Endoplasmic reticulum stress and autophagy in cerebral ischemia/reperfusion injury:PERK as a potential target for intervention

Several studies have shown that activation of unfolded protein response and endoplasmic reticulum(ER)stress plays a crucial role in severe cerebral ischemia/reperfusion *** occurs within hours after cerebral ischemia,but the relationship between ER stress and autophagy remains *** this study,we established experimental models using oxygen-glucose deprivation/reoxygenation in PC12 cells and primary neurons to simulate cerebral ischemia/reperfusion *** found that prolongation of oxygen-glucose deprivation activated the ER stress pathway protein kinase-like endoplasmic reticulum kinase(PERK)/eukaryotic translation initiation factor 2 subunit alpha(e IF2α)-activating transcription factor 4(ATF4)-C/EBP homologous protein(CHOP),increased neuronal apoptosis,and induced ***,inhibition of ER stress using inhibitors or by si RNA knockdown of the PERK gene significantly attenuated excessive autophagy and neuronal apoptosis,indicating an interaction between autophagy and ER stress and suggesting PERK as an essential target for regulating *** autophagy with chloroquine exacerbated ER stress-induced apoptosis,indicating that normal levels of autophagy play a protective role in neuronal injury following cerebral ischemia/reperfusion *** from this study indicate that cerebral ischemia/reperfusion injury can trigger neuronal ER stress and promote autophagy,and suggest that PERK is a possible target for inhibiting excessive autophagy in cerebral ischemia/reperfusion injury.

Visceral leishmaniasis(VL)(kala-azar)was most seriously prevalent in the plain regions of eight provinces/municipalities in the eastern and central parts of *** the early 1950s,the number of counties/cities endemic for VL and the number of cases in the plain regions accounted for 60%and 80%,respectively,of the total numbers in the entire *** implementing comprehensive control measures,including treatment of patients for eliminating the source of infection and spraying insecticide in endemic villages to kill sandflies,VL transmission has been brought under control in this region by the early 1960s,and no new infected cases have been found since 1983,achieving the goal of eliminating VL.

Schistosomiasis, caused by infections with Schistosoma japonicum, is one of the human parasitic diseases that remains a public health problem throughout the world＇s tropical and subtropical regions. Objective Pigs, as hosts of zoonotic Cryptosporidium species/genotypes, are domestic animals with public health significance. The present study was to characterize the infection rate and species/genotype of Cryptosporidium in pre-weaned and post-weaned pigs from Shanghai and Shaoxing, China. Methods A total of 208 fecal samples （42 from pre-weaned piglets, and 166 from post-weaned pigs） were examined by nested PCR of the 185 rRNA gene and analyzed by phylogenetic DNA fragment sequencing of secondary PCR products. Results Infection was detected in 79 samples （19/42 pre-weaned piglets, and 60/166 post-weaned pigs）. C. suis （14/79） and Cryptosporidium pig genotype II （65/79） were identified; piglets were more susceptible to the former （13/14） and post-weaned pigs to the latter （59/65）. Conclusion Infection of Cryptosporidium spp. in pigs was age-specific; piglets were more susceptible to C. suis while pigs were more susceptible to Cryptosporidium pig genotype II. These findings combined with the isolation of the two Cryptosporidium from water suggest that pigs may be a source of zoonotic Cryptosporidium water pollution. Improvements in pig feeding practices, sewage discharge, feces disposal and field worker protection are therefore important to prevent potential public health problems.

Surveillance of antimalarial drug resistance in China in the 1980s–1990s

Since the successful preparation of the microplates and the medium for field application,the resistance degree and its geographical distribution of chloroquine-resistant Plasmodium falciparum,the fluctuation of the resistance degree of *** to chloroquine,and the sensitivity of the parasite to commonly used antimalarial drugs were investigated between 1980 and 2003 by the in vitro microtest and the in vivo four-week test recommended by the World Health Organization(WHO).The results indicated that chloroquine-resistant falciparum malaria was present in all eight provinces/autonomous regions endemic for falciparum malaria in China,and the resistance was high and widely distributed in the Hainan and Yunnan *** the use of chloroquine was stopped or administered in a decreased quanity,the drug resistance gradually *** Hainan and Yunnan,*** was still highly resistant to chloroquine,amodiaquine and piperaquine,and sensitive to pyronaridine and artemisinin derivatives,but the sensitivity was gradually *** on these results,principles and therapeutic regimens for antimalarial drug use in China were formulated,the use of the antimalarials which had already developed resistance was stopped or reduced,and recommendations to use artemisinin derivatives or compound pyronaridine to promote a rational use of antimalarials and strengthen malaria control were *** results showed that malaria incidence had declined,and endemic areas of falciparum malaria have been gradually reducing since the mid-1980s.

Recombinant adenovirus containing hyper-interleukin-6 and hepatocyte growth factor ameliorates acute-on-chronic liver failure in rats

AIM: To investigate the protective efficacy of recombinant adenovirus containing hyper-interleukin-6 (Hyper-IL-6, HIL-6) and hepatocyte growth factor (HGF) (Ad-HGF-HIL-6) compared to that of recombinant adenovirus containing either HIL-6 or HGF (Ad-HIL-6 or Ad-HGF) in rats with acute-on-chronic liver failure (ACLF).METHODS: The recombinant adenoviruses containing HIL-6 and/or HGF were constructed. We established an ACLF model, and rats were randomly assigned to control, model, Ad-GFP, Ad-HIL-6, Ad-HGF or Ad-HGF-HIL-6 group. We collected serum and liver tissue samples to test pathological changes, biochemical indexes and molecular biological ***: Attenuated alanine aminotransferase, prothrombin time, high-mobility group box 1 (HMGB1), endotoxin, tumour necrosis factor (TNF)-α and interferon-γ were observed in the Ad-HGF-, Ad-HIL-6- and Ad-HGF-HIL-6-treated rats with ACLF. Likewise, reduced hepatic damage and apoptotic activity, as well as reduced HMGB1 and Bax proteins, but raised expression of Ki67 and Bcl-2 proteins and Bcl-2/Bax ratio were also observed in the Ad-HGF-, Ad-HIL-6- and Ad-HGF-HIL-6-treated rats with ACLF. More significant changes were observed in the Ad-HGF-HIL-6 treatment group without obvious side effects. Furthermore, caspase-3 at the protein level decreased in the Ad-HIL-6 and Ad-HGF-HIL-6 treatment groups, more predominantly in the latter ***: This study identifies that the protective efficacy of Ad-HGF-HIL-6 is more potent than that of Ad-HGF or Ad-HIL-6 in ACLF rats, with no significant side effects.

The present study is to determine the suitable protoscolices （PSCs） density for long-time culturing in vitro. The PSCs were divided into eight groups with different densities and the viability tests were carried out with 0.1% methylene blue staining. Then the infection ability of cultured PSCs was assessed by the mean cyst weight of mice inoculated intraperitoneally with PSCs after 8 months post-infection.

Analysis of diferentially expressed protein from primary and recurrent ovarian cancer serum

Objective:To study the value of the differentially expressed proteins from primary and recurrent ovarian cancer serum for early diagnosis of primary and recurrent ovarian ***: WCX kit(Bruker Daltonics GraBH) and matrix-assisted laser desorption/ionization time-offlight mass spectrometry(MALDI-TOF-MS]technology were used to detect serum samples from 49 patients with primary ovarian cancer and 21 patients with recurrent ***:In the mass range(Mr) from 1 000 to 12 000 Da,eight differentially expressed protein peaks were screened from primary ovarian cancer *** them,four protein peaks with Mr 1 457,1 857,2 202, 7 761 were lowly expressed and the others with Mr 2 946,5 333,5 859,5 901 were highly expressed. Ten differentially expressed protein peaks were screened from recurrent ovarian cancer serum. Among them.1 944,1 980,2 080,2 661,2 993,4 450,4 659,5 359 Da protein expressions were increased significantly,and 1897,7868 Da protein expressions were decreased *** pattern of primary ovarian cancer was applied to 8 early-stage ovarian cancer serum samples, and 7 serum samples were successfully predicted with the accuracy of 87.5%.The pattern of recurrent ovarian cancer was applied to 9 without pelvic:or abdominal mass recurrent ovarian cancer serum samples,and 8 serum samples were successfully predicled with the accuracy of 88.9%.Conclusions:Combination of MALDI-TOF-MS and WCX kit technology can directly screen the diferrential expressed protein from primary and recurrent ovarian cancer *** have clinical significance for enhancement of sensitivity and specificity of ovarian cancer diagnosis.
