Endoplasmic reticulum stress and autophagy in cerebral ischemia/reperfusion injury:PERK as a potential target for intervention

Several studies have shown that activation of unfolded protein response and endoplasmic reticulum(ER)stress plays a crucial role in severe cerebral ischemia/reperfusion *** occurs within hours after cerebral ischemia,but the relationship between ER stress and autophagy remains *** this study,we established experimental models using oxygen-glucose deprivation/reoxygenation in PC12 cells and primary neurons to simulate cerebral ischemia/reperfusion *** found that prolongation of oxygen-glucose deprivation activated the ER stress pathway protein kinase-like endoplasmic reticulum kinase(PERK)/eukaryotic translation initiation factor 2 subunit alpha(e IF2α)-activating transcription factor 4(ATF4)-C/EBP homologous protein(CHOP),increased neuronal apoptosis,and induced ***,inhibition of ER stress using inhibitors or by si RNA knockdown of the PERK gene significantly attenuated excessive autophagy and neuronal apoptosis,indicating an interaction between autophagy and ER stress and suggesting PERK as an essential target for regulating *** autophagy with chloroquine exacerbated ER stress-induced apoptosis,indicating that normal levels of autophagy play a protective role in neuronal injury following cerebral ischemia/reperfusion *** from this study indicate that cerebral ischemia/reperfusion injury can trigger neuronal ER stress and promote autophagy,and suggest that PERK is a possible target for inhibiting excessive autophagy in cerebral ischemia/reperfusion injury.

Willingness to pay for colorectal cancer screening in Guangzhou

AIM To measure the willingness to pay for colorectal cancer screening in Guangzhou, and to identify those factors associated with it. METHODS A face-to-face questionnaire survey for pre-screening population from free and non-free colonoscopy districts was used to collect information on demographic characteristics, health behaviours, the intention of the cancer screenings and willingness to pay for colorectal cancer screening. A total of 1243 participants who took part in the pre-screening for colorectal cancer in Guangzhou were collected in the study. Categorical data were compared using the χ~2 test to analyse significant differences. non-conditional logistic regression and multi-class logistic regression were also performed for multivariate analysis and to estimate the odds *** The percentage of participants willing to pay for colorectal cancer screening was 91.7%. "Unnecessary" was the dominant reason that participants gave for their unwillingness, accounting for 63.1%. Of those who were willing to pay, 29.2%, 20.7%, 14.8%, 13.0% and 22.4% of participants were willing to pay less than $100, $100-$199, $200-299, $300-$399 and more than $400, respectively. non-logistic regression analysis showed that respondents who were male, had a high level of education, were from the family with more children/older to raise, and accepted colorectal cancer screening were willing to pay for this screening. Multi-class logistic regression analysis showed that respondents with higher annual household income per capita, from government and private enterprises, government agency/institution and peasants, and less family medical expenditure were willing to pay *** Willingness to pay for colorectal cancer screening in Guangzhou is high, but the amount of willing to pay is not much.

Introduction:China faces a growing burden of chronic obstructive pulmonary disease(COPD).Previous mortality estimations were primarily based on the underlying cause of *** study analyzed COPD-associated death and its comorbidities using all COPD cases listed on the chain of events on death ***:A retrospective analysis of the National Mortality Surveillance System(NMSS)was conducted to estimate COPD-associated mortality from 2014 to ***-standardized mortality rates(ASMRs)were calculated stratified by sex,region,and *** regression was used to estimate the average annual percentage change(AAPC)during the study ***:From 2014 to 2021,the ASMR of COPD decreased from 91.85 to 45.90 per 100,000 *** but uneven decreases in COPD mortality were observed across gender[females:AAPC:−11.2%,95%confidence interval(CI):−11.9 to−10.4%;males:AAPC:−8.0%,95%CI:−9.2 to−6.8%],regions(eastern:AAPC:−10.7%,95%CI:−11.5 to−9.9%;central:AAPC:−9.9%,95%CI:−10.9 to−8.9%;western:AAPC:−7.7%,95%CI:−10.6 to−4.7%),and residential areas(urban:AAPC:−10.9%,95%CI:−12.3 to−9.5%;rural:AAPC:−8.3%,95%CI:−9.1 to−7.4%).Other than COPD,cardiovascular diseases and respiratory conditions were the major underlying causes of death in COPDassociated ***:COPD is a significant comorbidity of other disorders in *** COPDassociated mortality substantially decreased from 2014 to 2021,the burden remained high in underdeveloped regions.

China faces the greatest challenge from stroke in the *** death rate for cerebrovascular diseases in China was 149.49 per 100000,accounting for 1.57 million deaths in *** ranked third among the leading causes of death behind malignant tumours and heart *** age-standardised prevalence and incidence of stroke in 2013 were 1114.8 per 100000 population and 246.8 per 100000 person-years,*** to the Global Burden of Disease Study 2017,the years of life lost(YLLs)per 100000 population for stroke increased by 14.6%;YLLs due to stroke rose from third highest among all causes in 1990 to the highest in *** absolute numbers and rates per 100000 population for all-age disability-adjusted life years(DALYs)for stroke increased substantially between 1990 and 2017,and stroke was the leading cause of all-age DALYs in *** main contributors to cerebrovascular diseases include behavioural risk factors(smoking and alcohol use)and pre-existing conditions(hypertension,diabetes mellitus,dyslipidaemia and atrial fibrillation(AF)).The most prevalent risk factors among stroke survivors were hypertension(63.0%-84.2%)and smoking(31.7%-47.6%).The least prevalent was AF(2.7%-7.4%).The prevalences for major risk factors for stroke are high and most have increased over *** on the latest national epidemiological data,26.6%of adults aged≥15 years(307.6 million adults)smoked tobacco *** those aged≥18 years,age-adjusted prevalence of hypertension was 25.2%;adjusted prevalence of hypercholesterolaemia was 5.8%;and the standardised prevalence of diabetes was 10.9%.For those aged≥40 years,the standardised prevalence of AF was 2.31%.Data from the Hospital Quality Monitoring System showed that 3010204 inpatients with stroke were admitted to 1853 tertiary care hospitals during *** those,2466785(81.9%)were ischaemic strokes(ISs);447609(14.9%)were intracerebral haemorrhages(ICHs);and 95810(3.2%)were subarachnoid haemorrhages(SAHs).The average age

External quality assessment of the detection of rickettsioses in China

Objective:To report a training course on the laboratory diagnoses of rickettsioses that 10 provincial/city CDCs participated in laboratory external quality assurance(EQA) based on the serological specific antibodies detection and rapid PCR amplifying targeted genes of rickettsiae. Methods:An EQA program to evaluate the following laboratory procedures was developed to detect rickettsiae:(1) immunofluorescent assay(IFA) to detect specific antibodies of A. phagocytophilum,*** and *** respectively.(2) Two sets of nested PCR were used amplifying groEL genes for most members of the family Rickettsiaceae and amplifying 16SrRNA genes for the most members of family anaplasmae,respectively.A scoring scheme based on the distribution of the median antibody titer values of the serologic assays was designed and a ranking list of the scores of the PCR results based on the detected minimal copy numbers of reference DNA was ***:Among nine laboratories who reported the results on time,eight laboratories gave acceptable serologic results,the other one provided an unacceptable antibody titer(1:2 vs median 1:64) results for *** limits of detection(LOD) for the PCR amplifying for five references DNA ranged from 1copy/μL to 10~6 copy/μ***:We successfully trained and popularized modern diagnostic methods of rickettsiae in 10 provincial CDCs in China and first conducted the EQA projects and evaluated the results.

Interpretation Criteria for Standardized Western Blot for the Predominant Species of Borrelia Burgdorferi Sensu Lato in China

Objective Western blotting （WB;immunoblotting） is a widely used tool for the serodiagnosis of Lyme borreliosis （LB）,but so far,no generally accepted criteria for its performance and interpretation have been established in *** present study was designed to determine the criteria for standardized Western blot for the predominant species of Borrelia burgdorferi sensu lato in China,in which WB was produced with strain PD91 as the representative strain attributed to predominant genospecies Borrelia garinii of Borrelia burgdorferi sensu *** Approximately 13 bands between 14 and 100 kD were differentiated for strain PD91 by using Gel-Pro analysis *** a study with 631 serum samples （taken from 127 patients with Lyme borreliosis and 504 controls）,all observed bands were *** establish criteria for a positive WB result for strain PD91,receiver operating characteristic （ROC） curves were *** The following interpretation criteria were recommended：for IgG,at least one band of P83/100,P58,P39,P30,OspC,P17,P66,and OspA;for IgM,at least one band of P83/100,P58,OspA,P30,OspC,P17 or *** addition,syphilis,leptospirosis and other related diseases should be excluded when the positive band is P41 in *** IgG criteria,the sensitivity is 73.2%,the specificity is 99.4% and Youden index is 0.726;for IgM criteria,the sensitivity is 50.6%,the specificity is 93.1% and Youden index is *** Standardization of WB assays is necessary for comparison of results from different ***,the criteria of other genospecies of Borrelia burgdorferi sensu lato should be determined in the future to complete the criteria of WB for the diagnosis of the Lyme disease in China.

In the past 30 years,the prevalence of diabetes in China has been increasing,but the awareness rate,treatment rate and control rate of diabetes are relatively low,and the death rate,disability rate,disease burden of diabetes and its complications are constantly increasing,which brings heavy burden to individuals,families and social *** Healthy China Initiative(2019-2030)is a roadmap to a healthy China and an innovative public policy system *** Diabetes prevention and control Action is one of the four chronic and noncommunicable diseases prevention and control actions in the Healthy China Initiative(2019-2030).Based on the background,basic principles and key contents of diabetes prevention and control,this paper interpreted the diabetes prevention and control action from five aspects,that is,healthy lifestyle is the foundation,which emphasized that individuals are the first responsibility for their health,and advocates reducing the risk of diabetes by practicing healthy diet,increasing physical activity and maintaining a healthy weight;Early detection and early intervention are key links,which emphasized to pay attention to individual blood glucose level,high risk individuals regularly test blood glucose;Standardize health management is the priority,which emphasized to strengthen the implementation of graded diagnosis and treatment standards,and medical institutions provide guidance on self-management of health for patients with diabetes;The improvement of capacity in primary level is the guarantee,where emphasis is placed on improving the ability of medical personnel to screen,intervention and standardize diagnosis and treatment;And innovative health service models are the key measures which emphasized the use of information technology to improve the efficiency of health management.
