Currently,human health due to corona virus disease 2019(COVID-19)pandemic has been seriously *** coronavirus severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)spike(S)protein plays a crucial role in virus transmission and several S-based therapeutic approaches have been approved for the treatment of ***,the efficacy is compromised by the SARS-CoV-2 evolvement and *** we report the SARS-CoV-2 S protein receptor-binding domain(RBD)inhibitor licorice-saponin A3(A3)could widely inhibit RBD of SARS-CoV-2 variants,including Beta,Delta,and Omicron BA.1,XBB and ***,A3 could potently inhibit SARS-CoV-2 Omicron virus in Vero E6 cells,with EC50 of 1.016μ*** mechanism was related to binding with Y453 of RBD determined by hydrogen-deuterium exchange mass spectrometry(HDX-MS)analysis combined with quantum mechanics/molecular mechanics(QM/MM)***,phosphoproteomics analysis and multi fluorescent immunohistochemistry(mIHC)respectively indicated that A3 also inhibits host inflammation by directly modulating the JNK and p38 mitogen-activated protein kinase(MAPK)pathways and rebalancing the corresponding immune *** work supports A3 as a promising broad-spectrum small molecule drug candidate for COVID-19.

Human microorganisms,including bacteria,fungi,and viruses,play key roles in several physiological and pathological *** studies discovered that tumour tissues once considered sterile actually host a variety of microorganisms,which have been confirmed to be closely related to *** concept of intratumoural microbiota was subsequently *** could colonise tumour tissues through mucosal destruction,adjacent tissue migration,and hematogenic invasion and affect the biological behaviour of tumours as an important part of the tumour *** studies have demonstrated that intratumoural microbiota potentially promote the initiation and progression of tumours by inducing genomic instability and mutations,affecting epigenetic modifications,promoting inflammation response,avoiding immune destruction,regulating metabolism,and activating invasion and *** more comprehensive and profound insights about intratumoral microbiota are continuously emerging,new methods for the early diagnosis and prognostic assessment of cancer patients have been under *** addition,interventions based on intratumoural microbiota show great potential to open a new chapter in antitumour therapy,especially immunotherapy,although there are some inevitable ***,we aim to provide an extensive review of the concept,development history,potential sources,heterogeneity,and carcinogenic mechanisms of intratumoural microorganisms,explore the potential role of microorganisms in tumour prognosis,and discuss current antitumour treatment regimens that target intratumoural microorganisms and the research prospects and limitations in this field.

This work studied the transcriptional patterns of three antibacterial genes,attacin,defensin and cecropin,during the development of Musca domestica. Quantitative analysis by real-time PCR was performed on mRNA levels in different development stages and challenged 3rd-instar larva at different time points after challenge of Musca domestica. The results revealed a predominance of the transcripts of all three genes during the 3rd-instar larvae and the adults. In the meanwhile,it revealed the greatest increase in mRNA. The transcript levels increased to 801 times,1009 times and 2500 times respectively for cecropin,attacin and defensin in 3rd-instar larvae after challenging susceptible bacterium. The results suggested that the transcriptional patterns of Musca domestica antibacterial genes were different during the different growth stages as well as the microbial challenge encountered in 3rd-instar larvae.

Objective:To predict structure and function of translationally controlled tumor protein(TCTP)from Spirometraa mansoni by bioinformatics technology,and to provide a theoretical basis for further ***:Open reading frame(ORF)of EST sequence from Spirometra mansoni was obtained by ORE finder and was translated into amino acid residue by *** structure domain was analyzed by *** the method of online analysis tools:Protparam,InterProScan,***-3.0,PSORTⅡ,BepiPred,TMHMM,VectorNT1 Suite 9 packages and Phyre2,the structure and function of the protein were predicted and ***:The results showed that the EST sequence was Sm TCTP with 173 amino acid residues,theoretical molecular weight was 19 872.0 *** protein has the closest evolutionary status with Clonorchis *** mansoni,and Schistosoma *** it had no signal peptide site and transmembrane *** structure of TCTP contained twoα-helices and eightβ-***:Sm TCTP was a variety of biological functions of protein that may be used as a vaccine candidate molecule anti drug target.

Concomitant immunity to schistosome reinfection in vivo is generally thought to be the immune response induced by and relying on living adult worms. However, a few authors \' have suggested that the mechanism of resistance may be related to the hepatic pathology caused by the primary infection. The vascular pathology might enable the challenge schistosomula to escape from the hepatic portal system, thus preventing normal parasite sequestration and maturation in the liver. Phenol oxidase has been implicated in the process of eggshell formation. We hypothesized that allylthiourea (ATU) , the most effective phenol oxidase inhibitor, may block the formation of Schistosoma japonicum ( S. j) eggshell. Here we report the establishment of a novel model ( NM) based on ATU abbreviated infection that can be used to explore the real mechanism of immunity to reinfection with S. japonicum.

Alzheimer＇s disease （AD） is a complex neurological disor- der characterized by a progressive dementia. The amyloid hypothesis states that pathogenesis is driven by the accumu- lation of amyloid β（Aβ） peptides within the brain （Hardy and Higgins, 1992）, which have multiple effects including activation of glial cells and synapse degeneration. For many years therapeutic strategies were based upon the discovery of compounds that reduced the production of Aβ in vitro and in vivo. However, the amyloid hypothesis is not universally accepted; critics pointed out poor correlations between con- centrations of Aβ in the brain and clinical disease and that, although numerous compounds reduced the production of Aβ in animal models, few had any clinical benefit in AD pa- tients. The failure of Aβ-1owering therapies in translational research has resulted in important modifications of the amy- loid hypothesis.

Background:The impact of corticosteroids on humoral responses in coronavirus disease 2019(COVID-19)sur-vivors during the acute phase and subsequent 6-month period remains *** study aimed to determine how the use of corticosteroids influences the initiation and duration of humoral responses in COVID-19 survivors 6 months after infection ***:We used kinetic antibody data from the lopinavir-ritonavir trial conducted at Jin Yin-Tan Hospital in January 2020,which involved adults hospitalized with severe COVID-19(LOTUS,ChiCTR2000029308).Anti-body samples were collected from 192 patients during hospitalization,and kinetic antibodies were monitored at all available time points after ***,plasma samples were collected from 101 COVID-19 survivors for comprehensive humoral immune measurement at the half-year follow-up *** main focus was comparing the humoral responses between patients treated with systemic corticosteroid therapy and the non-corticosteroid ***:From illness onset to day 30,the median antibody titre areas under the receiver operating characteristic curve(AUCs)of nucleoprotein(N),spike protein(S),and receptor-binding domain(RBD)immunoglobulin G(IgG)were significantly lower in the corticosteroids *** AUCs of N-,S-,and RBD-IgM as well as neutralizing antibodies(NAbs)were numerically lower in the corticosteroids group compared with the non-corticosteroid ***,peak titres of N,S,RBD-IgM and-IgG and NAbs were not influenced by *** 6-month follow-up,we observed a delayed decline for most binding antibodies,except N-IgM(𝛽−0.05,95%CI[−0.10,0.00])in the corticosteroids group,though not reaching statistical *** significant difference was observed for ***,for the half-year seropositive rate,corticosteroids significantly accelerated the decay of IgA and IgM but made no difference to N-,S-,and RBD-IgG or ***,corticosteroids group showed a trend towards d
