Assessment of the Pesticides Utilization and the Pesticide Residues Presence in Fresh and Tomato Products for the Tomato Supply Chain in Rwanda

Pesticide residues are either natural or synthetic and are found in most edible products such as fresh fruits, vegetables, meat and other processed products. Excess of these pesticides’ residues is unsafe for consumption due to regulatory measures. Thus, it is beneficial to assess the level of residues of pesticides in the food diet in order to improve the standard of living of the population. This study intends to assess the pesticide usage and residue levels of selected pesticides in fresh tomatoes in the supply chain of Rwanda. The results of the study confirm that there are around 10 brands of pesticides used in Rwanda. 58.97% of the surveyed people around the country are males and 71.79% of farmers apply rockets as the main pesticide. More than 22% of surveyed farmers affirmed to consume the raw or/and unwashed tomatoes. This study revealed that the residues of the active ingredients contained in the pesticides are accumulated in unwashed tomatoes and the quantity of such residues is shown in this study. Deltamethrin was 0.64 ± 0. 009 mg·kg?1 in tomatoes collected in Rusizi district, cypermethrin 1.1435 ± 0. 0375 mg·kg?1 in tomatoes collected from Gisagara District, mancozeb residues was 0.620 ± 0.000 mg·kg?1 in tomatoes collected from Rusizi district. The results of the study also show that pesticide residues are not found in the washed tomatoes. Therefore, the detection of pesticide residues in unwashed tomatoes in some samples indicates the malpractice of residues among farmers in Rwanda. Also, consumers eating the unwashed fresh tomatoes are recommended to wash the tomatoes before use.

Probiotics are live microorganisms which when administered in adequate amounts confer a health benefit on the host. Human-origin Lactobacillus is a preferable source of probiotic bacteria. This study screened 14 vaginal Lactobacillus strains as probiotic candidates by investigating probiotic-related cell surface characteristics including cell surface hydrophobicity (CSH), Lewis acidity/basicity, autoaggregation, and biofilm formation. Moderate to high CSH and autoaggregation, high basicity and low acidity were prevalent in the 14 tested strains. Biofilm formation varied in a large range among the 14 tested strains. CSH showed a high correlation with Lewis acidity and autoaggregation, while Lewis acidity was highly correlated with autoaggregation and biofilm formation. Four strains were selected as promising probiotic strains. This study was the first one to compare antibiotic sensitivity between biofilm-forming cells and planktonic cells of Lactobacillus species, and found that biofilm-forming cells of a L. fermentum strain had a significantly higher survival rate than planktonic cells in cefotaxime, cefmetazole and tetracycline, but were as sensitive to oxacillin and ampicillin as planktonic cells were.

In cancer,myeloid cells have tumor-supporting *** reported that the protein GPNMB(glycoprotein nonmetastatic B)was profoundly upregulated in macrophages interacting with tumor ***,using mouse tumor models,we show that macrophage-derived soluble GPNMB increases tumor growth and metastasis in Gpnmb-mutant mice(DBA/2J).GPNMB triggers in the cancer cells the formation of self-renewing spheroids,which are characterized by the expression of cancer stem cell markers,prolonged cell survival and increased tumor-forming *** the CD44 receptor,GPNMB mechanistically activates tumor cells to express the cytokine IL-33 and its receptor UR1L. We also determined that recombinant IL-33 binding to IL-1R1L is sufficient to induce tumor spheroid formation with features of cancer stem ***,our results reveal a new paracrine axis,GPNMB and IL-33,which is activated during the cross talk of macrophages with tumor cells and eventually promotes cancer cell survival,the expansion of cancer stem cells and the acquisition of a metastatic phenotype.

SNP mutations in the HOXB13 gene associated with prostate cancer were determined in Moroccans prostate cancer patients (PCa). All PCa SNP mutations were new and belong to the SNP point-mutations located on the stop codon of HOXB13 exon 1 and 2 located in chromosome 17. The five mutations and their frequencies were as follows: rs1197613952 (12%), rs1597934612 (4%), rs1597933874 (4%), rs1597933837 (4%) and rs867793282 (4%). The European HOXB13-G84E (rs138213197) PCa mutation was not detected among Moroccan patients. The Y-chromosome genealogical haplotypes of the Western European (R1b1b2-M2G9) and the Eastern European (R191a-M-17) were not observed in Moroccans PCa patients. The patients have their own haplotypes E1b1 and J with a frequency of 55 and 35%, respectively. The results of the SNP mutations in the HOXB13, the absence of the HOXB13-G84E of the European in the Moroccans PCa patients, the absence of the European-lineage haplogroups (R1a1a-M17 and R1b1b2-M269) and the presence of E1b1b and J in Moroccans PCa patients would clearly indicate the absence of gene flow from European to Moroccans gene pool.

Understanding fatigue in primary biliary cholangitis:From pathophysiology to treatment perspectives

Fatigue is considered one of the most frequent and debilitating symptoms in primary biliary cholangitis(PBC),affecting over 50%of PBC *** in five patients with PBC suffer from severe fatigue,which significantly impairs quality of *** is made up of a central and a peripheral component,whose pathophysiology is still greatly *** fatigue is characterised by a lack of self-motivation and can manifest both in physical and mental activities(lack of intention).Peripheral fatigue includes neuromuscular dysfunction and muscle weakness(lack of ability).Peripheral fatigue could be explained by an excessive deviation from aerobic to anaerobic metabolism leading to excessive lactic acid accumulation and therefore accelerated decline in muscle function and prolonged recovery *** opposed to itching,and with the exception of endstage liver disease,fatigue is not related to disease *** objective of this review is to outline current understanding regarding the pathophysiology of fatigue,the role of comorbidities and contributing factors,the main tools for fatigue assessment,the failed therapeutic options,and future treatment perspectives for this disabling *** fatigue is an extremely common and debilitating symptom and there is still no licensed therapy for fatigue in PBC patients,further research is warranted to understand its causative mechanisms and to find an effective treatment.

Krill oil (KO) exhibits various biological activities， such as anti-inflammatory and antitumor effects. However， the inhibitory effects of benign prostatic hyperplasia (BPH) in vitro and in vivo have not yet been studied. This study investigated the anti-BPH effects of KO extracted by an enzymatic hydrolysis method. KO treatment inhibited the proliferation of WMPY-1 and BPH-1 cells by induction of G0/G1phase arrest through the modulation of positive and negative regulators in both prostate cell types. KO treatment stimulated phosphorylation of JNK and p38 signaling. In addition， KO changed the expression of BPH-related markers (5α-reductase， androgen receptor， FGF， Bcl-2， and Bax) and the activity of the proliferation-mediated NF-κB binding motif. KO-induced levels of proliferation-mediated molecules of prostate cells were attenuated in the presence of siRNA-specific p-38 (si-p38) and JNK (si-JNK). Furthermore， the administration of KO alleviated prostate size and weight and the cell layer thickness of prostate glands in a testosterone enanthate-induced BPH rat model. KO treatment altered the level of dihydrotestosterone in serum and the expression levels of BPH-related markers in prostate tissues. Finally， KO-mediated inhibition of prostatic growth was validated by histological analysis. These results suggest that KO has an inhibitory effect on BPH in prostate cells in vitro and in vivo. Thus， KO might be a potential prophylactic or therapeutic agent for patients with BPH.

Integration of internal and external cues into developmental programs is indispensable for growth and development of plants, which involve complex interplays among signaling pathways activated by the internal and external factors （IEFs）. However, decoding these complex interplays is still challenging. Here, we present a web-based platform that identifies key regulators and Network models delineating Interplays among Developmental signaling （iNID） in Arabidopsis. iNID provides a comprehensive resource of （1） transcriptomes previously collected under the conditions treated with a broad spectrum of IEFs and （2） protein and genetic interactome data in Arabidopsis. In addition, iNID provides an array of tools for identifying key regulators and network models related to interplays among IEFs using transcriptome and interactome data. To demonstrate the utility of iNID, we investigated the interplays of （1） phytohormones and light and （2） phytohormones and biotic stresses. The results revealed 34 potential regulators of the interplays, some of which have not been reported in association with the interplays, and also network models that delineate the involvement of the 34 regulators in the interplays, providing novel insights into the interplays collectively defined by phytohormones, light, and biotic stresses. We then experimentally verified that BME3 and TEM1, among the selected regulators, are involved in the auxin-brassinosteroid （BR）-blue light interplay. Therefore, iNID serves as a useful tool to provide a basis for understanding interplays among IEFs.

Efficacy of Children Neuropsychological and Behavioral Scale in Screening for Autism Spectrum Disorders through a Combination of Developmental Surveillance

Objective This study aimed to explore the clinical value of Children Neuropsychological and Behavioral Scale-Revision 2016(CNBS-R2016)for Autism Spectrum Disorder(ASD)screening in the presence of developmental *** All participants were evaluated by the CNBS-R2016 and Gesell Developmental Schedules(GDS).Spearman’s correlation coefficients and Kappa values were *** GDS as a reference assessment,the performance of the CNBS-R2016 for detecting the developmental delays of children with ASD was analyzed with receiver operating characteristic(ROC)*** efficacy of the CNBS-R2016 to screen for ASD was explored by comparing Communication Warning Behavior with Autism Diagnostic Observation Schedule,Second Edition(ADOS-2).Results In total,150 children aged 12–42 months with ASD were *** developmental quotients of the CNBS-R2016 were correlated with those of the GDS(r=0.62–0.94).The CNBS-R2016 and GDS had good diagnostic agreement for developmental delays(Kappa=0.73–0.89),except for Fine *** was a significant difference between the proportions of Fine Motor,delays detected by the CNBS-R2016 and GDS(86.0%vs.77.3%).With GDS as a standard,the areas under the ROC curves of the CNBS-R2016 were above 0.95 for all the domains except Fine Motor,which was *** addition,the positive rate of ASD was 100.0%and 93.5%when the cut-off points of 7 and 12 in the Communication Warning Behavior subscale were used,*** The CNBS-R2016 performed well in developmental assessment and screening for children with ASD,especially by Communication Warning Behaviors ***,the CNBS-R2016 is worthy of clinical application in children with ASD in China.
