L-type calcium channel blockers enhance 5-HTP-induced antinociception in mice

AIM: To investigate the involvement of L-type Ca2+ channels in antinociceptive action induced by the 5-HT precursor,5-hydroxytryptophan (5-HTP). METHODS: Female Kunming mice were treated with either 5-HTP (20-80 mg/kg,ip) alone, or the combination of 5-HTP and fluoxetine (2-8 mg/kg, ip), pargyline (15-60 mg/kg, ip), nimodipine (2.5-10 mg/kg, ip), nifedipine (2.5-10 mg/kg, ip), verapamil (2.5-10 mg/kg, ip), CaCl2 (5-20 mmol/L, icv), or EGTA(0.5-3 mmol/L, icv) prior to the hot-plate test (55 C, hind-paw licking latency). In addition, locomotor activity inmice treated with 5-HTP alone was measured using an ambulometer with five activity boxes. RESULTS: Ipinjection of 5-HTP alone had no influence on the spontaneous locomotor activity, whereas dose-dependently in-creased the latency to licking hind-paw in the hot-plate test in mice. The inhibitory effects of 5-HTP on nociceptiveresponse were significantly enhanced by fluoxetine in the mouse hot-plate test. At a sub-effective dose, pargylinecould cause a leftward shift in the dose-response curve of 5-HTP-induced antinociception. Co-administration with5-HTP and nimodipine, nifedipine, or verapamil obviously potentiated the antinociceptive effects elicited by ***, 5-HTP-induced antinociception was antagonized by CaCl2 and enhanced by EGTA injected icv in themouse hot-plate test. CONCLUSION: These findings suggest that systemic administration of 5-HTP may yield theantinociceptive effects, which are related to Ca2+ influx from extracellular fluid through L-type Ca2+ channels.

Nonlinear forced vibration in a subcritical regime of a porous functionally graded pipe conveying fluid with a retaining clip

This study examines the nonlinear behaviors of a clamped-clamped porous pipe made of a functionally graded material(FGM) that conveys fluids and is equipped with a retaining clip, focusing on primary resonance and subcritical dynamics. The nonlinear governing equations for the FGM pipe are derived by the extended Hamilton's principle, and subsequently discretized through the application of the Galerkin *** direct method of multi-scales is then used to solve the derived equations. A thorough analysis of various parameters, including the clip stiffness, the power-law index, the porosity, and the clip location, is conducted to gain a comprehensive understanding of the system's nonlinear dynamics. Through the analysis of the first natural frequency, the study highlights the influence of the flow velocity and the clip stiffness, while the comparisons with metallic pipes emphasize the role of FGM composition. The examination of the forced response curves reveals saddle-node bifurcations and their dependence on parameters such as the detuning parameter and the power-law index, offering valuable insights into the system's nonlinear resonant behavior. Furthermore, the frequency-response curves illustrate the hardening nonlinearities influenced by factors such as the porosity and the clip stiffness, revealing nuanced effects on the system response and resonance characteristics. This comprehensive analysis enhances the understanding of nonlinear behaviors in FGM porous pipes with a retaining clip, providing key insights for practical engineering applications in system design and optimization.

Crosstalk between degradation and bioenergetics: how autophagy and endolysosomal processes regulate energy production

Cells undergo metabolic reprogramming to adapt to changes in nutrient availability, cellular activity, and transitions in cell states. The balance between glycolysis and mitochondrial respiration is crucial for energy production, and metabolic reprogramming stipulates a shift in such balance to optimize both bioenergetic efficiency and anabolic requirements. Failure in switching bioenergetic dependence can lead to maladaptation and pathogenesis. While cellular degradation is known to recycle precursor molecules for anabolism, its potential role in regulating energy production remains less explored. The bioenergetic switch between glycolysis and mitochondrial respiration involves transcription factors and organelle homeostasis, which are both regulated by the cellular degradation pathways. A growing body of studies has demonstrated that both stem cells and differentiated cells exhibit bioenergetic switch upon perturbations of autophagic activity or endolysosomal processes. Here, we highlighted the current understanding of the interplay between degradation processes, specifically autophagy and endolysosomes, transcription factors, endolysosomal signaling, and mitochondrial homeostasis in shaping cellular bioenergetics. This review aims to summarize the relationship between degradation processes and bioenergetics, providing a foundation for future research to unveil deeper mechanistic insights into bioenergetic regulation.

Morphine attenuates frustration-like behavior induced by sucrose reward deprivation in rats

In this study, a T-maze-based frustration model in rats was established using sucrose-reward deprivation, The results revealed that rats maintained a 75% preference for the sucrose-reward arm in the reward phase. During the sucrose-deprivation frustration phase, both the preference for the sucrose-deprivation arm （62.5%） and time spent waiting in the sucrose-deprivation arm decreased. Acute injection of morphine increased the preference in a dose-dependent fashion, and prolonged the waiting duration in the sucrose-deprivation arm. These findings indicate that morphine specifically inhibited the frustration response induced by sucrose reward deprivation. To further elucidate the pharmacological mechanisms involved, the opioid receptor antagonist naloxone was given to model rats prior to the injection of morphine. The results revealed that naloxone administration markedly attenuated the anti-frustration-like effects of 3 mg/kg morphine treatment. These findings suggest that morphine attenuates the frustration-like response to reward deprivation in rats through the opioid receptor.

The Diagnostic Stability in Dual Diagnosis Study on a Sample of Patients with Opiate Addiction

It is known that in psychiatry, despite the introduction of standardized system, there is still a portion of diagnostic instability. This study aims to evaluate the stability of psychiatric diagnoses in a cohort of patients with dual diagnosis, under treatment of the dependence department of the Local Health Unit n.8 Veneto, Italy. 34 patients with opioid dependence, in treatment with methadone or buprenorphine, have been evaluated. They had participated to a previous study in 2006, in which psychiatric comorbidity was documented. These 34 patients have been retested with some of the tests already used in 2006： M.I.N.I. （for Axis I） and S.C.I.D. II （for Axis II）. The preceding evaluations and those current have been compared to assess the stability of psychiatric diagnosis. Almost all patients （95%） had at least a change of diagnosis. Psychoses are the most stable diagnoses. Also the substance use disorder is quite stable （80%）. Neuroses are more unstable. Personality disorders showed greater variability. Personality disorders appear more unstable in dual diagnosis. The diagnosis of personality disorder based on DSM, common in drug addicts, requires caution. Anyway, the longitudinal observation is important for a correct diagnosis.

Drug dependence and its risk factors in emergency department patients:A retrospective cross-sectional study

Objective:To determine the characteristics and risk factors of drug dependence among patients who were administered drugs with addictive potential(DAP)in an emergency department(ED).Methods:This retrospective cross-sectional study included patients who were administered DAP 3 or more times in the emergency room between September 1,2019 and March 1,*** demographic and baseline information were *** the prescibed DAP,the reasons to use these drugs,secondary drug dependence,the department where DAP were first prescribed,types of doctors who preferred to prescribed DAP,and the risk factors for the development of drug dependence were ***:A total of 3000 patients were screened from medical records,and among them,80 patients developed drug *** dependence only developed for tramadol(n=57,71.3%),diazepam(n=11,13.8%),and biperiden(n=12,15.0%).Tramadol was the most frequently prescribed drug(n=57,71.3%).The most common reason for drug dependence was psychiatric disorders(n=29,36.3%).Drug dependence developed in renal colic patients due to the administration of tramadol(n=7,100%).On the contrary,dependence to biperiden were mainly developed in patients with psychiatric complaints(n=12,41.4%).The rate of secondary drug dependence was 15%(n=12).Of the Biperiden users,41.7%developed secondary drug dependence on *** DAP were first prescribed in the ED(n=52,65%),and the specialist preferred to prescribe DAP(n=43,53.8%).For the development of dependence,the presence of renal colic(OR:3.387,95%confidence interval(CI):1.473-7.788,P=0.004)and low back pain(OR:5.778,95%CI:2.779-12.014,Pdepartments,and specialist are preferred to use *** is the most commonly used drugs caused drug *** disorder patients are easier to develope drug ***,renal colic and low back pain patients needs more attention to

BACKGROUNDIn the post-pandemic era, the emergence of sporadic cases of coronavirus disease 2019 (COVID-19) and the scale of the pandemic are unpredictable. Therefore, the impact ofsporadic cases of COVID-19 and isolation measures on mental health and sleep in different groupsof people need to be *** clarify the severity of psychological problems and insomnia of staff and community residentsaround a hospital with sporadic cases of COVID-19, and their relationship with quarantinelocation and long-term *** cross-sectional survey was conducted on community residents and medical staff. Many of thesemedical staff had been subjected to different places of quarantine. Community residents did notexperience quarantine. Hospital anxiety and depression scale (HADS), acute stress disorder scale(ASDS) and insomnia severity index (ISI) were used to evaluate anxiety and depression, acutestress disorder symptoms, and the severity of insomnia. Additionally, we conducted a 1-yearfollow-up study on medical staff, with related scales measurement immediately after and one yearafter the 2-wk quarantine *** included 406 medical staff and 226 community residents. The total scores of ISI and subscale inHADS of community residents were significantly higher than that of medical staff. Furtheranalysis of medical staff who experienced quarantine showed that 134 were quarantined in hotels,70 in hospitals and 48 at home. Among all subjects, the proportions of HADS, ASDS and ISI scoresabove normal cutoff value were 51.94%, 19.17% and 31.11%, respectively. Multivariable logisticregression analysis found that subjects with higher total ASDS scores had a greater risk to developanxiety and depression. The total ISI score for medical staff in hotel quarantine was significantlyhigher than those in home quarantine. Total 199 doctors and nurses who completed the 1-yearfollow-up study.
