Alzheimer's disease(AD)is the most common cause of dementia,costing about 1%of the global *** of clinical trials targeting amyloid-βprotein(Aβ),a key trigger of AD,have been explained by drug inefficiency regardless of the mechanisms of amyloid neurotoxicity,which are very difficult to address by available ***,we combine two imaging modalities that stand at opposite ends of the electromagnetic spectrum,and therefore,can be used as complementary tools to assess structural and chemical information directly in a single *** label-free super-resolution microspectroscopy for sub-cellular imaging based on novel optical photothermal infrared(O-PTIR)and synchrotron-based X-ray fluorescence(S-XRF)nano-imaging techniques,we capture elemental distribution and fibrillary forms of amyloid-βproteins in the same neurons at an unprecedented *** results reveal that in primary AD-like neurons,iron clusters co-localize with elevated amyloidβ-sheet structures and oxidized ***,our O-PTIR/S-XRF results motivate using high-resolution multimodal microspectroscopic approaches to understand the role of molecular structures and trace elements within a single neuronal cell.

Exploration behaviour and behavioural flexibility in orb-web spiders： A review

Orb-web spiders and their webs constitute an ideal model system in which to study behavioural flexibility and spatial cognition in invertebrates due to the easily quantifiable nature of the orb web. A large number of studies demonstrate how spiders are able to modify the geometry of their webs in response to a range of different conditions including the ability to adapt their webs to spatial constraints. However, the mechanisms behind this impressive web-building flexibility in these cognitively limited animals remain poorly explored. One possible mechanism though may be spatial learning during the spiders＇ exploration of their immediate surroundings. This review discusses the importance of exploration behaviour, the reliance on simple behavioural rules, and the use of already laid threads as guidelines for web-building in orb-web spiders. The focus is on the spiders＇ ability to detect and adapt their webs to space limitations and other spatial disruptions. I will also review the few published studies on how spatial information is gathered during the exploration phase and discuss the possibility of the use of ＇cognitive map＇-like processes in spiders. Finally, the review provides suggestions for designing experimental studies to shed light on whether spiders gather metric information during the site exploration （cognitive map hypothesis） or rely on more simple binary information in combination with previously laid threads to build their webs （stigmergy hypothesis）.

Tubulin detyrosination promotes human trophoblast syncytium formation

Human trophoblast syncytialization is one of the most important yet least understood events during placental *** this study,we found that detyrosinated a-tubulin(detyr-a-tub),which is negatively regulated by tubulin tyrosine Ugase(TTL),was elevated during human placental cytotrophoblast ***,relatively high expression of TTL protein was observed in first-trimester human placental cytotrophoblast cells,but fusing trophoblast cells exhibited much lower levels of T T L Notably,fusion of preeclamptic cytotrophoblast cells was compromised but could be partially rescued by knockdown of TTL ***,chronic downregulation of TTL in trophoblast cells resulted in significantly elevated expression of *** of detyr-a-tub thus contributed to the cell surface localization of the fusogenic protein Syncytin-2 and the gap junction protein Connexin 43(Cx43),which in turn promoted successful fusion between trophoblast *** together,the results suggest that tubulin detyrosination plays an essential role in human trophoblast fusogenic protein aggregation and *** tubulin detyrosination leads to defects in syncytialization and potentially to the onset of preeclampsia.

Thermochemical studies on complex of [Sm(o-NBA)_3phen]_2

A ternary complex [Sm(o-NBA)3phen]2 (o-NBA: o-Nitrobenzoate; phen: 1,10-phenanthroline) was synthesized and characterized by elemental analysis, IR, molar conductance, and thermogravimetric analysis. The dissolution enthalpies of SmCl3·6H2O(s), o-HNBA(s) and phen·H2O(s) in mixed solvent (VHCl :VDMF :VDMSO=2:2:1) were determined by calorimetry at 298.15 K. Protective effect of Eleteria cardamomum(L.) Maton against Pan masala induced damage in lung of male Swiss mice

Objective:To study the potential ameliorating properties of cardamom Elettaria cardamomum (***) *** against pan masala induced damage in lung of male Swiss mice. Methods:The experimental animals were divided into 3 groups(control,pan masala treated group and pan masala with cardamom treated group) to evaluate pan masala *** observations were substantiated with profound changes in the lung tissue as revealed in the histologic and transmission electron microscopic ***:Lung of pan masala treated group showed adenocarcinoma,edema,and inflammation with increased activity of acid phosphatase,alkaline phosphatase,and lactate *** deleterious effects were seen to be less in cardamom treated group and the enzymatic activity also decreased significantly (P<0.05)\ in the ameliorating ***:Thus,the present experiment exciting results are observed when cardamom is supplemented with pan masala,or when given alone.

Microwave-induced Apoptosis and Cytotoxicity of NK Cells through ERK1/2 Signaling

Objective To investigate microwave-induced morphological and functional injury of natural killer（NK） cells and uncover their mechanisms. Methods NK-92 cells were exposed to 10, 30, and 50 m W/cm^2 microwaves for 5 min. Ultrastructural changes, cellular apoptosis and cell cycle regulation were detected at 1 h and 24 h after exposure. Cytotoxic activity was assayed at 1 h after exposure, while perforin and NKG2 D expression were detected at 1 h, 6 h, and 12 h after exposure. To clarify the mechanisms, phosphorylated ERK（p-ERK） was detected at 1 h after exposure. Moreover, microwave-induced cellular apoptosis and cell cycle regulation were analyzed after blockade of ERK signaling by using U0126. Results Microwave-induced morphological and ultrastructural injury, dose-dependent apoptosis（P 〈 0.001） and cell cycle arrest（P 〈 0.001） were detected at 1 h after microwave exposure. Moreover, significant apoptosis was still detected at 24 h after 50 m W/cm^2 microwave exposure（P 〈 0.01）. In the 30 m W/cm^2 microwave exposure model, microwaves impaired the cytotoxic activity of NK-92 cells at 1 h and down regulated perforin protein both at 1 h and 6 h after exposure（P 〈 0.05）. Furthermore, p-ERK was down regulated at 1 h after exposure（P 〈 0.05）, while ERK blockade significantly promoted microwave-induced apoptosis（P 〈 0.05） and downregulation of perforin（P 〈 0.01）. Conclusion Microwave dose-dependently induced morphological and functional injury in NK-92 cells, possibly through ERK-mediated regulation of apoptosis and perforin expression.

Two-step production of monoamines in monoenzymatic cells in the spinal cord:a different control strategy of neurotransmitter supply?

Monoamine neurotransmitters play an important role in the modulation of sensory, motor and autonomic functions in the spinal cord. Although traditionally it is believed that in mammalian spinal cord, monoamine neurotransmitters mainly originate from the brain, accumulating evidence indicates that especially when the spinal cord is injured, they can also be produced in the spinal cord. In this review, I will present evidence for a possible pathway for two-step synthesis of dopamine and serotonin in the spinal cord. Published data from different sources and unpublished data from my own ongoing projects indicate that monoenzymatic cells expressing aromatic L-amino acid decarboxylase（AADC）, tyrosine hydroxylase（TH） or tryptophan hydroxylase（TPH） are present in the spinal cord and that these TH and THP cells often lie in close proximity to AADC cells. Prompted by the above evidence, I hypothesize that dopamine and serotonin could be synthesized sequentially in two monoenzymatic cells in the spinal cord via a TH-AADC and a TPH-AADC cascade respectively. The monoamines synthesized through this pathway may compensate for lost neurotransmitters following spinal cord injury and also may play specific roles in the recovery of sensory, motor and autonomic functions.

Empirical antibiotic treatment with piperacillin-tazobactam in Patients with microbiologically-documented biliary tract infections

AIM: To report our experience with empiric antimicrobial monotherapy (piperacillin/tazobactam, of which no data are available in such specific circumstances) in microbiologically-documented infections in patients with benign and malignant conditions of the biliary ***: Twenty-three patients, 10 with benign and 13 with malignant conditions affecting the biliary tree and microbiologically-documented infections were recruited and the efficacy of empirical antibiotic therapy was ***: The two groups featured similar demographic and clinical data. Overall, the infective episodes were most due to Gram negative agents, more than 60% of such episodes (mostly in malignant conditions) were preceded by invasive instrumental maneuvers. Empirical antibiotic therapy with a single agent (piperacillin/tazobactam) was effective in more than 80% of cases. No deaths were reported following infections. CONCLUSION: An empiric therapeutic approach with piperacillin/tazobactam is highly effective in biliary tract infections due to benign or malignant conditions.

Current status of the adjuvant therapy in uterine sarcoma: A literature review

Uterine sarcomas (US) are rare mesenchymal tumours accounting approximately for 3%–7% of all uterine cancers. Histologically, US are classified into mesenchymal tumours or mixed epithelial and mesenchymal tumours. The group of mesenchymal tumours includes uterine leiomyosarcoma (uLMS, 65% of cases), endometrial stromal sarcoma (ESS, 21%)– traditionally divided into low grade (LG-ESS) and high grade–undifferentiated uterine sarcoma (5%) and other rare subtypes such as alveolar or embryonal rhabdomyosarcoma. Despite the fact that several drugs demonstrated clinical activity in advanced or metastatic settings, the role of postoperative therapy in US remains controversial. In this review, we have summarised the current state of the art, including the chief trials on adjuvant treatment modalities in US, especially focusing on uLMS, LG-ESS and other rare histotypes.

High prevalence of nonalcoholic fatty liver in patients with idiopathic venous thromboembolism

AIM:To assess the prevalence of nonalcoholic fatty liver disease (NAFLD) in patients with idiopathic venous thromboembolism (VTE). METHODS:In a case-control study,after excluding subjects with well-consolidated risk factors for VTE,idiopathic VTE was documented in 138 consecutive patients who were referred to our department. Two hundred and seventy-six healthy sex/age/body-massindex-matched subjects,without any clinical/instrumental evidence of VTE,served as controls. All underwent a clinical/laboratory/ultrasound assessment for the presence of metabolic syndrome and NAFLD. RESULTS:NAFLD was detected in 112/138 cases (81%) and in 84/276 controls (30%) [risk ratio:2.7,95% confidence interval (CI):2.2-3.2,P < 0.0001]. Metabolic syndrome and smoking habit were more prevalent in patients with idiopathic VTE. The high prevalence of NAFLD in VTE was also confirmed after adjustment for inherited thrombophilia. NAFLD was clearly predicted by VTE (odds ratio:1.8,95% CI:1.2-2.7,P < 0.0001).
