STING signaling in islet macrophages impairs insulin secretion in obesity

The innate immune regulator stimulator of interferon genes(STING)mediates self-DNA sensing and leads to the induction of type I interferons and inflammatory cytokines,which promotes the progression of various inflammatory and autoimmune *** immune system plays a critical role in regulating obesity-induced islet dysfunction,whereas the potential effect of STING signaling is not fully ***,we demonstrate that STING is mainly expressed and activated in islet macrophages upon high-fat diet(HFD)***^(-/-)alleviates HFD-induced islet inflammation by inhibiting the expression of pro-inflammatory cytokines and the infiltration of ***,palmitic acid incubation promotes mitochondrial DNA leakage into the cytosol and subsequently activates STING pathway in ***,STING activation in macrophages impairs glucose-stimulated insulin secretion by mediating the engulfment ofβcell insulin secretory *** inhibiting STING activation enhances insulin secretion to control ***,our results reveal a regulatory mechanism in controlling the islet inflammation and insulin secretion in dietinduced obesity and suggest that selective blocking of the STING activation may be a promising strategy for treating type 2 diabetes.

Dear Editor,Protein histidine methylation has been found to be a widespread post-translational modification(PTM)in mammalian cells and represents more than 13%of all protein methylation events in the human methylome(Ning et al.,2016;Kapell and Jakobsson,2021).Recently,we and other groups independently identified METTL9 as the first broad-specificity histidine N1 methyltransferase in mammals(Davydova et al.,2021;Lv et al.,2021).METTL9 methylates a variety of substrates with a common x-His-x-His motif(His is for histidine;the second x is preferably small amino acids:A/C/G/S)and catalyzes methyl transfer to the last histidine.

HMGA2,a pivotal transcription factor,functions as a versatile regulator implicated in the progression of diverse aggressive *** this study,mass spectrometry was employed to identify ubiquitin-specific proteases that potentially interact with HMGA2,and USP48 was identified as a deubiquitinating enzyme of *** enforced expression of USP48 significantly increased HMGA2 protein levels by inhibiting its degradation,while the deprivation of USP48 promoted HMGA2 degradation,thereby suppressing tumor invasion and *** discovered that USP48 undergoes SUMOylation at lysine 258,which enhances its binding affinity to *** subsequent phenotypic screening of small molecules,we identified DUB-IN-2 as a remarkably potent pharmacological inhibitor of ***,the small-molecule inhibitor targeting USP48 induces destabilization of ***,upregulation of USP48 or HMGA2 in cancerous tissues is indicative of poor prognosis for patients with colorectal cancer(CRC).Collectively,our study not only elucidates the regulatory mechanism of DUBs involved in HMGA2 stability and validates USP48 as a potential therapeutic target for CRC,but also identifies DUB-IN-2 as a potent inhibitor of USP48 and a promising candidate for CRC treatment.

Metabolism is mobilized to meet the biosynthetic,bioenergetic,and regulatory demands of immune cell *** nothing is known about the metabolic basis of antibody class switch in B cells.A recent study published in Nature Communications through a joint effort by Chen Lab(metabolism biology)and Liu Lab(B cell biology)from Tsinghua University reveals that the monocarboxylate carrier MCT1(monocarboxylate transporter 1)promotes pyruvate oxidation and histone acetylation to support class switch recombination(CSR),the process by which B cells make antibodies of different isotypes with unique effector functions.

Thin-walled structures are widely used in cars due to their lightweight construction and energy-absorbing ***,issues such as high initial stress and lowenergy-absorbing efficiency *** study proposes a novel energy-absorbing structure inwhich a straight tube is combinedwith a conical tube and a bamboo-inspired bulkhead structure is *** configuration allows the conical tube to flip outward first and then fold together with the straight *** deformation mode absorbs more energy and less peak force than the conical tube sinking and flipping *** finite element numerical simulation,the specific energy absorption capacity of the structure is increased by 26%compared to that of a regular circular cross-section ***,the impact resistance of the bionic straight tapered tube structure is further improved through multi-objective optimization,promoting the engineering application and lightweight design of hybrid cross-section tubes.

Artificial light at night(ALAN) pollution alters bat lunar chronobiology:insights from broad-scale long-term acoustic monitoring

Background The timing of behavior and habitat use of nocturnal animals can be influenced by the lunar cycle in nature. The prevalence of artificial light at night(ALAN) has been recognized as a source of environmental pol-lution. The interaction between ALAN and the lunar cycle on bat behavior is important for understanding anthro-pogenic effects on bats. We utilized a decade(2012–2022) of acoustic monitoring data collected in North Carolina, United States, to investigate the relationship between bat activity, lunar cycle, and light pollution. We examined whether the amount of lunar illumination affected species-specific nightly activity and whether hourly bat activity patterns varied between nights with different moon phases. We further investigated if the relationship between bat activity and the lunar cycle might be altered by light *** We found that seven bat species showed activity variation across nights in relation to the amount of moon illumination when ALAN was absent. In general, bats were less active on full moon nights compared to new moon nights. Light pollution interacted with the bat–lunar relationship in five of the seven species, masking the effect of the lunar cycle. We identified delayed bat activity patterns on nights with a full or waxing moon in seven species, and light pollution altered that pattern in four species. Overall, ALAN was associated with decreased bat activity inde-pendent of lunar cycle *** Our study demonstrated that at a broad spatial scale, ALAN negatively affected many North American temperate bat species and altered their lunar chronobiology. As light pollution is spreading to historically dark areas and habitats, ALAN might couple with other threats, such as the white-nose syndrome or climate change, to cause cascading damage in the environment that depends on ecosystem services such as pest control provided by bats. Single-chain nanoparticles represent an emerging class of nanomaterials designed to mimic protein's folding *** covalent crosslinking toward the formation of single-chain nanoparticles encounters complex energy landscapes,leading to the potential occurrence of misfolding *** noncovalent crosslinking can circumvent this issue,the resulting single-chain nanoparticles exhibit lower structural stability compared to their covalently crosslinked *** this study,we present a novel approach for the synthesis of single-chain nanoparticles,achieved through the combination of non-covalent and covalent intramolecular *** grafted onto the linear polymer form intrachain non-covalent stacks aided by hydrogen bonds,leading to the formation of non-covalently crosslinked single-chain *** nanoparticles undergo conversion to covalently crosslinked nanostructures through subsequent photo-irradiation using[2+2]photocycloaddition,a process facilitated by the supramolecular confinement ***,the resulting single-chain nanoparticles demonstrate both intrachain folding efficiency and substantial stability,offering significant potential for advancing applications across diverse fields.
