P25/CDK5-mediated Tau Hyperphosphorylation in Both Ipsilateral and Contralateral Cerebra Contributes to Cognitive Deficits in Post-stroke Mice

Objective Post-stroke cognitive impairment(PSCI)develops in approximately one-third of stroke survivors and is associated with ***,the biochemical mechanisms underlying PSCI remain *** study aimed to establish an ischemic mouse model by means of transient unilateral middle cerebral artery occlusions(MCAOs)and to explore the biochemical mechanisms of p25/cyclin-dependent kinase 5(CDK5)-mediated tau hyperphosphorylation on the PSCI *** Cognitive behavior was investigated,followed by the detection of tau hyperphosphorylation,mobilization,activation of kinases and/or inhibition of phosphatases in the lateral and contralateral cerebrum of mice following ischemia in MACO ***,we treated HEK293/tau cells with oxygen-glucose deprivation(OGD)and a CDK5 inhibitor(Roscovitine)or a GSK3βinhibitor(LiCl)to the roles of CDK5 and GSK3βin mediating ischemia-reperfusion-induced tau *** Ischemia induced cognitive impairments within 2 months,as well as causing tau hyperphosphorylation and its localization to neuronal somata in both ipsilateral and contralateral ***,p25 that promotes CDK5 hyperactivation had significantly higher expression in the mice with MCAO than in the shamoperation(control)group,while the expression levels of protein phosphatase 2(PP2A)and the phosphorylation level at Tyr307 were comparable between the two *** addition,the CDK5 inhibitor rescued tau from hyperphosphorylation induced by *** These findings demonstrate that upregulation of CDK5 mediates tau hyperphosphorylation and localization in both ipsilateral and contralateral cerebra,contributing to the pathogenesis of PSCI.

Effects of <i>Baicalin</i>and <i>Ligustrazine</i>on airway inflammation and remodeling and underlying mechanism in asthmatic rats

Aim: To explore the effects of Baicalin and Ligustrazine on airway inflammation and construction and underlying mechanisms through the expressions of GATA-3, IL-5, MMP-9 and TIMP-1 in asthmatic rats. Methods: 30 Wistar rats were randomly divided equally into five groups. Lung tissues were sliced. WBC and Eos in lung tissue were estimated by HE stain and the expressions of IL-5, GATA-3, MMP-9, TIMP-1 and collagen type IV in lung tissue were observed by immunohistochemistry. The airway wall and airway smooth muscle thicknesses were measured by computed image analysis system. Results: Compared with asthma group, EOS counts and the expression of IL-5 and GATA-3 in the lung tissue were significantly lower in normal controlled groups (P Baicalin or Ligustrazine, EOS decreased, and the thicknesses of airway wall and airway smooth muscle became thinner compared with asthma group. Meanwhile, the expression of collagen type IV, IL-5, GATA-3, MMP-9 and TIMP-1 significantly decreased (P < 0.05). Airway wall thickness and collagen type Ⅳ were associated with Eos, IL-5, TAGA-3, MMP-9, TIMP-1 and MMP-9/ TIMP-1. Conclusion: Two herbs could diminish infiltration of EOS with inhibiting the expressions of IL-5, and GATA-3, meanwhile, decrease the deposition of collagen type IV and the thickness of the airway smooth muscle through regulating MMP-9, TIMP-1 level and the balance between MMP-9 and TIMP-1, additionally, had synergetic effects.

Regional tissue oxygen saturation as a predictor of post-spinal anesthesia hypotension for cesarean delivery

Since the discovery of neuraxial blockade,spinal anesthesia has been frequently used for cesarean *** to an international consensus statement on hypotension management,Kinsella et al[1]reported the incidence of post-spinal anesthesia hypotension in parturient women to be as high as 70%to 80%.Post-spinal anesthesia hypotension is mainly caused by sympathetic vasomotor block,which results in reduced systemic vascular resistance,venous pooling in capacitance vessels,and decreased cardiac *** hypotension may lead to maternal and fetal adverse *** maternal hypotension may contribute to reduced uterus blood flow resulting in decreased placental perfusion and fetal hypoxia leading to acute fetal *** women having a positive preoperative stress test were associated with an increased risk of post-spinal anesthesia hypotension during cesarean delivery.[2]However,the sensitivity of the stress test for predicting post-spinal anesthesia hypotension was only 69%.[2]In clinical practice,non-invasive,reliable,and readily available techniques are still much in need to predict the incidence of hypotension after spinal anesthesia as well as to improve the management of spinal anesthesia-induced hypotension for cesarean delivery.

Objective: Tumor markers play an important role in the early diagnosis and guidance of prognosis of lung cancer. This study focused on the significance of combined monitoring of IL-6 and tumor markers in improving the specificity and sensitivity of lung cancer diagnosis and ***: ELISA was used to detect the expression of IL-6, CEA, NSE and CYFRA21-1 in NSCLC and BPL patients to analyze the correlation between IL-6 and CEA, NSE or CYFRA21-1 expression. The expression of p-p44/42 MAPK and p-AKT were detected by ***: The expression of IL-6, CEA, NSE and CYFRA21-1 in serum of patients with NSCLC was significantly higher compared to patients with benign pulmonary lesions(BPL). The simultaneous increase in the expression of IL-6 and tumor markers worsened the prognosis of patients. The expression of CEA,NSE, CYFRA21-1, p-Erk1/2 and p-AKT in the IL-6 group was significantly higher compared to the other groups. Conclusion: These studies indicate that the combined detection of IL-6 and tumor markers is critical to improve the specificity and sensitivity of lung cancer diagnosis.

Impairment of Myocardial and Skeletal Mitochondria in Mice with Viral Myocarditis and Their Correlation

In order to investigate the impairment of mitochondrial membrane phospholipid local- ization and DNA3867 (mtDNA3867) deletion and the correlation between cardiac and skeletal muscle cells in mice with viral myocarditis, 50 BALB/c mice were divided into two groups randomly. In ex- perimental group (n=40), the mice were intraperitoneally injected with 0.1 mL Eagle liquid with CVB3 (TCID50=108), while in the control group (n=10), the mice were subjected to equal volume of Eagle liquid. The impairment of mitochondrial membrane phospholipid localization and mtDNA3867 deletion rate of cardiac and skeletal muscle were detected separately at day 3, 11 and 24 after injec- tion. The correlation of mitochondrial membrane phospholipid localization and mtDNA3867 deletion rate between cardiac and skeletal muscle cells cells was analyzed using Spearman method. At the day 3 after injection, in both cardiac and skeletal muscle cells, mtDNA3867 deletion rate was significantly higher in experimental group than in control group (P0.05). At day 11 after injec- tion, the mtDNA3867 deletion rate of both cells in experimental group was increased to the peak level (P0.05). At the day 24 after injection, the impairment of mitochondrial membrane phospholipid localization and mtDNA3867 deletion of both cells showed a recovery tendency, but still severer than those at the day 3 after injec- tion (P<0.05). The impairment of mitochondrial membrane phospholipid localization and mtDNA3867 deletion were consistent and synchronistic between cardiac and skeletal muscle cells, and showed good correlations (P<0.05). Experimental Study of Excitability and Autorhthmicity in Urinary Bladder Detrusor of Diabetes Rats

Summary： The changes in excitability and autorhthmicity of bladder detrusor in experimental non- insulin dependent diabetes mellitus （NIDDM） rats were observed. Sixty-nine NIDDM rats as NIDDM group and 69 normal rats as control group were enrolled into this experimental study. At 6th, 10th, 14th, 18th, 22nd and 26th week after the rats were iniected last time, the changes in the excitability and autorhthmicity of detrusor strips in vitro were observed. The results showed that the threshold of the tension which made the detrusor strips contract was significantly higher in NIDDM group （0.716±0.325 g） than in control group （0.323±0. 177 g）（F=59.63, P〈0.001）. At different stages, the threshold of the tension resulting the contract of the detrusor strips in NIDDM group was also higher than in control group. At 18th week after STZ iniection, the frequency of spontaneous contract of the detrusor strips in NIDDM was significantly higher than in control group （P〈0.05）, whereas at 22nd week, that in NIDDM group was significantly lower than in control group （P〈0. 05）. It was concluded that the decreased excitability of the bladder detrusor was the earliest and most obvious changes in bladder function in diabetes rats and the autorhthmicity had also changed at the early stage of diabetic bladder.

Objective: Correlation between lung cancer tumor markers and gender differences in lung cancer remains a clinical problem that is worthy of further study. This study investigated the significance of combined detection of 17β-Estrogen(E2) and tumor markers in the diagnosis and prognosis of lung ***: ELISA was used to detect the expression of E2, CEA, NSE and CYFRA21-1 in NSCLC and BPL patients to analyze the correlation between E2 and CEA, NSE or CYFRA21-1 expression. The expression of p-p44/42 MAPK and p-AKT were detected by ***: The expression levels of E2, CEA, NSE and CYFRA21-1 in NSCLC patients were significantly higher than in BPL(P<0.05), E2 was positively correlated with tumor markers(P<0.01). Patients with high expression of E2 and tumor markers showed poor prognosis(P<0.05).RT-qPCR and Western-blot analysis showed that the expression levels of CEA, NSE, CYFRA21-1,p-p44/42 MAPK and p-AKT in E2 group was higher than that in other groups(P<0.05). Conclusion:These studies indicate that the interaction of E2 and tumor markers can significantly improve the role of tumor markers in the diagnosis and prognosis of lung cancer.

To investigate the effects of sodium selenite on telomerase activity and expression of hTERT mRNA in cadmium-transformed 16HBE cells. Methods Telomerase activity and expression of genes were measured after cultured cadmium-transformed 16HBE cells were exposed to sodium selenite at different doses （0.625, 1.25, 2.50, 5.00 pmol/L） for 24 hours. Results Selenium decreased telomerase activity in cadmium-transformed 16HBE cells. There existed an obvious dose-effect relationship between the selenium concentration and these changes. The expression of hTERT and c-myc mRNA also decreased but the expression of madl mRNA increased after exposure to selenium for 24 hours. No difference was found in expression of hTRF1 and hTRF2 mRNA after incubated with sodium selenite for 24 hours, compared with control group. Conclusion Selenium inhibits telomerase activity by decreasing hTERT and c-myc mRNA expression and increasing madl mRNA expression in cadmium-transformed 16HBE cells and selenium concentration is significantly correlated with these changes.
