Exosome-encapsulated lncRNA HOTAIRM1 contributes to PM_(2.5)-aggravated COPD airway remodeling by enhancing myofibroblast differentiation

Emphysema,myofibroblast accumulation and airway remodeling can occur in the lungs due to exposure to atmospheric pollution,especially fine particulate matter(PM_(2.5)),leading to chronic obstructive pulmonary disease(COPD).Specifically,bronchial epithelium-fibroblast communication participates in airway remodeling,which results in *** increasing number of studies are now being conducted on the role of exosome-mediated cell-cell communication in disease ***,we investigated whether exosomes generated from bronchial epithelial cells could deliver information to normal stromal fibroblasts and provoke cellular responses,resulting in airway obstruction in *** studied the mechanism of exosome-mediated intercellular communication between human bronchial epithelial(HBE) cells and primary lung fibroblasts(pLFs).We found that PM_(2.5)-induced HBE-derived exosomes promoted myofibroblast differentiation in ***,the exosomal lncRNA expression profiles derived from PM_(2.5)-treated HBE cells and nontreated HBE cells were investigated using an Agilent Human LncRNA *** coculture assays and direct exosome treatment,we found that HBE cell-derived exosomal HOTAIRM1 facilitated the myofibroblast differentiation of ***,we discovered that exosomal HOTAIRM1 enhanced p LF proliferation to secrete excessive collagen secretion,leading to airway obstruction by stimulating the TGF-β/SMAD3 signaling ***,PM_(2.5)reduced FEV1/FVC and FEV1 and increased the level of serum exosomal HOTAIRM1 in healthy people;moreover,serum exosomal HOTAIRM1 was associated with PM_(2.5)-related reductions in FEV1/FVC and *** findings show that PM_(2.5)triggers alterations in exosome components and clarify that one of the paracrine mediators of myofibroblast differentiation is bronchial epithelial cell-derived HOTAIRM1,which has the potential to be an effective prevention and therapeutic target for PM_(2.5)-induced COPD.

Soleus arthrogenic muscle inhibition following acute lateral ankle sprain correlates with symptoms and ankle disability but not with postural control

Background:Acute lateral ankle sprains(ALAS)are associated with long-term impairments and instability tied to altered neural *** muscle inhibition(AMI)has been observed in this population;however,relationships with injury-related impairments are unclear,potentially due to the resting,prone position in which AMI is typically *** AMI during bipedal stance may provide a better understanding of this ***:AMI was assessed in 38 young adults(19 ALAS within 72 h of injury:10 males,21.4±2.7 years;19 healthy controls:10 males,21.9±2.2 years;mean±SD)using the Hoffmann reflex(H-reflex)during bipedal *** stimulation was administered to identify the _(max)imal H-reflex(H_(max))and _(max)imal motor response(M_(max))from the soleus,fibularis longus,and tibialis anterior *** primary outcome measure was the H_(max)/M_(max) *** outcomes included acute symptoms(pain and swelling),postural control during bipedal stance,and self-reported ***:No significant group-by-limb interactions were observed for any ***,a significant group main effect was observed in the soleus muscle(F(1,35)=6.82,p=0.013),indicating significantly lower H_(max)/M_(max) ratios following ALAS(0.38±0.20)compared to healthy controls(0.53±0.16).Furthermore,lower H_(max)/M_(max) ratios in the soleus significantly correlated with acute symptoms and self-reported function but not with postural ***:This study supports previous evidence of AMI in patients with ALAS,providing insight into neurophysiologic impacts of musculoskeletal *** results suggest that assessing AMI in a standing position following acute injury may provide valuable insight into how AMI develops and guide potential therapeutic options to curb and offset the formation of joint instability.

Understanding the educational inequalities in suicide attempts and their mediators:a Mendelian randomisation study

Background Educational inequalities in suicide have become increasingly prominent over the past *** modifiable risk factors that serve as intermediaries in the impact of low educational attainment on suicide has the potential to reduce health *** To examine the risk factors that mediate the relationship between educational attainment and suicide attempts and quantify their contributions to the mediation *** We conducted a two-sample Mendelian randomisation(MR)analysis to estimate the causal effect of educational attainment on suicide attempts,utilising genome-wide association study summary statistics from the Integrative Psychiatric Research(iPSYCH;6024 cases and 44240 controls)and FinnGen(8978 cases and 368299 controls).We systematically evaluated 42 putative mediators within the causal pathway connecting reduced educational attainment to suicide attempts and employed two-step and multivariable MR to quantify the proportion of the mediated *** In the combined analysis of iPSYCH and FinnGen,each standard deviation(SD)decrease in genetically predicted educational attainment(equating to 3.4 years of education)was associated with a 105%higher risk of suicide attempts(odds ratio(OR):2.05;95%confidence interval(Cl):1.81 to 2.31).0f the 42 risk factors analysed,the two-step MR identified five factors that mediated the association between educational attainment and suicide *** respective proportions of mediation were 47%(95%Cl:29%to 66%)for smoking behaviour,36%(95%Cl:0%to 84%)for chronic pain,49%(95%Cl:36%to 61%)for depression,35%(95%Cl:12%to 59%)for anxiety and 26%(95%Cl:18%to 34%)for *** MR implicated these five mediators collectively,accounting for 68%(95%Cl:40%to 96%)of the total *** This study identified smoking,chronic pain and mental disorders as primary intervention targets for attenuating suicide risk attributable to lower educational levels in the European population.

Modelling and optimization of nonylphenol biosorption by novel lowcost magnetic Chlorella vulgaris

Discharging untreated wastewater causes environmental *** research examined the efficiency of removal and the adsorption process of Emerging Contaminant nonylphenol(NP)using magnetic Chlorella vulgaris(M-Chlorella vulgaris).The characterization of M-Chlorella vulgaris involved XRD,FESEM,EDS-Mapping,BET,FTIR,and VSM *** impact of four different parametersdpH,MChlorella vulgaris dose,initial concentration of NP,and contact time on the biosorption process was *** model and optimize the study while minimizing costs and the number of experiments,Response Surface Methodology(RSM)with Central Composite Design(CCD)was *** to the findings,the quadratic model with adjusted R^(2)=0.96 was the best fitted among the other *** pH=3.5,dosage=2.5 g/l,NP concentration=4 mg/l,and reaction time=70 min,which are the optimal parameters,the maximum removal effectiveness was about 91%.The Langmuir model(R^(2)=0.998)and pseudo-second-order model(R^(2)=0.997)exhibited the best fits for the adsorption isotherm and kinetic studies,*** characteristics of M-Chlorella vulgaris were determined to facilitate a spontaneous nature,and thermodynamic experiments indicated that the process of adsorption of nonylphenol is *** inexpensiveness and availability of adsorbent,suitable efficiency in the biosorption of pollutant and magnetic collection of pollutant from the aquatic environment can be mentioned as the advantages of this process.

Ambient ozone(O3)exposure may be associated with a reduction of semen quality,yet the potential biological mechanisms remain *** investigated the effects of certain seminal plasma metabolites on mediating the links between O3 exposure and the deterioration of semen *** untargeted metabolomics analysis was performed on semen samples of 200 Chinese adult men to determine candidate metabolites associated with characteristics of semen *** analysis was adopted to examine whether these metabolites modulated the links between O3 exposure and semen *** found a significant reduction in sperm concentration by−28.1%(95%CI:−41.7%,−11.3%),and sperm count by−29.2%(95%CI:−43.7%,−11.0%)associated with each 10μg/m3 increase in ambient O3 concentration during the period of sperm *** delineated 7 metabolites in seminal fluid that substantially mediated the links between O3 exposure and declined semen quality,including myristoleic acid,aspartyl-isoleucine,phenylethyl primeveroside,ACar(18:2),ACar(18:1),FAHFA(22:6/22:3),and LPS(22:5).Among these,myristoleic acid exhibited the most pronounced mediation effects,with its indirect effect of which accounts for 46.4%of the overall *** findings suggested that exposure to ozone decreased sperm quality by disrupting fatty acid metabolism,particularly myristoleic acid.

Gut barrier disruption is a key event in bridging gut microbiota dysbiosis and high-fat diet(HFD)-associated metabolic ***,the underlying mechanism remains *** the present study,by comparing HFD-and normal diet(ND)-treated mice,we found that the HFD instantly altered the composition of the gut microbiota and subsequently damaged the integrity of the gut *** sequencing revealed that the HFD upregulates gut microbial functions related to redox reactions,as confirmed by the increased reactive oxygen species(ROS)levels in fecal microbiota incubation in vitro and in the lumen,which were detected using in vivo fluorescence *** microbial ROS-producing capability induced by HFD can be transferred through fecal microbiota transplantation(FMT)into germ-free(GF)mice,downregulating the gut barrier tight ***,mono-colonizing GF mice with an Enterococcus strain excelled in ROS production,damaged the gut barrier,induced mitochondrial malfunction and apoptosis of the intestinal epithelial cells,and exacerbated fatty liver,compared with other low-ROS-producing Enterococcus *** administration of recombinant high-stability-superoxide dismutase(SOD)significantly reduced intestinal ROS,protected the gut barrier,and improved fatty liver against the *** conclusion,our study suggests that extracellular ROS derived from gut microbiota play a pivotal role in HFD-induced gut barrier disruption and is a potential therapeutic target for HFD-associated metabolic diseases.
