Comparison of antioxidant activity of insulin, Ocimum gratissimum L., and Vernonia amygdalina L. in type 1 diabetic rat model

OBJECTIVE： Diabetes mellitus （DM） is known to be associated with increase of oxidative stress products. The direction of effect of any treatment on these products could therefore be a reliable measure of its efficacy on DM. So the aim of this study was to investigate the activity of insulin, Ocimum gratissimum L. （og） and Vernonia amygdafina L. （VA） on oxidative stress ***： Thirty-six female Wistar rats weighing 150-200 g were randomly divided into six groups of six rats each. Thirty rats were induced for type 1 DM （DM1） with a single intraperitoneal administration of 65 mg/ kg body weight of streptozotocin. Group 1 was normal control and was administered distilled water while Group 2 served as DM1 control group; Groups 3, 4, 5 and 6 were diabetic rats treated with 208 mg/kg og （DM1 ＋ og）, 52 mg/kg VA （DM1 ＋ VA）, 208 mg/kg og ＋ 52 mg/kg VA （DMI＋og ＋VA） and 0.16 IU insulin （DM1 ＋ insulin） respectively. Determination of methemoglobin and sulfhemoglobin was achieved by the absorption spectrum principle. Red blood cell （RBC） catalase was assayed by continuous spectrophotometric ***： The RBC catalase concentration was significantly decreased in the DM1 and DMI＋VA groups when compared with the normal control. DM1 ＋ og significantly increased RBC-catalase when compared to DMI. The methemoglobin concentration was significantly reduced in the DM1, DM1 ＋ VA, DM1 ＋ og ＋ VA and DM1 ＋ insulin groups when compared to the normal control group. The sulfhemoglobin concentration was significantly increased in the diabetic control and the diabetic treated groups when compared to the normal control. DM1 ＋ og reduced the sulfhemoglobin concentration when compared to DM1. AIM To investigate the role of minimally invasive surgery for gastric cancer and determine surgical, clinical, and oncological *** This is a propensity score-matched case-control study, comparing three treatment arms: robotic gastrectomy(RG), laparoscopic gastrectomy(LG), open gastrectomy(og). Data collection started after sharing a specific study protocol. Data were recorded through a tailored and protected web-based system. Primary outcomes: harvested lymph nodes, estimated blood loss, hospital stay, complications rate. Among the secondary outcomes, there are: operative time, R0 resections, POD of mobilization, POD of starting liquid diet and soft solid diet. The analysis includes the evaluation of type and grade of postoperative complications. Detailed information of anastomotic leakages is also *** The present analysis was carried out of 1026 gastrectomies. To guarantee homogenous distribution of cases, patients in the RG, LG and og groups were 1:1:2 matched using a propensity score analysis with a caliper = 0.2. The successful matching resulted in a total sample of 604 patients(RG = 151; LG = 151; og = 302). The three groups showed no differences in all baseline patients characteristics, type of surgery(P = 0.42) and stage of the disease(P = 0.16). Intraoperative blood loss was significantly lower in the LG(95.93 ± 119.22) and RG(117.91 ± 68.11) groups compared to the og(127.26 ± 79.50, P = 0.002). The mean number of retrieved lymph nodes was similar between the RG(27.78 ± 11.45), LG(24.58 ± 13.56) and og(25.82 ± 12.07) approach. A benefit in favor of the minimally invasive approaches was found in the length of hospital stay(P < 0.0001). A similar complications rate was found(P = 0.13). Current understanding and dispute on the function of the Wnt signaling pathway effector TCF7L2 in hepatic gluconeogenesis

Approximately 10 years ago,the Wnt signaling pathway effector TCF7L2(ZTCF-4)was recognized as a type 2 diabetes(T2D)risk gene through a genome wide association study(GWAS).As the correlation between TCF7L2 polymorphisms and T2D susceptibility has been reproducibly observed by numerous follow-up investigations among different ethnic groups,great efforts have been made to explore the function of TCF7L2 in metabolic organs including the pancreas,liver and adipose *** these explorations have enriched our general knowledge on the Wnt signaling cascade in metabolic homeostasis,studies conducted to date have also generated controversial *** I will provide a brief review on the Wnt signaling pathway as well as the milestone GWAS discovery and the follow-up studies.I will then discuss the two different opinions on the correlation between TCF7L2 variants and T2D risk,a gain-of-function event versus a loss-of-function *** will be followed by summarizing the relevant investigations on the metabolic function of hepatic TCF7L2 and presenting our view on the discrepancy and perspectives.

A facile synthesis of ω-aminoalkyl ammonium hydrogen phosphates

A series of og-aminoalkyl ammonium hydrogen phosphates were synthesized through a simple and efficient three-step method. The starting materials, co-aminoalkyl alcohols （AC-n, with carbon number n = 3, 4, 5, 6）, were amino-protected with 9- fluorenylmethyl chloroformate （Fmoc-C1）, followed by phosphorylation with POCI3 and deprotection in piperidine/DME The structures of each intermediate and final product were confirmed by 1H NMR, FTIR and mass spectrum. The yield of each step was about 77-92%, with a total yield higher than 56%. This new method was superior in low-cost raw materials, mild reaction temperatures （0-25 ℃） and easy purification methods.

POTENTIATION OF DOCETAXEL ANTITUMOR ACTIVITY BY BATIMASTAT AGAINST MOUSE FORESTOMACH CARCINOMA

Objective . To improve the treatment of tumors, we studied the combined effects of docetaxel and batimastat (BB 94) on mouse forestomach carcinoma (MFC), and compared them with doxorubicin. Methods and results. In vitro, growth curve analysis, MTT assay and clonogenic assay used to determine the cytotoxic effect of docetaxel or/and BB 94 on MFC cell showed that docetaxel but not BB 94 had a significant cytotoxicity, and the effect of docetaxel wasn’t enhanced by BB 94. In early stage MFC tumor model, obvious antitumor effect of docetaxel or doxorubicin given i.v. at maximum tolerated dose (MTD, docetaxel: 20mg/kg; doxorubicin: 6mg/kg) every 4 days for 3 injections (q4d×3), even that of BB 94 (30mg/kg i.p. qd×20) was observed. Tumor growth inhibition was greater for docetaxel batimastat (96.0%) than for doxorubicin-batimastat (88.0%), docetaxel (89.0%), doxorubicin (68.0%) and BB 94 (33.0%), and the effect of docetaxel could be potentiated by BB 94. Docetaxel also showed activity against advanced stage MFC tumor in dose-dependent manner, and was more effective at MTD than doxorubicin with 4/5 regressions, 46.5 days tumor growth delay and 2.8log10 tumor cell kill. Conclusion. Our results suggest that in the MFC model with dose and schedule used, docetaxel is an effective cytotoxic new drug against MFC tumor and BB 94 enchances the antitumor activity of docetaxel.
