Melatonin(N-acetyl-5-methoxytryptamine)is known as the hormone of darkness because it is synthesized at night and involved in regulating the circadian *** hormone is primarily synthesized by the vertebrate pineal gland,but is ubiquitous among invertebrates,unicellular organisms,plants,and even cyanobacteria(Hattori and Suzuki,2024).Melatonin is well-conserved evolutionarily and possesses several physiological functions,such as immune response,bone and glucose metabolism,and memory formation besides regulating the circadian rhythm.

Extrachromosomal DNA(ecDNA) is a form of circular DNA mostly found in tumor cells. Unlike the typical chromosomal DNA, ecDNA is circular, self-replicating, and carries complete or partial gene fragments. Although ecDNA occurrence remains a rare event in cancer, recent studies have shown that oncogene amplification on ecDNA is widespread throughout many types of cancer, implying that ecDNA plays a central role in accelerating tumor evolution. ecDNA has also been associated with increased tumor mutation burden, chromosomal instability, and even tumor microenvironment remodeling. ecDNA may be crucial in influencing tumor heterogeneity, drug sensitivity,oncogenic senescence, and tumor immunogenicity, leading to a worsening prognosis for tumor patients. In this way, several clinical trials have been conducted to investigate the importance of ecDNA in clinical treatment. In this review, we summarize the biogenesis, characteristics, and current research methods of ecDNA, discuss the vital role of ecDNA-caused tumor heterogeneity in cancers, and highlight the potential role of ecDNA in cancer therapy.

Modulation of antioxidant status in streptozotocin-induced diabetic male wistar rats following intake of red palm oil and/or rooibos

Objective:To investigate the role of red palm oil(RPO),rooibos tea extract(RTE)and their combined treatment(RPO+RTE)on antioxidant status in streptozotocin(STZ)-induced diabetic ***:Diabetes mellitus was induced by a single administration of streptozotocin(50 mg/kg)and the rats were treated for 7 *** enzymes[calalase(CAT),glutathione peroxidase(GPx),superoxide dismutase(SOD)],antioxidant capacity[trolox equivalence antioxidant capacity(TEAC),oxygen radical absorbance capacity(ORAC)]as well as total protein,albumin,globulin,total glutathione,conjugated diene and thiobarbituric acid reactive substances(TBARS)were ***:Treatment with RPO,RTE and RPO+RTE significantly(p>0.05)improved liver SOD and plasma ORAC in the diabetic ***,diabetic rats treated with RTE and RPO+RTE enhanced liver GPx.A significant(P<0.05)increase in the plasma TBARS in the diabetic control group was observed when compared with the normal control *** of diabetic rats with RTE and RPO+RTE reduced plasma TBARS to a level not significantly different at P<0.05 from the normal control ***:The results revealed the anti-oxidative potentials of red palm oil,rooibos and their combination in diabetic conditions and hence,they could be useful in the management of diabetes and its complications.

OBJECTIVE：The morphological and color characteristics of the tongue sublingual veins（SLVs）can manifest differently within the subjects,depending on the way their tongue is curled *** study was conducted in order to investigate the clinical relevancy of tongue SLV diagnosis in relation to menstrual clinical symptoms（pain,clots,heavy,and scanty）,using three different inspection procedures（IP1,IP2,and IP3）.METHODS：Three-hundred and seventy-seven female patients were asked to stick out their tongues in three specifi c ways which were intended to create different tongue protrusion *** SLV parameters for thickness（TK）,length（LE）,color（CL）,shape（SP）,and nodules（ND）were then ***：According to the results of the Waldχ2test,IP1 provides the best model for pain（R2=0.155）,IP3 for clots（R2=0.437）,IP2 for heavy（R2=0.268）,and scanty（R2=0.192）.Abnormal SLV diagnostic parameters were most strongly associated with the clinical symptom of clots（R2=0.492）.CONCLUSION：While the study showed the relations between tongue SLV features and menstrual clinical symptoms,as well it showed that IP2 was the best overall predictor for the symptomatic indexes used in this study,and using one particular SLV inspection procedure may not be suffi *** application of a particular inspection method alone may cause under-or over-estimation of SLV abnormalities.

Microtubule-associated protein Tau is responsible for the stabilization of neuronal microtubules under normal physiological *** attention has been focused on Tau’s contribution to cognition,but little research has explored its role in emotions such as pain,anxiety,and *** the current study,we found a significant increase in the levels of p-Tau(Thr231),total Tau,IL-1β,and brain-derived neurotrophic factor(BDNF)on day 7 after complete Freund's adjuvant(CFA)injection;they were present in the vast majority of neurons in the spinal dorsal *** of Mapt-shRNA recombinant adeno-associated virus into the spinal dorsal cord alleviated CFA-induced inflammatory pain and inhibited CFA-induced IL-1βand BDNF ***,Tau overexpression was sufficient to induce hyperalgesia by increasing the expression of IL-1βand ***,the activation of glycogen synthase kinase 3 beta partly contributed to Tau *** findings suggest that Tau in the dorsal horn could be a promising target for chronic inflammatory pain therapy.

Objectives Journaling is a common non-pharmacological tool in the management of mental illness,however,no clear evidence-based guideline exists informing primary care providers on its *** seek here to present this synthesis that may begin to inform future research and eventual evidence-based guideline *** Of the 3797 articles retrieved from MEDLINE,EMBASE,PsycINFO,20 peer-reviewed randomised control trials(31 outcomes)met inclusion *** studies addressed the impact of a journaling intervention on PTSD,other anxiety disorders,depression or a combination of the *** criteria Peer reviewed,randomised control trials on the impact of journaling on mental illness were *** sources MEDLINE,EMBASE and *** The data are highly heterogeneous(control arm=I^(2) of 71.2%,intervention arm=I^(2) of 83.8%)combined with a B-level Strength of Recommendation Taxonomy *** was additionally found that there is a significant pre-post psychometric scale difference between control(−0.01,95%CI−0.03 to 0.00)and intervention arms(−0.06,95%CI−0.09 to−0.03).This 5%difference between groups indicates that a journaling intervention resulted in a greater reduction in scores on patient health ***’s d effect size analysis of studies suggests a small to moderate *** Further studies are needed to better define the *** review suggests that while there is some randomised control data to support the benefit of journaling,high degrees of heterogeneity and methodological flaws limit our ability to definitively draw conclusions about the benefit and effect size of journaling in a wide array of mental *** the low risk of adverse effects,low resource requirement and emphasis on self-efficacy,primary care providers should consider this as an adjunct therapy to complement current evidence-based management.
