Objective:To evaluate if berberine can act on vitamin D receptors(VDR)and thereby regulate the expression of tight junction proteins(TJPs)in irritable bowel syndrome-diarrhea-predominant(IBS-D)***:The newborn rats were induced into IBS-D rat model via neonatal maternal separation combined with acetic acid chemical *** modeling,the model was evaluated and rats were divided into the control group and berberine treatment groups(0.85,1.7 and 3.4 mg/kg,once a day for 2 weeks).The distal colon was obtained and colonic epithelial cells(CECs)were isolated and cultured after IBS-D model *** vitamin D receptor response element(VDRE)reporter gene was determined in the CECs of IBS-D rats to analyze the effect of berberine on the VDRE *** overexpression or silencing technology was used to analyze whether VDR plays a role in promoting intestinal barrier repair,and to determine which region of VDR plays a role in berberine-regulated intestinal ***:The IBS-D rat model was successfully constructed and the symptoms were improved by berberine in a dose-dependent manner(P<0.05).The activity of VDRE promoter was also effectively promoted by berberine(P<0.05).Berberine increased the expression of TJPs in IBS-D CECs(P<0.05).VDR expression was significantly increased after transfection of different domains of VDR when compared to normal control and basic plasmid groups(all P<0.05).RT-qPCR and Western blot results showed that compared with the blank group,expressions of occludin and zonula occludens-1 were significantly higher in VDR containing groups(all P<0.05).Berberine plus pCMV-Myc-VDR-N group exerted the highest expression levels of occludin and zonula occludens-1(P<0.05).Conclusion:Berberine enhances intestinal mucosal barrier function of IBS-D rats by promoting VDR activity,and the main site of action is the N-terminal region of VDR.

The traditional simple random sampling(SRS)design method is inefficient in many *** proposed some new designs to increase *** this paper,as a variation of moving extremes ranked set sampling(MERSS),double MERSS(DMERSS)is proposed and its properties for estimating the population mean are *** turns out that,when the underlying distribution is symmetric,DMERSS gives unbiased estimators of the population ***,it is found that DMERSS is more efficient than the SRS and MERSS methods for usual symmetric distributions(normal and uniform).For asymmetric distributions considered in this study,the DMERSS has a small bias and it is more efficient than SRS for usual asymmetric distribution(exponential)for small sample sizes.

Objective: To establish the dynamic treatment strategy of Chinese medicine(CM) for metastatic colorectal cancer(m CRC) by machine learning algorithm, in order to provide a reference for the selection of CM treatment strategies for m CRC. Methods: From the outpatient cases of m CRC in the department of Oncology at Xiyuan Hospital, China Academy of Chinese Medical Sciences, 197 cases that met the inclusion criteria were screened. According to different CM intervention strategies, the patients were divided into 3 groups: CM treatment alone, equal emphasis on Chinese and Western medicine treatment(CM combined with local treatment of tumors, oral chemotherapy, or targeted drugs), and CM assisted Western medicine treatment(CM combined with intravenous regimen of Western medicine). The survival time of patients undergoing CM intervention was taken as the final evaluation index. Factors affecting the choice of CM intervention scheme were screened as decision variables. The dynamic CM intervention and treatment strategy for m CRC was explored based on the cost-sensitive classification learning algorithm for survival(CSCLSurv). Patients' survival was estimated using the Kaplan-Meier method, and the survival time of patients who received the model-recommended treatment plan were compared with those who received actual treatment plan. Results: Using the survival time of patients undergoing CM intervention as the evaluation index, a dynamic CM intervention therapy strategy for m CRC was established based on *** CM intervention strategies for m CRC can be selected according to dynamic decision variables, such as gender, age, Eastern Cooperative Oncology Group score, tumor site, metastatic site, genotyping, and the stage of Western medicine treatment at the patient's first visit. The median survival time of patients who received the modelrecommended treatment plan was 35 months, while those who receive the actual treatment plan was 26.0 months(P=0.06). Conclusions:

Recent advances in high-throughput chromosome conformation capture(Hi-C)techniques have allowed us to map genome-wide chromatin interactions and uncover higher-order chromatin structures,thereby shedding light on the principles of genome architecture and ***,statistical methods for detecting changes in large-scale chromatin organization such as topologically associating domains(TADs)are still ***,we proposed a new statistical method,DiffGR,for detecting differentially interacting genomic regions at the TAD level between Hi-C contact *** utilized the stratum-adjusted correlation coefficient to measure similarity of local TAD *** then developed a nonparametric approach to identify statistically significant changes of genomic interacting *** simulation studies,we demonstrated that DiffGR can robustly and effectively discover differential genomic regions under various ***,we successfully revealed cell type-specific changes in genomic interacting regions in both human and mouse Hi-C datasets,and illustrated that DiffGR yielded consistent and advantageous results compared with state-of-the-art differential TAD detection *** DiffGR R package is published under the GNU General Public License(GPL)≥2 license and is publicly available at https://***/wmalab/DiffGR.

Cardiac arrest poses a significant global public health challenge,manifesting in approximately 550,000 cases annually within the United States.[1]In-hospital cardiac arrest(IHCA)is commonly attributed to airways and respiratory issues.[2]Recommendations emphasize the expertise of responders in airway management.[3]Various options exist,such as chest compression-only cardiopulmonary resuscitation(CPR),bag-mask ventilation(BMV),and advanced *** BMV and advanced airways are not deemed equivalent or superior based on previous evidence.[4]Achieving consistency in choosing and timing the optimal airway approach during IHCA is *** current American Heart Association guidelines suggest an advanced airway strategy when endotracheal intubation(ETI)success rates are high,but the optimal time for advanced airway management remains unclear.[5]Wong et al[6]revealed that survival improved by less than 5 min with advanced airway *** to a subgroup analysis of IHCA patients in emergency departments(EDs),early intubation was associated with a 1.5-fold greater rate of return of spontaneous circulation(ROSC)than in other locations.[7]ED patients’constant monitoring and immediate management,with readily available intubation equipment,enhance early intubation and survival rates.[6]Nonetheless,IHCA patients intubated within the first 15 min had a lower ROSC rate.
