Sea buckthorn(Hippophae rhamnoides L.)is a natural homologous substance of medicine and ***,as one of its primary active components,has very superior biological activity and can be used as a dietary supplement for functional foods,with good commercial *** initial progress has been made in the study of sea buckthorn polysaccharides,related studies have been fragmented and lacked systematic and *** manuscript presents a critical analysis and systematic summary of the extraction and purification methods,structural characterization and physicochemical properties,biological activity and potential mechanisms,and structure-activity relationships of sea buckthorn *** evidence has indicated that sea buckthorn polysaccharides,which were widely prepared by water extraction and column chromatography purifications,exhibited exhibit superior biological activities in vitro and in vivo,including antioxidant,immunomodulatory,anti-inflammatory,hepatorenal protective,antibacterial,antiviral,and prebiotic *** analysis,it was concluded that there is a correlation between the relevant activities of sea buckthorn polysaccharides and that the structure of sea buckthorn polysaccharides has a great influence on their biological *** reviewed the challenges and limitations of sea buckthorn polysaccharides,summarized the critical aspects,and provided suggestions for potential breakthroughs in the research and application of sea buckthorn polysaccharide.

Objective:The objective of this study was to determine the level of methotrexate(MTX)toxicity in the intestines of mice and to evaluate the protective effect of probiotics composed of Streptococcus,Bifidobacterium,and Lactobacillus species on intestinal cells during MTX ***:Mice were divided into three groups:control,MTX group(received MTX injections),and MTX+probiotics group(received MTX injections along with a diet containing probiotics).Morphological and histological changes,the level of mitochondrial DNA(mtDNA)damage,the level of lipid peroxidation products,and gene expression in the mice’s small intestine were ***:We demonstrated that intraperitoneal MTX injections significantly increased mtDNA damage in the liver(p<0.001),small intestine(p<0.001),and blood of mice(p<0.01).MTX elevated the quantity of lipid peroxidation products in the liver and small intestine,indicating its strong prooxidative *** induced structural changes in the mice’s intestines,characterized by leukocytic infiltration of *** therapy in mice partially mitigated the morphological and histological changes in the small intestine induced by MTX,reduced oxidative stress,and promoted increased expression of quinone oxidoreductase 1(Nqo1),which participates in both cell protection against oxidative stress and drug/xenobiotic *** prevented the upregulation of the proinflammatory cytokine IL-1b in the small intestine and induced increased expression of genes associated with the Nuclear factor erythroid 2-related factor 2/Antioxidant response element(Nrf2/ARE)pathway,an important mechanism of cell ***:Probiotics can be considered an effective approach to reducing the toxicity of MTX during psoriasis or cancer treatment.

The ameliorative effects of ginger against heat-induced oxidative stress and mitochondrial dysfunction in Caenorhabditis elegans

Ginger， rich in gingerols and shogaols， exhibits multiple biological properties. However， the mechanisms underlying its thermotolerance remain unclear. The study employed network pharmacology and experimental validation in Caenorhabditis elegans to investigate how gingerol-related compounds within ginger extract (GE) mitigated damage caused by heat stress (HS). A total of 18 types of gingerol analogues were identified in GE， among which 6-， 8-， and 10-gingerol， as well as 6-， 8-， and 10-shogaol were quantified. Collectively， these six compounds accounted for 54.4% of the total composition. Supplementation with 15 μg/mL GE significantly extended heat-stress lifespan by 20.30%， while the combination of the six major gingerols and shogaols at the same concentration prolonged lifespan by 18.93%. Additionally， pretreatment with GE and the combination alleviated HS-induced oxidative damage by eliminating reactive oxygen species (ROS) and upregulating antioxidant enzymes. Network pharmacology analysis suggested that the MAPK pathway may play a crucial role in thermotolerance. Experimental findings confirmed that ginger attenuated oxidative damage through the activation of SKN-1/Nrf2 and DAF-16/FOXO via the MAPK pathway. Moreover， GE stabilized mitochondrial membrane potential and restored ATP levels， thus preserving mitochondrial function during heat exposure. Further investigations using molecular docking and molecular dynamics simulations revealed that shogaols， with more stable binding affinities for KEAP1 protein， exhibited more potent effects than gingerols in prolonging lifespan and reducing ROS levels under HS conditions. In short， gingerol analogues from ginger conferred thermal resistance to nematodes by mitigating oxidative damage and mitochondrial dysfunction.

The role of copper element has been an increasingly relevant topic in recent years in the fields of human and animal health, for both the study of new drugs and innovative food and feed supplements. This metal plays an important role in the central nervous system, where it is associated with glutamatergic signaling, and it is widely involved in inflammatory processes. Thus, diseases involving copper(Ⅱ) dyshomeostasis often have neurological symptoms, as exemplified by Alzheimer's and other diseases(such as Parkinson's and Wilson's diseases). Moreover, imbalanced copper ion concentrations have also been associated with diabetes and certain types of cancer, including glioma. In this paper, we propose a comprehensive overview of recent results that show the importance of these metal ions in several pathologies, mainly Alzheimer's disease, through the lens of the development and use of copper chelators as research compounds and potential therapeutics if included in multi-target hybrid drugs. Seeing how copper homeostasis is important for the well-being of animals as well as humans, we shortly describe the state of the art regarding the effects of copper and its chelators in agriculture, livestock rearing, and aquaculture, as ingredients for the formulation of feed supplements as well as to prevent the effects of pollution on animal productions.

Structural analysis of a neutral polysaccharide ATP-1 from the red seaweed Asparagopsis taxiformis and its immunomodulatory effects on RAW264.7 macrophages

As a seaweed supplement， the powdered， entire herb of Asparagopsis taxiformis can effectively promote immune response in animals. However， the active factors responsible for this effect are currently unknown. A novel polysaccharide， ATP-1， was isolated and purified from the red alga A. taxiformis， and its structural and immunomodulatory properties were determined. ATP-1 is a neutral polysaccharide with a molecular weight of 307.978 kDa and is primarily composed of galactose (94.07%)， glucose (1.27%)， and xylose (4.66%). Scanning electron microscopy and atomic force microscopy revealed that ATP-1 had an amorphous structure and was composed of molecular chain entanglement and intramolecular hydrogen bonding interactions. The microstructure of ATP-1 exhibited asymmetric sheet–like cracks and pores. Results from immunomodulatory activity showed that ATP-1 promoted inducible nitric oxide (NO) synthase and cyclooxygenase (COX)-2 protein expression in RAW264.7 macrophages， improved macrophage phagocytic ability， and stimulated cytokine production (NO， tumor necrosis factor [TNF]-α， and interleukin [IL]-6). Quantitative proteome analysis identified 178 differentially expressed proteins (70 upregulated and 108 downregulated) that were involved in various molecular pathways， among which， TNF was one of the 20 most common and significant signaling pathways in inflammation and immunoregulation. Western blotting indicated that ATP-1 significantly upregulated the expression of IKKα， IKKβ， COX-2， and TNF-α proteins in the TNF pathway. In contrast， ATP-1 downregulated the expression of IκBα， Fas， and AIPI. Results from molecular docking suggested that hydrogen bonding predominantly contributed to the affinity between TNF-α and galactose， glucose， and xylose. These findings indicated that ATP-1 may play an immunomodulatory role by activating the TNF signaling pathway. Development and Reproduction of Sweet Potato Whitefly Bemisia tabaci (Homoptera;Aleyroididae) on Four Vegetable Crops in Southern Sierra Leone

Bemisia tabaci is a polyphagous herbivore that feeds on a wide range of horticultural and ornamental crops cultivated under diverse ecological zones. In Sierra Leone, B. tabaci is found to infest a wide range of vegetable crops by directly feeding on phloem sap thereby inducing physiological disorders, and also serve as a vector to gemini viruses. Invariably the destructive feeding of B. tabaci affects the productivity and aesthetic values of vegetables and other horticultural crops and hence is considered a serious economic pest. A bioassay experiment was carried out by rearing B. tabaci populations on four vegetable crops under controlled laboratory conditions to determine its life table and demographic parameters. Results showed that the intrinsic rate of growth which measures the population size and growth pattern was highest for populations reared on tomato crops with the following values: rm 0.145 female female−1 day−1, the gross reproduction rate (Ro), and finite growth rate λ were highest for population reared on tomato, correspondingly the development period from egg-adult emergence was shortest with a value of 26 d. Conversely, the computed demographical parameters rm, λ and Ro for the population reared on sweet pepper were 0.106 female female−1 day−1 respectively, with a corresponding development period egg-adult emergence as 36d. The computed biological parameters for okra and garden egg varied with intermediary values between tomato and pepper host materials. The survivorship rates were quite significant for the smaller instars (Instars 1-III) with over 80% surviving to pre-pupa and pupa stage for the populations reared for all the test materials. High mortality was noticed for the pre-pupa and pupa stages as their survival rates were significantly low compared to the high survival rates of the smaller instars. In China, liver transplantation is an important discipline in the field of organ transplantation. China Liver Transplant Registry (CLTR) is a scientific project that has been set up to advance surgical techniques and procedures and to improve both short-and long-term post-transplant follow-up and outcome of the liver recipients. CLTR also serves as a robust data support platform for the National Liver Transplant quality Control Center in the quest to upscale its quality control protocols. The mission of CLTR is to register all liver transplantation activities in the mainland of China and to conduct scientific analyses of the collected data. The huge number of compiled cases and the scientific research conducted over the past decade based on this database drastically revolutionized the clinical practice in the country. All CLTR activities and projects will be a guarantee to foster progresses of liver transplantation in China in a more scientific way, to standardize the systematic care in the field of liver transplantation.

Childhood Environmental Unpredictability and Psychopathy:Mediating Roles of Insecure Attachment and Life History Strategy

Background:Childhood environmental unpredictability(CEU)is crucial to personal development,profoundly influence physical and psychological growth particularly,and psychopathy is recognized as a dark personality *** aim of this study is to investigate the effects of insecure attachment and life history strategy(LHS)on the relationship between CEU and psychopathy by establishing a sequential mediation model based on evolutionary life history theory and cognitive-affective personality ***:A group of 532 undergraduates completed the measurements for CEU,insecure attachment,LHS,and psychopathy,and Bootstrap mediation test through SPSS 25.0 and PROCESS macro tool was used to examine the relationship among these ***:The results revealed positive relations between CEU and insecure attachment(β=0.87,p<0.001),CEU and LHS(β=0.11,p<0.001),and CEU and psychopathy(β=0.14,p<0.001).Insecure attachment was also positively associated with LHS(β=0.62,p<0.001)and psychopathy(β=0.04,p<0.001).In addition,LHS was significantly associated with psychopathy(β=0.08,p<0.001).The relationship between CEU and psychopathy is mediated independently and sequentially by insecure attachment and ***:The current research shows the relationship between CEU and psychopathy,as well as the mechanism of insecure attachment and life history strategy as mediators.
