AIM: To investigate the role of hepatic peroxisome proliferator-activated receptor-γ (PPAR-γ) in increased susceptibility to endotoxin-induced toxicity in rats with bile duct ligation during ***: Male Sprague-Dawley rats were subjected to bile duct ligation (BDL). Sham-operated animals served as controls. DNA binding were determined by polymerase chain reaction, Western blotting analysis,and electrophoretic mobility shift assay, *** and sham-operated rats received a non-lethal dose of intraperitoneal lipopolysaccharide (LPS) injection (3 mg/kg, i.p.). Additionally, the potential beneficial effects of the PPAR-γ agonist rosiglitazone were determined in BDL and sham-operated rats treated with a non-lethal dose of LPS. Survival was assessed in BDL rats treated with a non-lethal dose of LPS and in sham-operatedrats treated at a lethal dose of LPS (6 mg/kg, i.p.).RESULTS: PPAR-γ activity in rats undergoing BDL wassignificantly lower than in the sham-controls. Hepatic PPAR-γ gene expression was downregulated at both them RNA and protein levels. In a parallel group, serumlevels of pro-inflammatory cytokines were nearly unde-tectable in the sham-operated rats. When challenged with a non-lethal dose of LPS (3 mg/kg), the BDL ratshad approximately a 2.4-fold increase in serum IL-6,a 2.7 fold increase in serum TNF-α, 2.2-fold increasein serum IL-1 and 4.2-fold increase in serum ALT. Thesurvival rate was significantly lower as compared with that in sham-operated group. Additionally, rosiglitazone significantly reduced the concentration of TNF-α, IL-1β, IL-6 and ALT in sham-operated rats, but not in BDL rats, in response to LPS (3 mg/kg). Also, the survival was improved by rosiglita zone in sham-operated rats challenged with a lethal dose of LPS, but not in BDL rats, even with a non-lethal dose of LPS (3 mg/kg).CONCLUSION: Obstructive jaundice downregulates hepatic PPAR-γ expression, which in turn may contributeto hypersensitivity towards endotoxin.

Dietary fat intake and liver cancer risk: A prospective cohort study in Chinese women

Objective:This study aimed to determine whether dietary fat intake increased liver cancer risk in Chinese women from a prospective population-based ***:A total of 72,704 Chinese women were followed up from the time of baseline recruitment(1996–2000)to the end of *** fat intake was calculated using a validated food frequency *** Cox regression model was used to assess the hazard ratio(HR)and 95%confidence intervals(CI)for dietary fat intake and liver cancer ***:We identified 252 incident liver cancer cases out of 1,267,845 person-years during the overall follow-up *** associations,neither in quartiles nor per standard deviation(SD)increment,were detected between liver cancer risk and dietary total fat,fat subtypes and subtype ratios,and food *** HR(95%CI)of the 1-SD increment was 1.03(0.90–1.17)for total fat,1.06(0.93–1.20)for saturated fat,1.06(0.93–1.21)for monounsaturated fat,and 1.00(0.89–1.13)for polyunsaturated *** null associations were observed in stratification analyses according to body mass index and menopausal ***:In our prospective cohort study,no significant association was observed in Chinese women between dietary fat and liver cancer risk,and in stratification and sensitivity analyses.

AIM： To investigate the role of subjective global assessment （SGA） in nutritional assessment and outcome prediction of Chinese patients with gastrointestinal cancer. METHODS： A total of 751 patients diagnosed with gastrointestinal cancer between August 2004 and August 2006 were enrolled in this study. Within 72 h after admission, SGA, anthropometric parameters, and laboratory tests were used to assess the nutritional status of each patient. The outcome variables including hospital stay, complications, and in-hospital medical expenditure were also ***： Based on the results of SGA, 389 （51.8%）, 332 （44.2%）, and 30 （4.0%） patients were classified into well nourished group （SGA-A）, mildly to moderately malnourished group （SGA-B）, and severely malnourished group （SGA-C）, respectively. The prevalence of malnutrition classified by SGA, triceps skinfold thickness （TSF）, mid-upper arm muscle circumference （MAMC）, albumin （ALB）, prealbumin （PA）, and body mass index （BMI） was 48.2%, 39.4%, 37.7%, 31.3%, 21.7%, and 9.6%, respectively. In addition, ANOVA tests revealed significant differences in body mass index （BMI）, TSF, PA, and ALB of patients in different SGA groups. The more severely malnourished the patient was, the lower the levels of BMI, TSF, PA, and ALB were （P 〈 0.05）. x^2 tests showed a significant difference in SGA classification between patients receiving different types of treatment （surgery vs chemotherapy/radiotherapy）. As the nutritional status classified by SGA deteriorated, the patients stayed longer in hospital and their medical expenditures increased significantly. Furthermore, multiple regression analysis showed that SGA and serum ALB could help predict the medical expenditures and hospital stay of patients undergoing surgery. The occurrence of complications increased in parallel with the increasing grade of SGA, and was the highest in the SGA-C group （23.3%） and the lowest in the SGA-A group （16.8%）. Covalent stabilization of DNA nanostructures on cell membranes for efficient surface receptor-mediated labeling and function regulations

One major challenge of using DNA nanostructures for cellular and in vivo applications is their insufficiently structural integrity that stems from the non-covalent base pairing and stacking in complex cellular and physiological environment. The establishment of covalent bonds in DNA nanostructures can link individual strands more stably and therefore should improve the performance of DNA nanostructures in different scenarios where structural integrity is required. Here, we developed a convenient and effective method for constructing covalently stabilized DNA nanostructures by chemically inserting photo-crosslinker(^(CNV)K) in DNA sequences. These covalently linked DNA nanostructures were found to be more resistant to external interference, such as low cation concentrations and unspecific displacement on cell membranes. We also demonstrated that our strategy could improve the efficiency of cell surface receptor-mediated labeling and function regulations in living cells, which sheds light on broadening the biomedical applications of DNA nanostructures.

Finite Element Modeling of Human Thorax Based on MRI Images for EIT Image Reconstruction

Electrical impedance tomography(EIT)image reconstruction is a non-linear *** general,finite element model is the critical basis of EIT image reconstruction.A 3D human thorax modeling method for EIT image reconstruction is proposed herein to improve the accuracy and reduce the complexity of existing finite element modeling *** contours of human thorax and lungs are extracted from the layers of magnetic resonance imaging(MRI)images by an optimized Otsu’s method for the construction of the 3D human thorax model including the lung ***,the GMSH tool is used for finite element subdivision to generate the 3D finite element model of human *** proposed modeling method is fast and accurate,and it is universal for different types of MRI *** effectiveness of the proposed method is validated by extensive numerical simulation in *** results show that the individually oriented 3D finite element model can improve the reconstruction quality of the EIT images more effectively than the cylindrical model,the 2.5D model and other human chest models.

Background Moyamoya disease (MMD) is a rare and complex cerebrovascular disorder characterized by the progressive narrowing of the internal carotid arteries and the formation of compensatory collateral vessels. The etiology of MMD remains enigmatic, making diagnosis and management challenging. The MOYAOMICS project was initiated to investigate the molecular underpinnings of MMD and explore potential diagnostic and therapeutic *** The MOYAOMICS project employs a multidisciplinary approach, integrating various omics technologies, including genomics, transcriptomics, proteomics, and metabolomics, to comprehensively examine the molecular signatures associated with MMD pathogenesis. Additionally, we will investigate the potential influence of gut microbiota and brain-gut peptides on MMD development, assessing their suitability as targets for therapeutic strategies and dietary interventions. Radiomics, a specialized field in medical imaging, is utilized to analyze neuroimaging data for early detection and characterization of MMD-related brain changes. Deep learning algorithms are employed to differentiate MMD from other conditions, automating the diagnostic process. We also employ single-cellomics and mass cytometry to precisely study cellular heterogeneity in peripheral blood samples from MMD *** The MOYAOMICS project represents a significant step toward comprehending MMD’s molecular underpinnings. This multidisciplinary approach has the potential to revolutionize early diagnosis, patient stratification, and the development of targeted therapies for MMD. The identification of blood-based biomarkers and the integration of multiple omics data are critical for improving the clinical management of MMD and enhancing patient outcomes for this complex disease.

Radiofrequency ablation of hepatocellular carcinoma: Current status,challenges, and prospects

Local ablation technologies, such as radiofrequency ablation (RFA), microwave ablation (MWA) andcryoablation, have become a standard treatment option for hepatocellular carcinoma (HCC) less than5 cm in size, particularly in individuals who are not candidates for hepatectomy. Except for equivalentprognosis and efficiency, RFA has various advantages over surgical excision, including a lower rate ofcomplications, a cheaper cost, more normal tissue preservation, and a shorter hospital stay. However, therate of tumor recurrence and/or distant metastasis after RFA therapy is still high. RFA has been widelyemployed in multiple cancers, large cancer, and lesion identified at “high-risk” sites in recent years, withthe advancement of ablation types and operating techniques, particularly the combined use of manytechnologies. The real value of RFA technology has been more fully *** will examine the status,progress, and problems of RFA in the treatment of HCC in this review.
