Neurochemical features of endomorphin-2-containing neurons in the submucosal plexus of the rat colon

AIM:To investigate the distribution and neurochemical phenotype of endomorphin-2(EM-2)-containing neurons in the submucosal plexus of the rat colon.METHODS:The mid-colons between the right and left flexures were removed from rats,and transferred into Kreb's solution. For whole-mount preparations,the mucosal,outer longitudinal muscle and inner circularmuscle layers of the tissues were separated from the submucosal layer attached to the submucosal plexus. The whole-mount preparations from each rat mid-colon were mounted onto seven gelatin-coated glass slides,and processed for immunofluorescence histochemical double-staining of EM-2 with calcitonin gene-related peptide(CGRP),choline acetyltransferase(Ch AT),nitric oxide synthetase(NOS),neuron-specific enolase(NSE),substance P(SP) and vasoactive intestinal peptide(VIP). After staining,all the fluorescence-labeled sections were observed with a confocal laser scanning microscope. To estimate the extent of the co-localization of EM-2 with CGRP,Ch AT,NOS,NSE,SP and VIP,ganglia,which have a clear boundary and neuronal cell outline,were randomly selected from each specimen for this analysis. RESULTS:In the submucosal plexus of the mid-colon,many EM-2-immunoreactive(IR) and NSE-IR neuronal cell bodies were found in the submucosal plexus of the rat mid-colon. Approximately 6 ± 4.2 EM-2-IR neurons aggregated within each ganglion and a few EM-2-IR neurons were also found outside the ganglia. The EM-2-IR neurons were also immunopositive for Ch AT,SP,VIP or NOS. EM-2-IR nerve fibers coursed near Ch AT-IR neurons,and some of these fibers were even distributed around Ch AT-IR neuronal cell bodies. Some EM-2-IR neuronal cell bodies were surrounded by SP-IR nerve fibers,but many long processes connecting adjacent ganglia were negative for EM-2 immunostaining. Long VIP-IR processes with many branches coursed through the ganglia and surrounded the EM-2-IR neurons. Lipid rafts participate in aberrant degradative autophagic-lysosomal pathway of amyloid-beta peptide in Alzheimer's disease

Amyloid-beta peptide is the main component of amyloid plaques, which are found in Alzhei- mer＇s disease. The generation and deposition of amyloid-beta is one of the crucial factors for the onset and progression of Alzheimer＇s disease. Lipid rafts are glycolipid-rich liquid domains of the plasma membrane, where certain types of protein tend to aggregate and intercalate. Lipid rafts are involved in the generation of amyloid-beta oligomers and the formation of amyloid-beta peptides. In this paper, we review the mechanism by which lipid rafts disturb the aberrant deg- radative autophagic-lysosomal pathway of amyloid-beta, which plays an important role in the pathological process of Alzheimer＇s disease. Moreover, we describe this mechanism from the view of the Two-system Theory of fasciology and thus, suggest that lipid rafts may be a new target of Alzheimer＇s disease treatment.

Neural stem cells over-expressing brain-derived neurotrophic factor promote neuronal survival and cytoskeletal protein expression in traumatic brain injury sites

Cytoskeletal proteins are involved in neuronal survival.Brain-derived neurotrophic factor can increase expression of cytoskeletal proteins during regeneration after axonal injury.However,the effect of neural stem cells genetically modified by brain-derived neurotrophic factor transplantation on neuronal survival in the injury site still remains unclear.To examine this,we established a rat model of traumatic brain injury by controlled cortical impact.At 72 hours after injury,2 × 10~7 cells/m L neural stem cells overexpressing brain-derived neurotrophic factor or naive neural stem cells（3 m L） were injected into the injured cortex.At 1–3 weeks after transplantation,expression of neurofilament 200,microtubule-associated protein 2,actin,calmodulin,and beta-catenin were remarkably increased in the injury sites.These findings confirm that brain-derived neurotrophic factor-transfected neural stem cells contribute to neuronal survival,growth,and differentiation in the injury sites.The underlying mechanisms may be associated with increased expression of cytoskeletal proteins and the Wnt/β-catenin signaling pathway.

Honokiol attenuates mitochondrial fission and cell apoptosis by activating Sirt3 in intracerebral hemorrhage

Background:Sirtuin-3(Sirt3)has been documented to protect against mitochondrial dysfunction and apoptosis.Honokiol(HKL)is a Sirt3 pharmacological activator with reported neuroprotective effects in multiple neurological disorders.The present study aimed to explore the neuroprotective effects of HKL and the role of Sirt3 following intracerebral hemorrhage(ICH).Methods:An in vivo ICH model in rats was established by injecting autologous blood into the right basal ganglia.PC12 cells were stimulated with hemin.For the in vivo investigation,the modified Neurological Severity Scores and the Morris water maze test were performed to assess neurological deficits.Hematoxylin-Eosin and Terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling staining were employed to evaluate the histopathology and apoptosis.Immunohistochemical staining was used to investigate the expression of Sirt3.Adenosine triphosphate(ATP)levels were quantified to assess mitochondrial dysfunction.Cell counting kit-8,lactate dehydrogenase assay,and flow cytometry were used to analyze cell vitality and apoptosis in vitro.Immunofluorescence staining was performed to observe mitochondrial morphology and dynamin-related protein 1(Drp1)localization to mitochondria.Western blot was applied to quantify the expression of Sirt3,Bax,Bcl-2,cleaved-caspase-3,Drp1,phosphorylation of Drp1 at serine-616,and phosphorylation of Drp1 at serine-637 in vivo and in vitro.Results:HKL treatment alleviated neurological deficits,attenuated the histopathological damage and cell apoptosis,and restored the decreased ATP levels in ICH rats.HKL improved cell survival rate,reduced cell apoptosis,and inhibited mitochondrial fission in PC12 cells.Moreover,both in vivo and in vitro models showed increased phosphorylation of Drp1 at Ser616,and reduced phosphorylation of Drp1 at Ser637.Meanwhile,immunofluorescence co-localization analysis revealed that hemin increased the overlap of Drp1 and mitochondria in PC12 cells.The phosphorylation and mitochondrial translocation of Drp1

Protective effect of basic fibroblast growth factor on laser induced retinopathy

AIM:To investigate the side effects of the commonly used laser treatment along with testing the neuroprotective effect of bFGF on a potential retinal impairment.METHODS:To do this,30 chinchilla pigmented adult male rabbits were divided into the control and experimental groups.The control and experimental groups underwent both laser application and bFGF treatment.The retinal tissue impairment and its renewal rate were tested under the light and electron microscopical levels.RESULTS:The focal laser application on rabbit eyes caused morphological alterations both in the application region and in the neighbouring areas.In the damaged areas,the outer nuclear layer of the neural retina was almost disappeared,retina pigment epithelium was interrupted,the retina pigment epithelium migrated intraretinally,and the damaged region along with neighbouring areas seemed to be not separated.bFGF application just after the laser photocoagulation,revealed better results in application areas.CONCLUSION:It could be suggested that the bFGF application following laser photocoagulation might have protective,repairing and wound healing effects on the retina.

Ultrastructural view of colon anastomosis under propolis effect by transmission electron microscopy

AIM: To evaluate the effect of propolis administration on the healing of colon anastomosis with light and transmission electron microscopes. METHODS: Forty-eight Wistar-Albino female rats were divided into two groups and had colon resection and anastomosis. In group Ⅰ, rats were fed with standard rat chow pre- and postoperatively. The rats in group Ⅱ were fed with standard rat chow and began receiving oral supplementation of propolis 100 mg/kg per day beginning 7 d before the operation and continued until they were sacrificed. Rats were sacrificed 1, 3, 7 and 14 d after operation, and anastomotic bursting pressures measured. After the resection of anastomotic segments, histopathological examination was performed with light and transmission electron microscopes by two blinded histologists and photographed. RESULTS: The colonic bursting pressures of the propolis group were statistically significantly better than the control group. Ultrastructural histopathological analysis of the colon anastomosis revealed that propolis accelerated the phases of the healing process and stimulated mature granulation tissue formation and collagen synthesis of fibroblasts. CONCLUSION: Bursting pressure measurements and ultra structural histopathological evaluation showedthat administration of propolis accelerated the healing of colon anastomosis following surgical excision.

Angiogenic Effect of Intercellular Adhesion Molecule-1

In order to investigate the angiogenic effect of intercellular adhesion molecule-1 （ICAM-1）, two parts of experiment were performed. Chick embryo chorioallantoic membrane （CAM） assay was used for in vivo angiogenic research. The chick embryos were divided into 4 groups： ICAM-1 group （divided into 3 subgroups, Ⅰ, Ⅱ and Ⅲ） for screening the angiogenic effect of ICAM-1 by adding different concentrations of ICAM-1 （0.1, 0.2 and 0.3 μg/μL） 5 μL into the chick embryo CAMs on the day 10 after incubation for every subgroup; Anti-ICAM-1 group A （divided into 2 subgroups, Ⅰ and Ⅱ） by adding different concentrations of Anti-ICAM-1 （1：100, 1：50） 5 μL into the chick embryo CAMs on the day 10 after incubation for every subgroup to evaluate the effect of ICAM-1 on the survival of microvessels through observing whether Anti-ICAM-1 could induce involution of the microvessels on CAMs; Anti-ICAM-1 group B （divided into 2 subgroups, Ⅰ and Ⅱ ） by adding different concentrations of Anti-ICAM-1 （1：100, 1：50） 5 μL into the chick embryo CAMs on the day 6 after incubation for every subgroup to evaluate whether ICAM-1 involved in embryonic angiogenesis through observing the growth of microvessels on CAMs; Control group： ICAM-1 or Anti-ICAM-1 was substituted by PBS 5 μL on the day 10 or day 6 after incubation. Three days later, the CAMs were photographed in vivo, excised, sectioned and the number of microvessels was counted. In ICAM-1 group, there was increased number of microvessels arranged radially with ＂spoked-wheel＂ pattern around the gelatin sponges. The new microvessels growing perpendicularly to gelatin sponges were observed. The number of the microvessels growing in the CAM mesenchymes around the sponges in 3 subgroups was higher than that in control group （P〈0.01）, however, there was no significant difference among the 3 subgroups （P〉0.05）. In anti-ICAM-1 group A, the radially arranged microvessels were very unclear around the sponges contrast to that of ICAM-1 group. Few new microvessels were detected in the center of the sponges

Whether embryonic development is affected by the maternal gut dysbacteriosis still remains ambiguous.In this study,firstly we show elevated level of lipopolysaccharides (LPS) in a gut micro biota dysbiosis mouse model.Secondly, we demonstrate that the cardiogenesis, neurogenesis and somitogenesis were inhibited in both gut microbiota dysbiosis mice and LPS exposed chicken embryos.

Angelica sinensis polysaccharides ameliorate 5-flourouracil-induced bone marrow stromal cell proliferation inhibition via regulating Wnt/β-catenin signaling

Chemotherapy may cause cellular oxidative stress to bone marrow.Oxidative damage of bone marrow hematopoietic microenvironment is closely related to chronic myelosuppression after chemotherapeutic treatment.Angelica sinensis polysaccharides(ASP)are major effective ingredients of traditional Chinese medicine Angelica with multi-target anti-oxidative stress features.In the current study,we investigated the protective roles and mechanisms of ASP on chemotherapy-induced bone marrow stromal cell(BMSC)damage.The human bone marrow stromal cell line HS-5 cells were divided into control group,5-FU group,5-FU+ASP group,and 5-FU+LiCl group to investigate the mechanism of ASP to alleviate 5-FU-induced BMSC proliferation inhibition.The results showed that 5-FU inhibits the growth of HS-5 cells in a time and dose-dependent manner;however,ASP partially counteracted the 5-FU-induced decrease in cell viability,whereas Wnt signaling inhibitor Dkk1 antagonized the effect of ASP on HS-5 cells.ASP reversed the decrease in total cytoplasmicβ-catenin,p-GSK-3β,and CyclinD1 following 5-FU treatment and modulated nuclear expression ofβ-catenin,Lef-1,and C-myc proteins.Furthermore,ASP also enhanced the antioxidant capacity of cells and reduced 5-FU-induced oxidative stress,attenuated FoxO1 expression,thus weakened its downstream apoptosis-related proteins and G0/G1 checkpoint-associated p27^(Kip1) expression to alleviate 5-FU-induced apoptosis and to promote cell cycle progression.All the results above suggest that the protective role of ASP in 5-FU-treated BMSCs proliferation for the chemotherapy may be related to its activating Wnt/β-catenin signaling and keeping homeostasis betweenβ-catenin and FoxO1 under oxidative stress.The study provides a potential therapeutic strategy for alleviating chemotherapeutic damage on BMSCs.

Pregnancy with diabetes mellitus (PGDM) has been known to affect the embryonic development of various systems.No doubt that it has a negative impact on the respiratory system,but the specific pathogenesis is not clear.In this study,firstly we demonstrated that the fetuses had immature lung development in PGDM,including pulmonary epithelial cell decreased and fibrous tissue increased.
