Screening Research of Pharmaceutical Compositions Based on Succinic Acid, Ascorbic Acid and Rutin

We conduct research on the development of new medicines based on succinic acid, ascorbic acid and rutin. We studied the anti-inflammatory action （exudative inflammation, model of carrageenan-induced paw edema of rats）, the hepatoprotective （injury of rats with carbon tetrachloride） and renal protectie action. In the study, the anti-inflammatory activity of our medicines （exudative inflammation, karahenin model of inflammatory edema of paws rats） is set to ability of suppression of the inflammatory response by 24.4 %. Against the background, the action of carbon tetrachloride observed a positive effect on cholesterol- and pigment- forming liver function. Reducing the activity of enzymes ALT （alanine transaminase） and AST （aspartate transaminase） in groups of animals, which treated with our medication, talks about their ability to recover morpho functional integrity of the membranes of hepatocytes and warn of changes in the liver damage with carbon tetrachloride. Renal protective action screening study found that investigated mixture capable of increasingly lower level of creatinine in the blood of rats： 27.27-39.18 mmol/L in Lespeflan. Similarly, the concentration of urea in the smaller of the studied using mixtures is 5.18 mmol/L, while the application lespeflanu urea concentration slightly higher is 6.78 mmol/L. This shows that hypoazotemic effect is traced compared with the control group, and better than the comparator Lespeflan. Study of acute toxicity showed that the investigated mixture refers to practically nontoxic drugs. It is confirmed the need for further studies on the pharmacological activity of our facility to determine the effect on capillaries and the immune system, and as a result, prevention and treatment of influenza and ARI （acute respiratory infections）.

Quantification of aflatoxins and health risk assessment through intake of herbal medicine formulations

Aflatoxins(AFs)are frequent contaminants of herbs,which pose a health risk to regular *** study quantified AFs(AFB1,AFB2,AFG1,AFG2,and Total AFs)in 100 of frequently used herbal medicine formulations in Northern Jordan,and assessed the health risk associated with its *** half of the samples had levels lower than the detection limit,while the concentrations of AFB1 and Total AFs were higher than the European limits in 10%and 23%of the collected samples,*** mean average daily dose(ADD)of AFB1 through the consumption of the formulations was 3.32×10^(-6)and ranged between 1.21×10^(-9)and 0.00045 ng/kg bwt/day,while the mean ADD of Total AFs was 6.80×10^(-5)and ranged between 3.21×10^(-9)and 0.0015 ng/kg bwt/*** margin of exposure(MOE)for each medicine herein was far greater than 10,000 as recommended by European Food Safety Authority(EFSA)for carcinogenic and genotoxic *** cancer risk due to life time exposure to AFB1 in herbal medicines was 1.03×10^(-5)(ranged between 4.51×10^(-4)to 1.19×10^(-9)cancers/10^(5) individuals/75 year).The frequent occurrence of AFs in the analyzed samples raised questions on their potential harmful consequences on heavy consumers,and suggested that further official measures could minimize contamination and improve management of the associated cancer risk.

Salvia officinalis L.(Lamiaceae)leaves and its essential oil is used for mouth and throat disorders,skin disorders,minor wounds,and gastrointestinal disorders,and is widely used *** research aimed to conduct a comparative study of the composition of *** essential oils from commercial samples,and their main *** volatile constituents from *** leaves were investigated using gas chromatography(GC).The commercial samples of sage leaves were obtained from retail pharmacies in nine mainly European *** yield of essential oil in *** commercial leaves was between 10.0 and 24.8 mL/*** principal components(>5%)among the main identified 25 compounds were 1,8-cineole(8.3%–45.3%),α-thujone(3.0%–34.0%),сamphor(11.3%–29.3%),β-thujone(1.5%–12.9%),viridiflorol(1.1%–10.4%),camphene(2.6%–7.1%),andα-pinene(1.3%–5.8%).In seven(Estonia,England,France,Hungary,Belgium,Ukraine,Georgia)samplesα-thujone *** samples(Estonia,Georgia,England,Hungary)belong to the most common chemotypeα-thujone>camphor>1,*** chemotypes of *** essential oils have been *** thujones are widespread compounds among them.

Dear Editor,Pancreatic cancer*is one of the deadliest cancers,with an overall 5-year survival rate of less than 10%unchanged over the last 40 *** 80-85%of patients are diagnosed as advanced stage,which are not eligible for curative surgery.1 Therapeutic options for advanced pancreatic cancer patients are still chemotherapy drugs with limited outcomes to improve patient survival and life quality.

Bioavailability enhancers of herbal origin:An overview

Recently,the use of herbal medicines has been increased all over the world due to their therapeutic effects and fewer adverse effects as compared to the modern ***,many herbal drugs and herbal extracts despite of their impressive in-vitro findings demonstrates less or negligible in-vivo activity due to their poor lipid solubility or improper molecular size,resulting in poor absorption and hence poor *** with the advancement in the technology,novel drug delivery systems open the door towards the development of enhancing bioavailability of herbal drug delivery *** last one decade many novel carriers such as liposomes,microspheres,nanoparticles,transferosomes,ethosomes,lipid based systems *** been reported for successful modified delivery of various herbal *** herbal compounds including quercetin,genistein,naringin,sinomenine,pipeline,glycvrrhizin and nitrile glycoside have demonstrated capability to enhance the *** objective of this review is to summarize various available novel drug delivery technologies which have been developed for delivery of drugs(herbal),and to achieve better therapeutic *** attempt has also been made to compile a profile on bioavailability enhancers of herbal origin with the mechanism of action(wherever reported)and studies on improvement in drug bioavailability,exhibited particularly by natural compounds.

Adherence of Community Pharmacies in Riyadh, Saudi Arabia, to Optimal Conditions for Keeping and Selling Good-Quality medicines

Exposure to excessive temperature during distribution or storage has unfavorable consequences on the quality of medicines, particularly in hot climates. This could be one of the possible reasons behind the existence of substandard amoxicillin in community pharmacies in Riyadh, Saudi Arabia. This study explored the conditions under which medicines were kept in a random sample of 181 community pharmacies in Riyadh. The pharmacist in charge in each pharmacy was interviewed and our observations about the quality of storage were recorded. The inspection revealed that in 9% of the sample the readings of the existing room thermometers were >25℃, and that 13% of the sample lacked thermometers. Also in 33% of the sample the readings of the refrigerator thermometers were outside the accepted range, and 7% of the sample lacked refrigerator thermometers. About 15% of pharmacists were not informed about the local regulations of community pharmacy practice, neither before nor after taking up their current positions. Surprisingly, incorrect answers to simple questions about the system were frequently given by the informed pharmacists. Certain aspects of substandard storage conditions existed, in varying degrees, in significant percentages of pharmacies regardless of the pharmacists’ qualifications, experience, or awareness about the local regulations of community pharmacy practice. Stricter monitoring by the authorities regarding the adherence of community pharmacies to optimal conditions of storing medicines is recommended. Continuing education of community pharmacists must also be improved.

目的:印度贾尔贡地区森林里的榕属植物多年来被当地人用来治疗腹泻。本研究根据当地人的用药传统选取3类榕属植物的特定部位来研究它们对不同大鼠腹泻模型的治疗及缓解作用。方法:不限性别地选取体质量在180~200 g的Wistar大鼠240只,在每个独立实验中,大鼠分别被分为8组,每组10只。通过蓖麻油诱导大鼠腹泻、大鼠胃肠蠕动测试及前列腺素E2诱导大鼠小肠淤积的3个独立实验来评估孟加拉榕树树皮、聚果榕树叶以及无花果叶的乙醇提取物治疗腹泻的作用。结果:孟加拉榕树树皮、聚果榕树叶和无花果叶的提取物具有显著抑制由蓖麻油诱发的大鼠腹泻及前列腺素诱导的大鼠小肠淤积的作用。当给予大鼠400、600 mg/kg的乙醇提取物时,腹泻显著减少。与标准对照药组对比,服用3种植物乙醇提取物的大鼠表现出显著的剂量依赖性腹泻缓解现象(P<0.01)。与蓖麻油诱导大鼠腹泻实验中的对照组相比,榕属植物的乙醇提取物被证实具有显著的降低粪便排泄量及排泄次数的作用(P<0.01)。所有的植物提取物具有显著减缓木炭餐在大鼠小肠内的推进速度并抑制由前列腺素E2引起的大鼠小肠淤积的作用。结论:印度贾尔贡地区的榕属植物具有止泻作用。这些植物的作用机制主要通过解痉和抗小肠淤积来缓解腹泻,这可能与榕属植物的乙醇提取物内含有鞣酸及类黄酮有关。

The Quality of medicines in Community Pharmacies in Riyadh, Saudi Arabia: A Lot Quality Assurance Sampling (LQAS)-Based Survey

Objectives: To classify community pharmacies (CPs) in Riyadh, Saudi Arabia, in terms of the quality of medicines sold by them, using?the lot quality assurance sampling (LQAS) technique with a predefined threshold. Methods: Riyadh CPs were divided into 2 categories (“lots” for the purpose of LQAS), i.e., chain and independent CPs. Upper and lower rate thresholds for CPs that sell low-quality medicines were predefined as 20% and 5%, respectively. Consumer and provider risks were predefined as 0.05 and 0.10, respectively. The calculated number of randomly selected CPs required in each lot was 36;then, sale of low-quality medicines in >3 CPs implies a prevalence of >20% of such CPs according to LQAS. A randomly selected brand of amoxicillin (selected as a quality indicator of medicines because it is both widely counterfeited and heat-sensitive) was purchased from each pharmacy by a “mystery shopper”, checked for authenticity, and analyzed for drug content and content uniformity using a validated HPLC method. Results: Substandard amoxicillin was purchased in 9 pharmacies (4 chains and 5 independent). Both lots were thus rejected as unacceptable, which may indicate that consumers in Riyadh are at risk of purchasing substandard medicines at CPs. Conclusions: The quality of medicines sold in CPs in Riyadh did not meet our acceptability criterion, and appropriate intervention by decision makers is recommended. LQAS proved to be a practical, economical, and statistically valid sampling method for surveying the quality of medicines. It should enable decision makers to allocate resources for improvement more efficiently.

Gastroprotective effect of the iridoid fraction from Barleria prionitis leaves on experimentally-induced gastric ulceration

AIM: To study the gastroprotective effect and in vivo antioxidant potential of a standardized iridoid fraction from B. prionitis leaves(BPE) against different gastric ulcer models in rats. METHOD: The standardized iridoid fraction from BPE at 50, 100, and 200 mg/kg body weight was administered orally, twice daily for 5 days for prevention from aspirin, ethanol, cold-restraint stress(CRS), and pylorus ligation(PL)-induced ulcers. Estimation of the antioxidant enzyme activity was carried out in a CRS-induced ulcer model, and various gastric secretion parameters including volume of gastric juice, acid output, and pH value were estimated in the PL-induced ulcer model. RESULTS: BPE showed a dose-dependent ulcer protective effect in PL(18.67%–66.26% protection), aspirin(24.65%–63.25% protection), CRS(20.77%–59.42% protection), and EtOH(16.93%–77.04% protection)-induced ulcers. BPE treatment in PL-rats showed a decrease in acid-pepsin secretion, and enhanced mucin and mucosal glycoproteins. However, BPE reduced the ulcer index with significant decrease in LPO(P < 0.01–0.001), SOD(P < 0.01–0.001), and an increase in CAT(P < 0.01–0.001), activity in the CRS-induced model. CONCLUSION: The data shows that the iridoid fraction from BPE possesses anti-ulcerogenic and antioxidant potential.
