We analyzed RNA-sequencing(RNA-seq)and clinical data from head and neck squamous cell carcinoma(HNSCC)patients in The Cancer Genome Atlas(TCGA)Genomic Data Commons(GDC)portal to investigate the prognostic value of anoikis-related genes(ARGs)in HNSCC and develop new targeted drugs.Differentially expressed ARGs were screened using bioinformatics methods;subsequently,a prognostic model including three ARGs(CDKN2A,BIRC5,and PLAU)was constructed.Our results showed that the model-based risk score was a good prognostic indicator,and the potential of the three ARGs in HNSCC prognosis was validated by the TISCH database,the model’s accuracy was validated in two independent cohorts of the Gene Expression Omnibus database.Immune correlation analysis and half-maximal inhibitory concentration were also performed to reveal the different landscapes of TIME between risk groups and to predict immuno-and chemo-therapeutic responses.Potential small-molecule drugs for HNSCC were subsequently predicted using the L1000FWD database.Finally,in vitro experiments were used to verify the database findings.The relative ARG mRNA expression levels in HNSCC and surrounding normal tissues remained consistent with the model results.BIRC5 knockdown inhibited anoikis resistance in WSU-HN6 and CAL-27 cells.molecular docking,real-time PCR,cell counting kit-8(CCK-8),plate clone,and flow cytometry analyses showed that small-molecule drugs predicted by the database may target the ARGs in the prognostic model,inhibit HNSCC cells survival rate,and promote anoikis in vitro.Therefore,we constructed a new ARG model for HNSCC patients that can predict prognosis and immune activity and identify a potential small-molecule drug for HNSCC,paving the way for clinically targeting anoikis in HNSCC.

摘要目的评估CT引导下间断(IRT)高剂量(HDR)近距离放疗(BRT)在无法切除的原发性和继发性肝恶性肿瘤的疗效,本文更新和扩展了我们先前描述该治疗方法的经验。方法 41例共50个肝门部或胆管分叉附近肿瘤,59次行CT引导下IRT HDR BRT治疗。肿瘤直径均>4 cm,体积中位数为84 cm3(38～1 348 cm3)。IRT HDR BRT释放的中等物理总剂量约20.0 Gy(7.0～32.0 Gy),19例病人每天2次,每次约7.0 Gy(4.0～10.0 Gy),22例每天1次约8.0 Gy(7.0～14.0 Gy)。

Hepatocellular carcinoma(HCC)is the fifth most common type of cancer and the fourth leading cause of cancer-related deaths in the world.HCC has a reported recurrence rate of 70%-80%after 5 years of follow-up.Controlling tumor recurrence is the most critical factor associated with HCC mortality.Conventional salvage therapies for recurrent HCC include re-hepatectomy or liver transplantation,transcatheter arterial chemoembolization,Y-90,target therapy,and immunotherapy;however,these conventional treatment modalities have yet to achieve consistently favorable outcomes.Meanwhile,previous studies have demonstrated that conventional therapies in combination with traditional Chinese medicine(TCM),acupuncture,moxibustion or dietary supplements could notably benefit patients with HCC recurrence by strengthening and augmenting the overall management strategy.However,systemic reviews related to the interactions between complementary therapies and conventional therapy in recurrent HCC are limited.In this review,we discuss the molecular mechanisms underlying the functions of complementary therapies for recurrent HCC,which include augmenting the local control to improve the congestion status of primary tumors and reducing multicentric tumor occurrence via inducing autophagy,apoptosis or cell cycle arrest.TCM and its derivatives may play important roles in helping to control HCC recurrence by inhibiting epithelial-mesenchymal transition,migration,invasion,and metastasis,inhibiting cancer stem cells,and ameliorating drug resistance.

Breast cancer is a frightful disease and serious concern in women around the world causing significant health care burden in both developed and developing countries. Extensive research work has shown that breast cancer provides strong resistance to chemical agents, U V radiation,and hormonal treatments. It is generally accepted that cell genetics is not the only main reason for breast cancer and genetic risk factors, for example, mutations in RRCAI and BRCA2 genes constitute 5%-10% of all breast cancer rates. Other related factors include age, gender,race, ethnicity, weight, reproductive factors, exo-and endogenous hormonal exposures, oral contraceptives use, ultraviolet radiation, diet, and night work(circadian disruption). Many studies have revealed that dietary isoflavones regulate breast cancer occurrence, recurrence and prognosis. Dietary isoflavones have long been part of Asian population diet and there is a significant increase as compared to dietary isoflavones intake among other populations. Dietary isoflavones are natural phytoestrogens having both estrogenic and anti-estrogenic potentials on breast cancer cells in culture, animal models and in experimental trials. This literature survey provides a comprehensive overview on the tumor preventive and tumor promoting potentials of dietary isoflavones on breast cancer. In addition, this paper provides a literature review of dietary isoflavones and their effects on up-regulation and down-regulation of different signaling pathways, genes and proteins. Finally, future perspectives of dietary isoflavones and breast cancer researchers are also critically discussed, which will provide a deeper insight regarding the inner molecular mechanisms of action.

The morbidity and mortality rates of cancer have gradually increased in China,making it an major obstacle to the development of the national economy,public health and society in general.Traditional Chinese medicine(TCM)has played an irreplaceable role in cancer therapy.TCM significantly reduces side effects and great ly improves the cancer patients’prognosis and quality of life following surgery,radio-chemotherapy or treatment with biological agents.With the advances in modern drug extraction technology and molecular biology techniques,Chinese herbal extracts from natural plants have gradually garnered increasing attention in the clinical setting because of their potent efficacy,relative safety,and convenience.Chinese herbal extracts have also become an important source of antitumor drugs and tumor adjuvant therapy.Studies have demonstrated that numerous Chinese herbal extracts not only have direct anti-tumor effects,but also improve the tumor immune microenvironment,enhance tumor immune surveillance,protect the intestinal mucosal barrier,regulate tumor metabolism and promote tumor cell apoptosis.In this review,we provide an overview of the mechanisms of action for Chinese herbal extracts,focusing on the relationship between Chinese herbal extracts and immune organs,inflammatory cytokines,cell defense,antioxidant effects and direct anti-tumor effects.

OBJECTIVE Platinum compounds such as cisplatin and carboplatin are frequently used as the first-line chemotherapy for the treatment of the head and neck squamous cell carcinoma(HNSCC).In the present study,we investigated whether garcinol,apolyisoprenylated benzophenone can chemosensitize HNSCC to cisplatin.METHODS The effect of garcinol and cisplatin on HNSCC was assessed by MTT,Western blotting,real time PCR,FACS,immunohistochemistry,DNA binding assay and xenograft mouse model.RESULTS We found that garcinol inhibited the viability of a panel of diverse HNSCC cell lines,enhanced the apoptotic effect of cisplatin,suppressed constitutive as well as cisplatin-induced NF-κB activation,and downregulated the expression of various oncogenic gene products(cyclin D1,Bcl-2,survivin and VEGF).In vivo study showed that administration of garcinol alone(0.5 mg·kg-1,ip five times/week)significantly suppressed the growth of the tumor,and this effect was further increased by cisplatin.Both the markers of proliferation index(Ki-67)and microvessel density(CD31)were downregulated in tumor tissues by the combination of cisplatin and garcinol.The pharmacokinetic results of garcinol indicated that good systemic exposure was achievable after ip administration of garcinol at 0.5and 2mg·kg-1 with mean peak concentration(cmax)of 1825.4 and 6635.7nmol·L-1 in the mouse serum,respectively.CONCLUSION Overall,our results suggest that garcinol can indeed potentiate the effects of cisplatin by negative regulation of various inflammatory and proliferative biomarkers.

Relationship between miR-7-5p expression and ^(125)I seed implantation efficacy in pancreatic cancer and functional analysis of target genes

Background:Forkhead box P3(Foxp3)is known as the optimum maker for regulatory T cells(Tregs),which are conven-tionally thought to induce immune tolerance to disturb the antitumor immunity.However,the research on the prognostic significance of tumor-infiltrating Foxp3+Tregs in breast cancer is still limited and the result is controversial.Methods:We searched for studies in Pub Med,EMBASE and Web of Science prior to January 2015.The correlation between Foxp3+tumor-infiltrating lymphocytes(TILs)and clinicopathological features and breast cancer prognosis was analyzed.The meta-analysis was performed using STATA 11.0.Pooled hazard ratios(HRs)with 95%confidence intervals(CIs)were used to estimate the strength of the association between Foxp3+TILs and prognosis of breast cancers,while relative ratios(RRs)were used to evaluate the relationship between Foxp3+TILs and clinicopathological features of breast cancers.Result:A total of 14 studies comprising 8475 breast cancer patients met the inclusion criteria.Our results showed that higher Foxp3+TILs level was significantly associated with poor prognosis in terms of overall survival(OS)(pooled HR:1.78,95%CI:1.04-3.06;P=0.036).We found that breast cancer with higher Foxp3+TILs level was positively correlated with c-erb B-2 positive status(pooled RR:1.51,95%CI:1.28-1.78;P=0.000),lymph node positive status(pooled RR:1.20,95%CI:1.07-1.35;P=0.002)while there is negative association with ER positive status(pooled RR:0.67,95%CI:0.57-0.79;P=0.000)and PR positive status(pooled RR:0.68,95%CI:0.50-0.93;P=0.016).Conclusion:The present meta-analysis showed that higher Foxp3+TILs level in patients with breast cancer indicated poor overall survival(OS)and was significantly associated with c-erb B-2 status,lymph node status,ER status and PR status.Foxp3+TILs level is an promising prognostic factor in breast cancers.

Objective The aim of this study was to investigate the relationship between miR-7-5p expression and intertissue-^(125)I irradiation sensitivity in pancreatic cancer tissues and to analyze the function of target genes.Methods Thirty-seven patients with unresectable pancreatic ductal adenocarcinoma(PDAC)treated with radioactive ^(125)I seed implantation were enrolled.RT-PCR was used to detect the expression level of miR-7-5p in cancer tissues and analyze the relationship between miR-7-5p expression and ^(125)I radiation sensitivity.Bioinformatic software and online tools were used to predict the miR-7-5p target genes and analyze their functional annotation and pathway enrichment.Results Radioactive ^(125)I seed implantation was followed up for 2 months.The objective response rate of the miR-7-5p high expression group was 65.0%(13/20),whereas the objective response rate of the miR-7-5p low expression group was 5.88%(1/17),and the difference between the two groups was statistically significant(χ^(2)=13.654,Pmolecular complex detection analysis by Cytoscape revealed that PIGH,RAF1,EGFR,NXT2,PIK3CD,PIK3R3,ERBB4,TRMT13,and C5orf22 were the key modules of miR-7-5p target gene clustering.Conclusion The expression of miR-7-5p in pancreatic cancer tissues positively correlated with the radiosensitivity of ^(125)I seeds.Via targeted gene regulation,miR-7-5p
