PBQ[1-(4-chlorophenyl)-3-(pyridin-3-yl)urea],an enormous potent molluscicide,showed excellent Pomacea canaliculata(***)control activity and low toxicity for other aquatic organisms,but its snail-killing mechanisms are still not fully *** employed an optical method to elucidate PBQ action via a novel fluorescent viscosity probe,*** the viscosity in the test solutions increased,compared with that in pure ethanol,a 54-fold fluorescence intensity enhancement of NCV was observed in 310 cP of 90%***,NCV successfully exhibited a selective fluorescence response towards monensin-induced cellular viscosity changes in HepG2 *** liver,stomach,and foot plantar of the tested snails were frozen and sectioned for fluorescent imaging experiments after the treatment with different PBQ concentrations over various times.A significant fluorescent increase in the snail's liver was observed upon exposure to 0.75 mg/L PBQ for 72 h,which highlighted an increase in *** and eosin(HE)staining further supported PBQ-induced liver damage with a viscosity increase in *** study provides a new rapid optical visualization method to study the killing mechanisms of PBQ and may help discover new chemicals that control snail populations.

The phylogenetic and evolutionary history of Kokobera virus

Objective: To estimate the genetic diversity of Kokobera virus, the date of origin and the spread among different viruses in the endemic regions of Australia. Methods: Two datasets were built. The first consisting of 29 sequences of the NS5/3' UTR region of Kokobera group downloaded from Gen Bank, the second including only 24 sequences of Kokobera viruses, focus is on this group. Results: Bayesian time analysis revealed two different entries in Australia of Kokobera virus in the 50 s years with the dated ancestor in 1861 year. Clade A and B showed a clear separation of the Kokobera sequences according to the geographic region. Conclusions: Data from the study showed as Kokobera virus, despite of its ancient origin and its circulation before the European colonization, remained limited to the Australian Country and nowadays limited mostly to the regions were Australian marsupials are mostly found.

Nipah virus(Ni V) is a member of the genus Henipavirus of the family Paramyxoviridae,characterized by high pathogenicity and endemic in South *** is classified as a Biosafety Level-4(BSL-4) *** case-fatality varies from 40%-70% depending on the severity of the disease and on the availability of adequate healthcare *** present no antiviral drugs are available for Ni V disease and the treatment is just *** and evolutionary analyses can be used to help in understanding the epidemiology and the temporal origin of this *** review provides an overview of evolutionary studies performed on Nipah viruses circulating in different *** phylogenetic studies have been published from 2000 to 2015 years,searching on pub-med using the key words ‘Nipah virus AND phylogeny' and twenty-eight molecular epidemiological studies from 2006 to 2015 have been performed,typing the key words ‘Nipah virus AND molecular epidemiology'.Overall data from the published study demonstrated as phylogenetic and evolutionary analysis represent promising tools to evidence NiV epidemics,to study their origin and evolution and finally to act with effective preventive measure.

Recombinant adenovirus containing hyper-interleukin-6 and hepatocyte growth factor ameliorates acute-on-chronic liver failure in rats

AIM: To investigate the protective efficacy of recombinant adenovirus containing hyper-interleukin-6 (Hyper-IL-6, HIL-6) and hepatocyte growth factor (HGF) (Ad-HGF-HIL-6) compared to that of recombinant adenovirus containing either HIL-6 or HGF (Ad-HIL-6 or Ad-HGF) in rats with acute-on-chronic liver failure (ACLF).METHODS: The recombinant adenoviruses containing HIL-6 and/or HGF were constructed. We established an ACLF model, and rats were randomly assigned to control, model, Ad-GFP, Ad-HIL-6, Ad-HGF or Ad-HGF-HIL-6 group. We collected serum and liver tissue samples to test pathological changes, biochemical indexes and molecular biological ***: Attenuated alanine aminotransferase, prothrombin time, high-mobility group box 1 (HMGB1), endotoxin, tumour necrosis factor (TNF)-α and interferon-γ were observed in the Ad-HGF-, Ad-HIL-6- and Ad-HGF-HIL-6-treated rats with ACLF. Likewise, reduced hepatic damage and apoptotic activity, as well as reduced HMGB1 and Bax proteins, but raised expression of Ki67 and Bcl-2 proteins and Bcl-2/Bax ratio were also observed in the Ad-HGF-, Ad-HIL-6- and Ad-HGF-HIL-6-treated rats with ACLF. More significant changes were observed in the Ad-HGF-HIL-6 treatment group without obvious side effects. Furthermore, caspase-3 at the protein level decreased in the Ad-HIL-6 and Ad-HGF-HIL-6 treatment groups, more predominantly in the latter ***: This study identifies that the protective efficacy of Ad-HGF-HIL-6 is more potent than that of Ad-HGF or Ad-HIL-6 in ACLF rats, with no significant side effects.

We employed an optical method to elucidate PBQ action via a novel fluorescent viscosity probe, NCV. As the viscosity in the test solutions increased,compared with that in pure ethanol, a 54-fold fluorescence intensity enhancement of NCV was observed in 310 cP of 90% glycerol. Furthermore, NCV successfully exhibited a selective fluorescence response towards monensin-induced cellular viscosity changes in HepG2 cells. The liver, stomach, and foot plantar of the tested snails were frozen and sectioned for fluorescent imaging experiments after the treatment with different PBQ concentrations over various times. A significant fluorescent increase in the snail's liver was observed upon exposure to 0.75 mg/L PBQ for 72 h, which highlighted an increase in viscosity. Hematoxylin and eosin(HE) staining further supported PBQ-induced liver damage with a viscosity increase in P. canaliculata. Our study provides a new rapid optical visualization method to study the killing mechanisms of PBQ and may help discover new chemicals that control snail populations.
