Genetic variant in a BaP-activated super-enhancer increases prostate cancer risk by promoting AhR-mediated FAM227A expression

Genetic variants in super-enhancers(SEs)are increasingly implicated as a disease risk-driving *** studies have reported an associations between benzo[a]pyrene(BaP)exposure and some malignant tumor ***,it is unclear whether BaP is involved in the effect of genetic variants in SEs on prostate cancer risk,nor the associated intrinsic molecular *** the current study,by using logistic regression analysis,we found that rs5750581T>C in 22q-SE was significantly associated with prostate cancer risk(odds ratio=1.26,P=7.61×10^(-5)).We also have found that the rs6001092T>G,in a high linkage disequilibrium with rs5750581T>C(r^(2)=0.98),is located in a regulatory aryl hydrocarbon receptor(AhR)motif and may interact with the FAM227A promoter in further bioinformatics *** then performed a series of functional and BaP acute exposure experiments to assess biological function of the genetic variant and the target ***,the rs6001092-G allele strengthened the transcription factor binding affinity to AhR,thereby upregulating FAM227A,especially upon exposure to BaP,which induced the malignant phenotypes of prostate *** current study highlights that AhR acts as an environmental sensor of BaP and is involved in the SE-mediated prostate cancer risk,which may provide new insights into the etiology of prostate cancer associated with the inherited SE variants under environmental carcinogen stressors.

Resilience effects for household food expenditure and dietary diversity in rural western China

A more resilient livelihood is increasingly recognized as an efficient way to improve vulnerable households’food security and optimize their dietary *** study quantifies rural household resilience in western China,identifies the three pillars(absorptive capacity,adaptive capacity,and transformative capacity)contribution to resilience,and then establishes the estimated Resilience Capacity Index(RCI)linked with food security and dietary diversity supported by the multiple indicator multiple cause(MIMIC)*** show that,despite geographical heterogeneity,the RCI consistently increased from 2015 to *** with a higher RCI inheriting better capacity to deal with risk and shocks are significantly and positively correlated with increasing food expenditure and diversifying food *** can be because resilient households will allocate more money to food expenditure instead of saving for livelihood ***,policymakers can provide more incentives for rural households to adopt more dynamic and effective risk management ***,in turn,could yield positive spillover effects by preventing human capital loss associated with dietary-related chronic diseases and mortality.

BACKGROUND Suprasellar germinomas are rare intracranial tumors frequently associated with permanent endocrine *** present the clinical picture,treatment,and complications of suprasellar germinoma at pediatric age which,besides being lifethreatening,has lifelong endocrinological *** SUMMARY A 12-year-old female patient was presented having had intensive headaches for three weeks and visual disturbances for six *** ophthalmological examination revealed bilateral papilledema and a marked loss of *** brain magnetic resonance imaging(MRI)showed a suprasellar tumor,involving the infundibulum and the optic chiasm,extending to the third *** tests confirmed decreased levels of thyroxine,cortisol,gonadotropins,and insulin-like growth factor *** tumor reduction was performed,and immunohistopathology established the diagnosis of suprasellar *** of the spine and cerebrospinal fluid cytology confirmed the localized *** chemotherapy and radiotherapy were performed according to the SIOP CNS GCT II protocol.A post-treatment MRI showed no residual tumor,but pituitary function had not *** and a half years after the end of the treatment,the patient is in a complete remission,requiring hormonal replacement therapy,continuous education,and psychological *** This complex case highlights the importance of timely diagnosis,a multidisciplinary approach,and close follow-up in children with suprasellar germinomas.

Tauopathies are a group of neurological disorders,including Alzheimer’s disease and frontotemporal dementia,which involve progressive neurodegeneration,cognitive deficits,and aberrant tau protein *** development of tauopathies cannot currently be stopped or slowed down by treatment *** the significant contribution of tau burden in primary tauopathies and the strong association between pathogenic tau accumulation and cognitive deficits,there has been a lot of interest in creating therapies that can alleviate tau pathology and render neuroprotective ***,small molecules,immunotherapies,and gene therapy have been used to reduce the pathological tau burden and prevent neurodegeneration in animal models of ***,the major pitfall of the current therapeutic approach is the difficulty of drugs and gene-targeting modalities to cross the blood-brain barrier and their unintended side *** this review,the current therapeutic strategies used for tauopathies including the use of oligonucleotide-based gene therapy approaches that have shown a promising result for the treatment of tauopathies and Alzheimer’s disease in preclinical animal models,have been discussed.

Antitumor therapies based on adoptively transferred T cells or oncolytic viruses have made significant progress in recent years,but the limited efficiency of their infiltration into solid tumors makes it difficult to achieve desired antitumor effects when used *** this study,an oncolytic virus(rVSV-LCMVG)that is not prone to induce virus-neutralizing antibodies was designed and combined with adoptively transferred T *** transforming the immunosuppressive tumor microenvironment into an immunosensitive one,in B16 tumor-bearing mice,combination therapy showed superior antitumor effects than *** occurred whether the OV was administered intratumorally or *** therapy significantly increased cytokine and chemokine levels within tumors and recruited CD8^(+)T cells to the TME to trigger antitumor immune *** with adoptively transferred T cells and subsequent oncolytic virotherapy sensitizes refractory tumors by boosting T-cell recruitment,downregulating the expression of PD-1,and restoring effector T-cell *** offer a combination therapy with greater translational value,mRNA vaccines were introduced to induce tumor-specific T cells instead of adoptively transferred T *** combination of OVs and mRNA vaccine also displays a significant reduction in tumor burden and prolonged *** study proposed a rational combination therapy of OVs with adoptive T-cell transfer or mRNA vaccines encoding tumor-associated antigens,in terms of synergistic efficacy and mechanism.

Background:The aim of the study was to evaluate the outcomes of dissatisfed patients reporting poor visual quality following implantation of multifocal intraocular lenses(MF-IOLs),managed by IOL exchange with another multifocal optical ***:This is a retrospective series of ***-IOL exchange was done in 15 dissatisfed patients(30 eyes)with the perception of poor visual quality for far distance afected by neuroadaptation *** underwent a bilateral exchange of a MF-IOL with another MF-IOL of a diferent optical *** outcomes and complications were *** including Quality of Vision(QoV),Visual Function Index-14(VF-14)and its Rasch-revised version(VF-8R)and a satisfaction questionnaire were also used for outcome ***:The mean elapsed time from implantation to explantation-reimplantation was 11.8 *** QoV scores improved signifcantly across all the three *** function improved with a change in VF-14 score from 60.41±24.81 to 90.16±10.91(P<0.001).The VF-8R score improved as *** uncorrected distance visual acuity improved from 0.24 to 0.12 logMAR after exchange(P<0.001)and corrected distance visual acuity improved from 0.15 to 0.04 logMAR(P<0.001).Safety and efcacy indexes reached 1.46 and 1.16,*** patients’satisfaction following MF-IOL exchange,80%of the patients reported they would have the MF-IOL reimplantation procedure ***:Patient dissatisfaction with neuroadaptation failure following MF-IOL implantation can be managed in 80%of our cases by MF-IOL exchange with a diferent MF-IOL optical profle.

In the era of rapid development of Internet of Things(IoT),numerous machine-to-machine technologies have been applied to the industrial *** to the divergence of IoT solutions,the industry is faced with a need to apply various technologies for automation and *** fact leads to a demand for an establishing interworking mechanism which would allow smooth interoperability between heterogeneous *** of the major protocols widely used today in industrial electronic devices is ***,data generated by Modbus devices cannot be understood by IoT applications using different protocols,so it should be applied in a couple with an IoT service layer ***2M,a global IoT standard,can play the role of interconnecting various protocols,as it provides flexible tools suitable for building an interworking framework for industrial ***,in this paper,we propose an interworking architecture between devices working on the Modbus protocol and an IoT platform implemented based on oneM2M *** the proposed architecture,we introduce the way to model Modbus data as oneM2M resources,rules to map them to each other,procedures required to establish interoperable communication,and optimization methods for this *** analyze our solution and provide an evaluation by implementing it based on a solar power management use *** results demonstrate that our model is feasible and can be applied to real case scenarios.

Tripartite partnership, compared with the bilateral model, is envisioned as an opportunity to foster stronger and more trusting partnerships [1]. However, China has historically been characterized as principally reliant on bilateral engagement in global health [2]. The China-Tanzania Cooperation Project on Malaria Control was conducted by National Institute of Parasitic Diseases at China CDC and Chinese Center for Tropical Diseases Research, Ifakara health Institute in Tanzania and funding agencies from 2015 to 2022, with an aim to explore the applicability of Chinese experience on malaria control to reduce malaria disease burden in the local context of Tanzania. In the tripartite project funded by the former UK department for International development (DFID) and later the Bill & Melinda Gates Foundation (BMGF), China's partners were expected to share the extensively parallel practices achieved from the control and elimination of malaria for over 70 years, UK's partners were to deliver resources and design global health strategies, while Tanzanian local staff collaborated with triangular partners to ensure the project implementation [3]. In the project, the trilateral cooperation has transparent management mechanism to ensure the concrete management and implementation of the project [4] (Additional file 1: Figure S1). This Brief is to provide recommendations for China's engagement in global health by distilling the experience from the innovative tripartite partnership of the project.
