LC-MS/MS analysis and pharmacokinetic study on five bioactive constituents of Tanreqing injection in rats

Tanreqing injection(TRQ), a well-known traditional Chinese medicine formula, is commonly used to treat respiratory diseases. In the present study, a rapid, selective, and sensitive liquid chromatography-tandem mass spectrometry(LC-MS/MS) method was developed and validated to simultaneously determinate the plasma contents of 5 major constituents of TRQ, including chlorogenic acid(CHA), caffeic acid(CFA), baicalin(BA), ursodeoxycholic acid(UDCA) and chenodeoxycholic acid(CDCA) in rats after intravenous administration of TRQ. Chromatographic separation was performed on an Agilent Zorbax SB-C_(18) column(3.5 μm, 100 mm × 2.1 mm), with acetonitrile and 0.1% aqueous formic acid as mobile phase at a flow rate of 0.3 m L·min^(^(-1)). The calibration curves were linear over the ranges of 27.0–13 333.0 ng·m L^(-1) for CFA, 30.0–14 933.0 ng·m L^(-1) for CHA, 50.0–50 333.0 ng·m L^(-1) for BA, 550.0–55 000.0 ng·m L^(-1) for UDCA, and 480.0–48 000.0 ng·m L^(-1) for CDCA, respectively. Intra- and inter-day precisions(relative standard deviations, RSDs) were from 3.11% to 14.08%. The extraction recoveries were greater than 71% and accuracy(relative recovery) was from 89% to 137% for all analytes, except endogenous bile acids. This validated method was successfully applied to the first pharmacokinetic study of CFA, CHA, BA, UDCA and CDCA in rat plasma after intravenous administration of TRQ.

Antidiabetic fallacy of Vernonia amygdalina(bitter leaves) in human diabetes

Objective:Aim of present study is to scientifically,verify the antidiabetic activity/potency of Vernonia amygdalina in human ***:A search was made at Nnewi,South - East Nigeria for known diabetes who use Vernonia amygdalina either as their sole or supplementary antidiabetic.A total of ten volunteers comprising, eight females and two males were *** were all of age range of 36-50 and average weight of 78 kg and suffering from non - insulin form of *** purpose of the study was explained to them and their consent *** were asked not to take any other antidiabetic outside Vernonia amygdalina throughout the four weeks study *** was however,no form of restrictions to their choice of diet or life style. Despite significant advances in imaging techniques, the incidence of colorectal cancer has been increasing in recent years, with many cases still being diagnosed in advanced stages. Early detection and accurate staging remain the main factors that lead to a decrease in the cost and invasiveness of the curative techniques, significantly improving the outcome. However, the diagnosis of pedunculated early colorectal malignancy remains a current challenge. Data on the management of pedunculated cancer precursors, apart from data on nonpolypoid lesions, are still limited. An adequate technique for complete resection, which provides the best long-term outcome, is mandatory for curative intent. In this context, a discussion regarding the diagnosis of malignancy of pedunculated polyps, separate from nonpedunculated variants, is necessary. The purpose of this review is to provide a critical review of the most recent literature reporting the different features of malignant pedunculated colorectal polyps, including diagnosis and management strategies.

Development and validation of a simple UV spectrophotometric method for the determination of levofloxacin both in bulk and marketed dosage formulations

A rapid, specific and economic UV spectrophotometric method has been developed using a solvent composed of water：methanol：acetonitrile （9：0.5：0.5） to determine the levofloxacin content in bulk and pharmaceutical dosage formulations. At a pre-determined 2 of 292 nm, it was proved linear in the range of 1.0 12.0 μg/mL, and exhibited good correlation coefficient （R2=0.9998） and excellent mean recovery （99.0-100.07%）. This method was successfully applied to the determination of levofloxacin content in five marketed brands from Bangladesh and the results were in good agreement with the label claims. The method was validated statistically and by recovery studies for linearity, precision, repeatability, and reproducibility. The obtained results proved that the method can be employed for the routine analysis of levofloxacin in bulks as well as in the commercial formulations.

Role of clove oil in solvent exchange-induced doxycycline hyclate-loaded Eudragit RS in situ forming gel

Solvent exchange induced in situ forming gel(ISG) is the promising drug delivery system for periodontitis treatment owing to the prospect of maintaining an effective high drug level in the gingival crevicular fluid. In the present study, the influence of clove oil(CO) on the characteristics of doxycycline hyclate(DH)-loaded ISG comprising Eudragit RS(ERS) was investigated including viscosity/rheology, syringeability, in vitro gel formation/drug release, matrix formation/solvent diffusion and antimicrobial activities. CO could dissolve ERS and increase the viscosity of ISG and its hydrophobicity could also retard the diffusion of solvent and hinder the drug diffusion; thus, the minimization of burst effect and sustained drug release were achieved effectively. All the prepared ISGs comprising CO could expel through the 27-gauge needle for administration by injection and transform into matrix depot after exposure to the simulated gingival crevicular fluid. The antimicrobial activities against Staphylococcus aureus, Escherichia coli, Streptococcus mutans and Porphyromonas gingivalis were increased when the ratio of CO and N-methyl pyrrolidone(NMP) was decreased from 1:1 to 1:10 owing to higher diffusion of DH except that for C. albicans was increased as CO amount was ***, CO could minimize the burst while prolonging the drug release of DH-loaded ERS ISG for use as a local drug delivery system for periodontitis treatment.
