Cancer cell cycle heterogeneity as a critical determinant of therapeutic resistance

Intra-tumor heterogeneity is now arguably one of the most-studied topics in tumor biology,as it represents a major obstacle to effective cancer *** tumor cells are highly diverse at genetic,epigenetic,and phenotypic levels,intra-tumor heterogeneity can be assumed as an important contributing factor to the nullification of chemotherapeutic effects,and recurrence of the *** on the role of heterogeneous subpopulations of cancer cells with varying cell-cycle dynamics and behavior during cancer progression and treatment;herein,we aim to establish a comprehensive definition for adaptation of neoplastic cells against *** discuss two parallel and yet distinct subpopulations of tumor cells that play pivotal roles in reducing the effects of chemotherapy:"resistant"and"tolerant"***,this review also highlights the impact of the quiescent phase of the cell cycle as a survival mechanism for cancer *** understanding the mechanisms under-lying the quiescence,it provides an insightful perspective on cancer stem cells(CsCs)and their dual and intertwined functions based on their cell cycle state in response to ***-over,CSCs,epithelial-mesenchymal transformed cells,circulating tumor cells(CTCs),and disseminated tumor cells(DTCs),which are mostly in a quiescent state of the cell cycle are proved to have multiple biological links and can be implicated in our viewpoint of cell cycle heterogeneity in ***,increasing our knowledge of cell cycle heterogeneity is a key to identifying new therapeutic solutions,and this emerging concept may provide us with new opportunities to prevent the dreadful cancer recurrence.

Fermented calcium butyrate supplementation in post-peak laying hens improved ovarian function and tibia quality through the“gut-bone” axis

The compromised egg quality and leg abnormality during the end of the laying cycle(after 40 weeks)have been leading to poor animal welfare and substantial economic losses. Therefore, the effects of fermented calcium(Ca) butyrate, produced by fermentation by Clostridium butyricum, on production,eggshell quality, and tibial property of hens were explored. A total of 192 Hy-line brown laying hens at50-week-old were assigned to a basal diet or the basal diet with 300 mg/kg of the fermented Ca butyrate from 50 to 58 weeks of age. Each treatment had 6 replicates with 16 hens each. The diet supplemented with 300 mg/kg fermented Ca butyrate notably increased egg weight, ovarian follicle number, and eggshell strength(P = 0.072) as compared to the basal diet, which were associated with cytokine secretion, toll-like receptor signaling pathways, and intestinal immunity based on the RNA-seq data from the granulosa. Dietary Ca butyrate inclusion decreased the expression of ileal tumor necrosis factor-alpha and serum pro-inflammatory cytokine concentration, as well as increased the content of serum immunoglobulin A when compared to the basal diet(both P 0.05). In addition, the diet with fermented Ca butyrate reduced the number of osteoclasts in the proximal tibia and the level of C-terminal cross-linked telopeptide of type I collagen, a bone resorption marker(P < 0.05), whereas it tended to increase the concentration of the procollagen type I N-terminal propeptide that reflects bone formation marker in serum. Moreover, the layers fed fermented Ca butyrate diets possessed higher(P < 0.05) bone area and trabecular number of the proximal tibia, yield load, and ultimate load than those that consumed basal diets. Numerical and analytical studies on the coupling effects of unloading and cutterhead vibration on tunnel face in dry sandy ground

When tunnelling in difficult ground conditions,shield machine would inevitably produce significant ground loss and vibration,which may disturb the ground ahead of the tunnel *** this paper,discrete element models calibrated by model tests were established to investigate the response of tunnel face under the coupling effects of unloading and cutterhead *** results show that the friction angle reduction under cyclic loading and vibration attenuation in the sandy ground are significant and can be estimated by the fitted exponential *** cutterhead vibration,the tunnel face stability is undermined and the limit support pressure(LSP)increases to 1.4 times as that in the static case with the growth of frequency and ***,the loosening zone becomes wider and the arching effect is weakened with the reduction of peak horizontal stress and the increase of vertical stress above the *** on the numerical results,a pseudo-static method was introduced into the limit equilibrium analysis of the wedge-prism model for calculating the LSP under *** an error rate less than 5.2%,the proposed analytical method is well *** analytical calculation reveals that the LSP would increase with the growth of vibration amplitude,vibration frequency and covered depth but decrease with the increase of friction *** study can not only lay a solid foundation for the further investigation of ground loss,ground water and soft-hard heterogeneous ground under cutterhead vibration,but also provide meaningful references for the control of environmental disturbance in practice.

Flexible load can optimize the load curve,which is an important means to promote renewable energy *** peculiarities of electricity,heat,cooling and gas loads are analyzed in this paper,considering the fuzzy degree of human perception for water temperature,and the characteristic model of hot water load is *** the fuzzy degree of human perception of ambient temperature,the characteristic model of cooling load is established by using PMV and PPD ***,considering four combinations of cut load,translatable load,transferable load and alternative load,and considering the coupling relationship of composite parts,different response models of load are established *** the minimum cost of the system,including operation and compensation costs as the objective function,the optimization scheduling model of the regional integrated energy system is established,and the Gurobi solver is used for simulation analysis to solve the optimal output and load response curve of each piece of *** results show that the load curve can be optimized,the flexible regulation ability of the regional integrated energy system can be enhanced,the energy loss of the system can be reduced,and the wind power consumption ability of the system can be increased by considering the integrated demand response.

Benefits of jejunostomy feeding in patients who underwent gastrectomy for cancer treatment

BACKGROUND Gastric cancer is associated with significant undernutrition responsible for an increase in morbidity and mortality after *** To evaluate the impact of enteral nutrition by jejunostomy feeding in patients undergoing gastrectomy for *** Between 2003 and 2017,all patients undergoing gastrectomy for cancer treatment were included retrospectively.A group with jejunostomy(J+group)and a group without jejunostomy(J-group)were *** Of the 172 patients included,60 received ***,the two groups were comparable with respect to the nutritional parameters studied(body mass index,albumin,etc.).In the postoperative period,the J+group lost less weight and albumin:5.74±8.4 vs 9.86±7.5 kg(P=0.07)and 7.2±5.6 vs 14.7±12.7 g/L(P=0.16),*** morbidity was 25%in the J+group and 36.6%in the J-group(P=0.12).The J+group had fewer respiratory,infectious,and grade 3 complications:0%vs 5.4%(P=0.09),1.2%vs 9.3%(P=0.03),and 0%vs 4.7%(P=0.05),*** 30-day mortality was 6.7%in the J+group and 6.3%in the J-group(P=0.91).CONCLUSION Jejunostomy feeding after gastrectomy improves nutritional characteristics and decreases postoperative morbidity.A prospective study could confirm our results.

CONSTANS alters the circadian clock in Arabidopsis thaliana

An innovative heterogeneous transfer learning framework to enhance the scalability of deep reinforcement learning controllers in buildings with integrated energy systems

Deep Reinforcement Learning(DRL)-based control shows enhanced performance in the management of integrated energy systems when compared with Rule-Based Controllers(RBCs),but it still lacks scalability and generalisation due to the necessity of using tailored models for the training *** Learning(TL)is a potential solution to address this ***,existing TL applications in building control have been mostly tested among buildings with similar features,not addressing the need to scale up advanced control in real-world scenarios with diverse energy *** paper assesses the performance of an online heterogeneous TL strategy,comparing it with RBC and offline and online DRL controllers in a simulation setup using EnergyPlus and *** study tests the transfer in both transductive and inductive settings of a DRL policy designed to manage a chiller coupled with a Thermal Energy Storage(TES).The control policy is pre-trained on a source building and transferred to various target buildings characterised by an integrated energy system including photovoltaic and battery energy storage systems,different building envelope features,occupancy schedule and boundary conditions(e.g.,weather and price signal).The TL approach incorporates model slicing,imitation learning and fine-tuning to handle diverse state spaces and reward functions between source and target *** show that the proposed methodology leads to a reduction of 10% in electricity cost and between 10% and 40% in the mean value of the daily average temperature violation rate compared to RBC and online DRL ***,online TL maximises self-sufficiency and self-consumption by 9% and 11% with respect to ***,online TL achieves worse performance compared to offline DRL in either transductive or inductive ***,offline Deep Reinforcement Learning(DRL)agents should be trained at least for 15 episodes to reach the same level of performance as the online

Flexural behavior of textile reinforced mortar-autoclaved lightweight aerated concrete composite panels

To improve the deficiencies of prefabricated autoclaved lightweight aerated concrete(ALC)panel such as susceptibility to cracking and low load-bearing capacity,a textile-reinforced mortar-autoclaved lightweight aerated concrete(TRM-ALC)composite panel was developed in this *** group of reference ALC panels and five groups of TRM-ALC panels were fabricated and subjected to four-point flexural *** was applied on the tensile side of the ALC panels to create *** variable parameters were the plies of textile(one or two),type of textile(basalt or carbon),and whether the matrix(without textile)was applied on the compression side of *** results showed that a bonding only 8-mm-thick TRM layer on the surface of the ALC panel could increase the cracking load by 180%−520%.The flexural capacity of the TRM-ALC panel increased as the number of textile layers *** reinforcement of the matrix on the compressive side could further enhance the stiffness and ultimate loadbearing capacity of the TRM-ALC *** panels with basalt textile failed in flexural mode,with the rupture of fabric *** with carbon textile failed in shear mode due to the ultra-high tensile strength of *** addition,analytical models related to the different failure modes were presented to estimate the ultimate load-carrying capacity of the TRM-ALC panels.
