Regulatory Effect of Electroacupuncture on Ceramide Galactosyltransferase Expression in Sciatic Nerve of Experimental Diabetic Rats

Objective: To observe the change of ceramide galactosyltransferase(CGT) expression in sciatic nerve of experimental diabetic rats after electroacupuncture(EA) treatment. Methods: The rat model of diabetic peripheral neuropathy(DPN) was established with Streptozotocin(STZ). Twenty-eight rats were randomly divided into a normal group, a model group, an EA acupoint group, and an EA non-acupoint group, with 7 rats in each group. acupuncture on points Shenshu(BL 23) and Zusanli(ST 36) was performed in the EA group using EA electronic device with continuous wave, 2 Hz, 20 min every time, once every other day for 12 times. acupuncture(on the tip of rat tail) in EA non-acupoint group was performed with the same electronic parameters and time. The model group and the normal group were not given any treatment, except the same fixation as that in the EA acupoint group. After 12 treatments, the albumen and mRNA expressions of CGT of sciatic nerve in rats from each group were measured with real-time polymerase chain reaction(RT-PCR), fluorescence quantitative PCR(FQ-PCR) and immunohistochemical methods. Results: After modeling, the CGT expression was significantly lower than that in the normal group(P<0.01); however, after 12 EA treatments, the CGT positive cells in the EA acupoint group were up-regulated and significantly higher than those in the EA non-acupoint group and the model group(P<0.01). The CGT mRNA expression of the rats after modeling was also higher than that of the normal rats(P<0.01), and the expression in the EA acupoint group was significantly lower than those in the model group and the EA non-acupoint group(P<0.01, P<0.05). Conclusion: EA treatment with acupoints displays some therapeutic or preventive effect in the recovery of DPN via reversing the abnormal expression of CGT mRNA in sciatic nerves of experimental diabetic neuropathy rats, and it is worthy of further study.

OBJECTIVE: To evaluate the curative effect of Traditional chinese medicine(TCM) multi-channel interventional therapy on women with Assisted Reproductive Technology(ART) failure, to compare the curative effect of the dual therapy and triple therapy on women with ART Failure, and to choose the best TCM interventional therapeutic ***: The 95 cases with ART Failure from West China second University Hospital of Sichuan University meeting the inclusion criteria were randomly divided into three groups:dual therapy group(31 cases), triple therapy group(33 cases)and control group(31 cases). According to the intervene treatment of in vitro Fertilization and Embryo Transfer(IVF-ET) long cycle scheme, the control group wait naturally for 3 months before IVFET. The dual therapy group take TCM prescriptionⅡ of cultivating emotion and assisting reproduction and auricular acupoint therapy for 3 months before IVF-ET, then Western medicine treatment progestin supporting as well as auricular application and Antai Recipe after IVF-ET *** triple therapy group take TCM prescription Ⅱof cultivated emotion and assisted reproduction,auricular acupoint therapy and retention enema of TCM, and combination treatment the same as dual therapy group after transplantation. The natural pregnancy number, the period condition of secondary IVF-ET and the improvement of the kidney deficiency, liver depression and blood stasis syndrome among those three groups were ***: It was showed from analysis in 95 cases with ART failure that the number of natural preg-nancy was as followings: 3 patients from the dual therapy group, 10 patients from the triple therapy group, and no patient from the control group. BACKGROUND Carotid blowout syndrome(CBS)refers to rupture of the extracranial carotid artery and its branches;as a severe complication,it usually occurs after surgery or radiotherapy for malignant tumours of the head and *** present a case of CBS caused by chronic infection of the external carotid artery(ECA).In this case,we did not find any evidence of head and neck *** SUMMARY A 42-year-old man was referred to the Emergency department with a complaint of a lump found on the left side of his neck with pain and fever for 4 *** diagnosed the condition as neck infection with abscess formation based on physical examination,routine blood examination,ultrasound examination and plain computed tomography(CT)and decided to perform emergency *** the operation,30 mL of grey and smelly pus was drained from the deep surface of the sternocleidomastoid *** second day after the operation,the patient suddenly exhibited a large amount of haemoptysis and incision *** enhanced CT showed distal occlusion of the left ECA and irregular thickening of the broken ends of the artery encased in an uneven enhancement of soft tissue *** ECA occlusion and rupture were *** patient was transferred to a vascular unit for transcatheter ECA embolization and recovered *** Surgeons need to pay attention to vascular lesions caused by chronic infection that may develop into acute CBS.

Objective:The effectiveness of statins in reducing atherosclerotic calcification remains *** aim of this study was to confirm that simvastatin reduces atherosclerotic calcification and stabilizes plaque by restricting endoplasmic reticulum stress(ERS)-mediated ***:Twenty-four 8-week-old male apolipoprotein E(ApoE)-/-mice(C57BL/6J genetic background)were selected and randomly divided into model(n=12)and simvastatin(n=12)*** male C57BL/6J mice were selected as control group(n=12).The mice were adaptively fed for 2 weeks and were put on a high-fat diet *** 9 weeks,they were treated with simvastatin(20 mg/kg)or phosphate-buffered saline daily for 8 *** sinus samples were obtained from ApoE-/-and C57BL/6J mice for hematoxylin and eosin,von Kossa,alizarin Red S,terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling,and immunohistochemical staining after in vivo treatment with *** addition,mouse vascular smooth muscle cells were analyzed after exposure to simvastatin in ***:Administration of simvastatin in vivo drastically attenuated the atherosclerosis,calcification,and apoptosis,and decreased the serum levels of triglycerides,total cholesterol,low-density lipoprotein cholesterol,and high-density lipoprotein *** expression levels of glucose-regulated protein,78 kDa(GRP78),C/EBP homologous protein(CHOP),and caspase 12(CASP12)in the aortic sinus decreased in the simvastatin group compared with the model *** vitro,simvastatin or simvastatin plus ERS inhibitor(taurine)attenuated calcification and apoptosis,and reduced the expression of ERS-related proteins GRP78,CHOP,and ***:Treatment with simvastatin suppressed atherosclerotic *** effect may be mediated through the inhibition of ERS-related apoptosis.

The cochlear auditory epithelium contains two types of sound receptors,inner hair cells(IHCs)and outer hair cells(OHCs).Mouse models for labelling juvenile and adult IHCs or OHCs exist;however,labelling for embryonic and perinatal IHCs or OHCs are ***,we generated a new knock-in Fgf8^(P2A-3×GFP/+)(Fgf8^(GFP)/+)strain,in which the expression of a series of three GFP fragments is controlled by endogenous Fgf8 cis-regulatory *** confirming that GFP expression accurately reflects the expression of Fgf8,we successfully obtained both embryonic and neonatal IHCs with high purity,highlighting the power of Fgf8^(GFP)/+.Furthermore,our fate-mapping analysis revealed,unexpectedly,that IHCs are also derived from inner ear progenitors expressing Insm1,which is currently regarded as an OHC ***,besides serving as a highly favorable tool for sorting early IHCs,Fgf8^(GFP)/+will facilitate the isolation of pure early OHCs by excluding IHCs from the entire hair cell pool.

Observation on Clinical Effects of Acupotomy plus Cupping for Knee Osteoarthritis

Objective: To observe the clinical effect of acupotomy plus cupping for knee osteoarthritis(KOA). Methods: Sixty cases with KOA were randomly divided into a treatment group and a control group, 30 cases in each group. The patients in the treatment group were given acupotomy and cupping, while the patients in the control group were given injection of Sodium Hyaluronate. The index of severity for osteoarthritis(ISOA), the change of the effusion of knee joint and clinical effects were observed after treatment. Results: The total effective rate was respectively 96.7% in the treatment group and 66.7% in the control group. The difference in the clinical effects between the two groups was statistically significant(P<0.05). After treatment, ISOA scores and scores of knee effusion in the two groups were remarkably reduced than those before treatment(P<0.05). The changes of the scores of the two indexes were statistically significant(both P<0.05). Conclusion: Acupotomy plus cupping is better than injection of Sodium Hyaluronate in treatment of KOA.

Sleep deprivation reorganizes the dynamic configurations of default mode network activity during recovery sleep

Sleep deprivation causes disturbances of the neural activity, leading to the impairment of brain functions. However, the exact mechanism of sleep deprivation and how it affects the dynamics of brain activity during the recovery sleep remains unclear. In the current study, we performed sleep deprivation experiments on ten adult rats, and recorded the local field potentials from default mode network(DMN) regions during sleep before and after sleep deprivation. The DMN dynamics was assessed with the configurations of coactive micropatterns(CAMPs) using our previously proposed CAMP method. Our analysis revealed that the effects of sleep deprivation on DMN dynamics in the slow-wave sleep(SWS) state and the rapid eye-movement sleep(REM)state were disparate. Dynamic configurations of DMN activity in the SWS state were significantly impaired after sleep deprivation, with increased occurrence of low-activity CAMP and reorganized transition structure across three CAMPs. Moreover,enhanced functional connectivity and improved efficiencies in all CAMP networks were observed during the SWS state in the recovery sleep. However, there were no significant alterations in either DMN dynamics or CAMP network structures in the REM sleep state after sleep deprivation. Our results described the alterations of DMN dynamics in different sleep states after sleep deprivation, and illustrated the differential effects of sleep deprivation on two sleep states. These findings demonstrated the underlying neural mechanisms of the effects of sleep deprivation on DMN activity during sleep and increased our understanding of the physiological roles of the DMN in maintain sleep homeostasis after sleep deprivation.

Amide proton transfer (APT) magnetic resonance imaging (MRI) is an important molecularimaging technique at the protein level in tissue. Neurodegenerative diseases have a highlikelihood of causing abnormal protein accumulation in the brain, which can be detectedby APT MRI. This article briefly introduces the principles and image processing technologyof APT MRI, and reviews the current state of research on Alzheimer's disease and Parkinson's disease using this technique. Early applications of this approach in these twoneurodegenerative diseases are encouraging, which also suggests continued technicaldevelopment and larger clinical trials to gauge the value of this technique.

Objective Tongue Inspection on 142 Liver Cancer Patients with Damp-Heat Syndrome

Objective： To establish the diagnosis evidence of objective tongue inspection for liver cancer （LC） patients with damp-heat syndrome （DHS） by dynamically observing their tongue figures using modern tongue image analytic apparatus, and to explore the effect of intervention on the tongue figures. Methods： Tongue figures were collected from 142 LC patients with DHS by tongue image analytic apparatus. Red （R）, green （G） and blue （B） values were analyzed. The r and g values were calculated requesting r=R/（R＋G＋B）, g=G/（R＋G＋B）, and b=l-r-g, and scored in combination with chinese medical symptoms scale. The tongue figure and correlated scores were collected from 59 of them 3 days after transcatheter arterial chemoembolization intervention. Results： The range of objective tongue inspection of LC patients with DHS was as follows： as for tongue fur, 0.360〈r〈0.402 and 0.280〈g〈0.322; as for tongue proper, 0.404〈r〈0.470 and 0.243〈g〈0.301. The tongue figures and the average scores of quality of life, DHS, poor appetite, aggravated pain, decreased sleep quality and aggravated fever were obviously changed in the 59 LC patients with DHS after intervention, showing statistical difference when compared with before intervention （P〈0.05 or P〈0.01）. Conclusion： The range of objective tongue inspection of LC patients with DHS could be known by collecting and analyzing objective indicator of tongue figures, thus laying foundation for further studies with analysis of correlation between intervention and chinese medicine based on tongue figures.

To the Editor:Amyloidosis is caused by the extracellular deposition of insoluble beta-pleated protein fibrils in tissues and is classified according to the constituent *** cause of amyloidosis can be traced to more than 30 kinds of malfolded proteins,and accurate amyloid typing is crucial for appropriate therapy.[1]The pathological diagnosis is confirmed with positive Congo red(CR)staining under a light microscope and 7 to 12 nm branching fibrils under an electron *** identification of some amyloid precursor proteins can resort to immunofluorescence(IF)and immunochemistry(IHC);however,the accuracy depends on the selected antibodies to a large *** microdissection coupled with liquid chromatography-tandem mass spectrometry(LMD/MS)can identify these proteins without the restriction of IF and IHC,although exquisite equipment and skilled specialists are required.[2]This study aimed to establish LMD/MS analysis of renal biopsy tissue and compare it with IHC on diagnostic sensitivity.
