to TGF β is not associated with a lack of TβRⅡ.

Objective:To evaluate the efficiency of p16/Ki-67 dual stain used as a triage in cervical cancer ***:In this study,we did 468 p16/Ki-67 dual stain in human papillomavirus(HPV)16/18-positive or 12 other high-risk HPV(OHR-HPV)positive Thinprep cytologic test(TCT)atypical squamous cells of undetermined significance(ASCUS)/lower-grade squamous intraepithelial lesion(LSIL)*** evaluated the sensitivity,specificity,positive predictive value(PPV)and negative predictive value(NPV)of the triage ***:The sensitivity,specificity,PPV and NPV of p16/Ki-67 dual stain in HPV 16/18-positive women were91.5%/68.4%,77.0%/75.0%,73.9%/59.1%and 92.8%/81.8%.In 12 OHR-HPV positive TCT ASCUS/LSIL women,the results were 79.1%/95.0%,88.5%/66.7%,88.5%/70.4%and 89.2%/94.1%.The risk of precancerous lesions in p16/Ki-67 dual stain positive cases was much higher than before,and the negative cases had lower ***,there was no cervical intraepithelial neoplasia(CIN)III case missed after triaged by p16/Ki-67 dual-stained *** p16/Ki-67 dual-stained cytology positive women with benign pathology or CIN I,the 1-year progression rate is 20.5%and in p16/Ki-67 dual-stained cytology negative women,the 1-year progression rate is5.6%.Conclusions:hr-HPV genotyping test plays an important role in cervical cancer screening.p16/Ki-67 dual stain may be a promising triage *** for chronic cervicitis or CIN I patients,a positive p16/Ki-67 dual-stained cytology suggests a high risk in progression and need to be followed up closely.
