Although the dynamic instability of microtubules(MTs)is fundamental to many cellular functions,quiescent MTs with unattached free distal ends are commonly present and play important roles in various events to power cellular ***,how these free MT tips are stabilized remains poorly ***,we report that centrosome and spindle pole protein 1(CSPP1)caps and stabilizes both plus and minus ends of static ***-time imaging of laser-ablated MTs in live cells showed deposition of CSPP1 at the newly generated MT ends,whose dynamic instability was concomitantly ***,MT ends in CSPP1-overexpressing cells were hyper-stabilized,while those in CSPP1-depleted cells were much more *** CSPP1-elicited stabilization of MTs was demonstrated to be achieved by suppressing intrinsic MT catastrophe and restricting ***,CSPP1-bound MTs were resistant to mitotic centromere-associated kinesin-mediated *** findings delineate a previously uncharacterized CSPP1 activity that integrates MT end capping to orchestrate quiescent MTs.

Maize(Zea mays)endosperm is an important food source for humans and *** addition,it is considered an excellent model for research on seed development because of its relatively large size for both embryo and endosperm compared to those of other crop species,such as rice(Oryza sativa)and wheat(Triticum aestivum).After double fertilization,early maize endosperm development includes three stages:(i)a multinucleate coenocyte stage;(i)a cellularization stage,in which the cellular endosperm is formed;and(ii)a stage of differentiation into distinct *** early differentiation,four major cell types with specific functions differentiate within the endosperm:starchy endosperm,aleurone layer,basal endosperm transfer layer(BETL),and embryo surrounding *** subaleurone,conducting zone,basal intermediate zone,and endosperm adjacent to scutellum arise in late differentiation(Sabelli and Larkins 2009).After the main cell types are established,maize endosperm undergoes intensive cell proliferation,which marks the transition to the endosperm-filling *** the endosperm-flling stage,the maternal sucrose products are transported to seeds through the BETL region,where they will act as the main carbon substrate for storage products such as starch and proteins(Chen et al.,2023).In fact,the endosperm of maize seeds accounts for approximately 90%of the total grain weight,serving as an important nutrient reservoir(Flint-Garcia et al.,2009).

Combinatorial interactions between different regulators diversify and enrich the chance of transcriptional regulation in eukaryotic ***,a dose-dependent functional switch of homologous transcriptional repressors has rarely been ***,we show that SHY2,an auxin/indole-3-acetic acid(Aux/IAA)repressor,exhibits a dose-dependent bimodal role in auxin-sensitive root-hair growth and gene transcrip-tion in Arabidopsis,whereas other Aux/lAA homologs consistently repress the auxin *** co-repressor(TOPLESS[TPL])-binding affinity of a bimodal Aux/IAA was lower than that of a consistently re-pressing Aux/*** switch of a single amino acid residue in the TPL-binding motif between the bimodal form and the consistently repressing form switched their TPL-binding affinity and transcriptional and bio-logical roles in auxin *** on these data,we propose a model whereby competition between homologous repressors with different co-repressor-binding affinities could generate a bimodal output at thetranscriptional anddevelopmental levels.

The Cenomanian-Turonian boundary oceanic anoxic event(OAE-2; Bonarelli event; ～94 Ma) is associated with a species turnover in the marine calcareous microfossil record, widespread marine anoxia,black shale deposition and positive δ13C excursions. This study reviews 141 CTBE sites globally, aiming to understand micropaleontological, geochemical, and sedimentological expressions of OAE-2. There is a clear palaeogeographic and palaeobathymetric heterogeneity in the development of OAE-2 marine anoxia. A majority of the documented OAE-2 sites are from deep marine environments. The calcareous nannoplankton and benthic foraminifera record a diversity decline, while planktic foraminifera shows community level shifts and no major mass extinction. The variability of total organic carbon in OAE-2 sediments across sites(10 wt.%) and the δ13C profiles(diachronous) have been attributed to different mechanisms of anoxia *** increased primary “productivity model” gains support from productivity proxies(e.g., Ba, P, Cu, Ni),redox-sensitive elements(e.g., Mn, Mo, U, V, As), and eutrophic genera(e.g., benthic foraminifera Gabonita spp., calcareous nannofossils Biscutum spp. and Zeugrhabdotus erectus). The enhanced organic carbon“preservation model” in stratified(semi) restricted basins gains support from sites with lower enrichment of redox-sensitive elements and oligotrophic flora and fauna in the OAE-2 records. Geochronology of the Caribbean and the High Atlantic Large Igneous Province events(CLIP and HALIP) mark them as likely triggers of the OAE-2 related global perturbation of marine biogeochemistry. Volcanic triggers may have caused climate warming, altered hydrological cycles, enhanced continental weathering, shifts in ocean circulation, and nutrient flows. Addressing knowledge gaps, further research is urged, utilizing innovative proxies, and exploring underrepresented depositional systems to comprehensively understand OAE-2 onset and biotic crisis.

RNA interfering therapy has emerged as a promising therapeutic modality to treat *** specific and efficient delivery of RNA into a tumor is crucial for achieving effective cancer gene therapy but remains a huge ***,we report a novel furin-responsive small interfering RNA(siRNA)delivery vehicle with multiple functions for colorectal tumor treatment.A peptide-based siRNA delivery vehicle RVRR-P18-Gd,consisting of furin enzyme-specific peptide substrate Arg-Val-Arg-Arg(RVRR)with positive charge for siRNA binding,a Gd(III)chelated 1,4,7,10-tetraazacyclododecane-N,N′,N″,N‴-tetraacetic acid(DOTA)(DOTA-Gd)for magnetic resonance imaging,and purpurin 18 as photosensitizer for photodynamic therapy(PDT),was rationally designed and *** advantage of the cationic amphiphilic feature,RVRR-P18-Gd molecules spontaneously self-assembled with negatively charged Hif-1αsiRNA into stable nanoparticles via attractive electrostatic interaction,which effectively prevented siRNA degradation by nucleases,prolonged the circulation half-life,and enhanced tumor ***,the specific release of Hif-1αsiRNA mediated by endogenous furin significantly downregulated Hif-1αexpression in colorectal cancer cells,resulting in enhanced therapeutic susceptibility,and with the PDT effect,effectively suppressed HCT116 tumor growth in living *** work highlights a powerful and universal approach to precisely deliver siRNA to targeted tumors for efficient synergistic therapy.

The Myc gene is the essential oncogene in triple-negative breast cancer(TNBC).This study investigates the synergistic effects of combining Myc decoy oligodeoxynucleotides-encapsulated niosomes-selenium hybrid nanocarriers with X-irradiation exposure on the MDA-MB-468 cell *** and scramble ODNs for Myc transcription factor were designed and synthesized based on promoter sequences of the Bcl2 *** nanocarriers were synthesized by loading Myc ODNs and selenium into chitosan(Chi-Se-DEC),which was then encapsulated in niosome-nanocarriers(NISM@Chi-Se-DEC).FT-IR,DLS,FESEM,and hemolysis tests were applied to confirm its characterization and physicochemical ***,cellular uptake,cellular toxicity,apoptosis,cell cycle,and scratch repair assays were performed to evaluate its anticancer effects on cancer *** anticancer assessments were repeated under X-ray irradiation conditions(fractionated 2Gy).Physicochemical characteristics of niosomes containing SeNPs and ODNs showed that it is synthesized *** revealed that the anticancer effect of NISM@Chi-Se-DEC can be significantly improved in combination with X-ray irradiation *** can be concluded that NISM@Chi-Se-DEC nanocarriers have the potential as a therapeutic agent for cancer treatment,particularly in combination with radiation therapy and in-vivo experiments are necessary to confirm the efficacy of this nano-drug.
