Succinylation modification:a potential therapeutic target in stroke

Obeticholic acid(OCA),a farnesoid X receptor(FXR)agonist with favorable effects on fatty and glucose metabolism,has been considered the leading candidate drug for nonalcoholic steatohepatitis(NASH)***,its limited effectiveness in resolving liver fibrosis and lipotoxicity-induced cell death remains a major ***,a newly recognized form of cell death characterized by uncontrolled lipid peroxidation,is involved in the progression of *** oxide(NO)is a versatile biological molecule that can degrade extracellular *** this study,we developed a PEGylated thiolated hollow mesoporous silica nanoparticles(MSN)loaded with OCA,as well as a ferroptosis inhibitor liproxsatin-1 and a NO donor S-nitrosothiol(ONL@MSN).Biochemical analyses,histology,multiplexed flow cytometry,bulk-tissue RNA sequencing,and fecal 16S ribosomal RNA sequencing were utilized to evaluate the effects of the combined nanoparticle(ONL@MSN)in a mouse NASH *** with the OCA-loaded nanoparticles(O@MSN),ONL@MSN not only protected against hepatic steatosis but also greatly ameliorated fibrosis and ***@MSN also displayed enhanced therapeutic actions on the maintenance of intrahepatic macrophages/monocytes homeostasis,inhibition of immune response/lipid peroxidation,and correction of microbiota *** findings present a promising synergistic nanotherapeutic strategy for the treatment of NASH by simultaneously targeting FXR,ferroptosis,and fibrosis.

The high incidence,mortality,and disability associated with ischemic stroke pose a significant threat to human *** intestinal microbiota significantly influences the onset,progression,and prognosis of ischemic *** flora plays a pivotal role in brain-gut *** reflection of changes in the gut and brain caused by gut microbes faciltates the investigation of early warning biomarkers and potential therapeutic targets for ischemic *** this narrative review of the relationship between gut microbiota and ischemic stroke,we primarily discuss three topics,grounded in real-world human and animal ***,we examined the relationship between ischemic stroke and intestinal microbiota and its metabolites,delineate the overall characteristics of intestinal microbiota dysregulation in ischemic stroke,and assess the potential clinical value,prevailing research controversies,and unique phenomena of intestinal microbiota metabolites such as trimethylamine N-oxide and short-chain fatty acids in ischemic ***,we explored the potential communication pathways between intestinal flora and ischemic stroke based on the brain-gut axis,encompassing metabolic pathways,immune pathways,and neural ***,we encapsulated the factors influencing the severity of ischemic stroke via intestinal flora,the pharmacological and nonpharmacological interventions that modulate intestinal flora in treating ischemic stroke,and the current research landscape of intestinal flora in the context of ischemic stroke sequelae.

The mechanism by which hyperbaric oxygen treatment alleviates spinal cord injury: genome-wide transcriptome analysis

Accumulating studies have demonstrated that hyperbaric oxygen(HBO)treatment alleviates spinal cord injury(SCI).However,the underlying mechanism by which HBO alleviates SCI remains to be *** this study,we performed genome-wide transcriptional profiling of the spinal cord between SCI mice and mice that received HBO treatment by high-throughput RNA sequencing at 1 week after *** also compared genome-wide transcriptional profiles from SCI mice and sham-operated *** found 76 differentially co-expressed genes in sham-operated mice,SCI mice,and HBO-treated SCI *** Gene Ontology and Kyoto Encyclopedia of Genes and Genomes enrichment analysis,we identified the biological characteristics of these differentially expressed genes from the perspectives of cell component,biological process,and molecular *** also found enriched functional pathways including ferroptosis,calcium signaling pathway,serotonergic synapse,hypoxia-inducible factor-1 signaling pathway,cholinergic synapse,and neuroactive ligand-receptor *** performed quantitative reverse transcription-polymerase chain reaction and validated that HBO treatment decreased the expression of Hspb1(heat shock protein beta 1),Hmox1(heme oxygenase 1),Ftl1(ferritin light polypeptide 1),Tnc(tenascin C)and Igfbp3(insulin-like growth factor binding protein 3)and increased the expression of Slc5a7(solute carrier family 5 choline transporter member 7)after *** results revealed the genome-wide transcriptional profile of the injured spinal cord after HBO *** findings contribute to a better understanding of the mechanism by which HBO treats SCI and may provide new targets for SCI intervention.

Acute carbon monoxide poisoning in Shandong, China: an observational study

Background: Carbon monoxide (CO) poisoning remains a major cause of accidental injuries and multiple studies have indicated that CO is also associated with significantly severe or long-term toxicity to the central nervous system. Given that CO poisoning causes serious morbidity and mortality, a better understanding of epidemiological features and clinical characteristics of acute CO poisoning in China is ***: We collected the clinical data of acute CO poisoning in patients between November 2019 and April 2020 across Shandong province, China and analyzed its characteristics focusing on the weekly amount and the severity of the confirmed ***: A total number of 21,088 acute CO poisoning cases were diagnosed. The overall incidence of acute CO poisoning was approximately 0.021%. On severity rankings, 63% of confirmed cases (n=13,378) were mild, 27%(n=5635) were moderate, and 10% (n=2075) were severe. Interestingly, the coastal cities had more confirmed cases than the inland/suburban areas in Shandong. Meanwhile, the number of confirmed cases was negatively correlated with the local mean daily temperature (P=0.0167).Conclusions: Mild acute CO poisoning cases accounted for the majority of all confirmed cases during the winter of 2019. In Shandong province, which is located in east China, residents of the coastal cities are more susceptible to CO poisoning than residents of inland cities.
