Background:Sapindus mukurossi extract(SME)is a kind of natural surface active ingredient with potential applications in cleansing ***,the polyphenols and pigments contained in the extract may cause color browning of the products during storage especially at elevated temperatures,affecting its high level addition in the ***:To explore a decolorization process suitable for industrialization realize the high level addition of SME and explore the potential of SME in the field of controlling sebum *** and Methods:SME was prepared by adsorbing polyphenols on the D301 resin and oxidation decoloring *** its sebum-control efficacy by SZ95 model and clinical ***:The results demonstrate that the D301 resin displays the best adsorption selectivity for polyphenols in SME,and the polyphenol adsorption ratio of D301 resin(5 wt%)can reach 83.6%;The optimal decolorization conditions are pH=7.8,temperature 73℃and decolorization time 5.7 h when H2O2 content is 6%,The prepared SME shows no obvious changes in color and retain stable during the high temperature(50°C)test period of 28 days.4μg/mL of SME decreases the lipid synthesis of SZ95 cells by 24.8%.The clinic efficacy of the shampoo containing 10%SME(by dry extract weigh)is further *** significant changes in the skin moisture content and transepidermal water loss(TEWL)are observed within four weeks after using the product,while the scalp sebum level is significantly ***:In this study,we prepared a light-colored,highly stable SME,enabled its high-level addition in cleansing and care products and found its sebum-control efficacy.

Metastases of uveal melanoma(UM)spread predominantly to the *** to low response rates to systemic therapies,liver-directed therapies(LDT)are commonly used for tumor *** impact of LDT on the response to systemic treatment is unknown.A total of 182 patients with metastatic UM treated with immune checkpoint blockade(ICB)were included in this *** were recruited from prospective skin cancer centers and the German national skin cancer registry(ADOReg)of the German Dermatologic Cooperative Oncology Group(DeCOG).Two cohorts were compared:patients with LDT(cohort A,n=78)versus those without LDT(cohort B,n=104).Data were analyzed for response to treatment,progression-free survival(PFS),and overall survival(OS).The median OS was significantly longer in cohort A than in cohort B(20.1 vs.13.8 months;P=0.0016)and a trend towards improved PFS was observed for cohort A(3.0 vs.2.5 months;P=0.054).The objective response rate to any ICB(16.7%vs.3.8%,P=0.0073)and combined ICB(14.1%vs.4.5%,P=0.017)was more favorable in cohort *** data suggest that the combination of LDT with ICB may be associated with a survival benefit and higher treatment response to ICB in patients with metastatic UM.

Enhancing the efficiency of direct reprogramming of human primary fibroblasts into dopaminergic neuron-like cells through p53 suppression

Dopaminergic(DA)neuron-like cells obtained through direct reprogramming of primary human fibroblasts offer exciting opportunities for treatment of Parkinson’s disease.A significant obstacle is the low efficiency of conversion during the reprogramming ***,we demonstrate that the suppression of p53 significantly enhances the efficiency of transcription factor-mediated conversion of human fibroblasts into functional dopaminergic *** particular,blocking p53 activity using a dominant-negative p53(p53-DN)in IMR90 cells increases the conversion efficiency by 5–20 *** induced DA neuron-like cells exhibit dopamine neuron-specific gene expression,significant dopamine uptake and production capacities,and enables symptomatic relief in a rat Parkinson’s disease *** together,our findings suggest that p53 is a critical barrier in direct reprogramming of fibroblast into dopaminergic neurons.

Chinese herbal medicine Yougui Pill reduces exogenous glucocorticoid-induced apoptosis in anterior pituitary cells

Long-term glucocorticoid use may result in sustained suppression of one or more secreted components from the hypothalamo-pituitary-adrenal axis, and often results in apoptosis. Yougui Pill（YGP）, a 10-component traditional Chinese herbal medicine, has been shown to be clinically effective for glucocorticoid-induced suppression of the hypothalamo-pituitary-adrenal axis. However, the pharmacological and molecular mechanisms remain unclear. We hypothesized that YGP would exert an anti-apoptosis effect on dexamethasone-treated anterior pituitary cells. In vivo experiments showed that YGP significantly reduced the number of apoptotic cells, down-regulated m RNA expression of cytochrome c, caspase-3, and caspase-9, and up-regulated m RNA expression of Bcl-2. These findings suggest that YGP reduced glucocorticoid-induced apoptosis in rat anterior pituitary cells by regulating the mitochondria-mediated apoptosis pathway.

Elderly patients had more severe postoperative complications after pancreatic resection:A retrospective analysis of 727 patients

AIM To examine the impact of aging on the short-term outcomes following pancreatic resection(PR) in elderly *** A retrospective cohort study using prospectively collected data was conducted at the China National cancer center. Consecutive patients who underwent PR from January 2004 to December 2015 were identifiedand included. ‘Elderly patient' was defined as ones age 65 and above. Comorbidities, clinicopathology, perioperative variables, and postoperative morbidity and mortality were compared between the elderly and young patients. Univariate and multivariate analyses were performed using the Cox proportional hazard model for severe postoperative complications(grades Ⅲb-Ⅴ).RESULTS A total of 454(63.4%) patients were < 65-yearsold and 273(36.6%) patients were ≥ 65-yearsold, respectively. Compared to patients < 65-yearsold, elderly patients had worse American Society of Anesthesiologists scores(P = 0.007) and more comorbidities(62.6% vs 32.4%, P < 0.001). Elderly patients had more severe postoperative complications(16.8% vs 9.0%, P = 0.002) and higher postoperative mortality rates(5.5% vs 0.9%, P < 0.001). In the multivariate Cox proportional hazards model for severe postoperative complications, age ≥ 65 years [hazard ratio(HR) = 1.63; 95% confidence interval(CI): 1.18-6.30], body mass index ≥ 24 kg/m^2(HR = 1.20, 95%CI: 1.07-5.89), pancreaticoduodenectomy(HR = 4.86, 95%CI: 1.20-8.31) and length of operation ≥ 241 min(HR = 2.97; 95%CI: 1.04-6.14) were significant(P = 0.010, P = 0.041, P = 0.017 and P = 0.012, respectively).CONCLUSION We found that aging is an independent risk factor for severe postoperative complications after PR. Our results might contribute to more informed decision-making for elderly patients.

Objective Most acute promyelocytic leukemia cases are characterized by the PML-RARa fusion oncogene and low white cell counts in peripheral *** Based on the frequent overexpression of miR-125-family miRNAs in acute promyelocytic leukemia,we examined the consequence of this phenomenon by using an inducible mouse model overexpressing human *** MiR-125b expression significantly accelerates PML-RARa-induced leukemogenesis,with the resultant induced leukemia being partially dependent on continued miR-125b ***,miR-125b expression led to low peripheral white cell counts to bone marrow blast percentage ratio,confirming the clinical observation in acute promyelocytic leukemia *** This study suggests that dysregulated miR-125b expression is actively involved in disease progression and pathophysiology of acute promyelocytic leukemia,indicating that targeting miR-125b may represent a new therapeutic option for acute promyelocytic leukemia.
