Biodegradable magnesium materials regulate ROS-RNS balance in pro-inflammatory macrophage environment

The relationship between reactive oxygen and nitrogen species(ROS-RNS)secretion and the concomitant biocorrosion of degradable magnesium(Mg)materials is poorly understood.We found that Mg foils implanted short term in vivo(24 h)displayed large amounts of proinflammatory F4/80+/iNOS+macrophages at the interface.We sought to investigate the interplay between biodegrading Mg materials(98.6%Mg,AZ31&AZ61)and macrophages(RAW 264.7)stimulated with lipopolysaccharide(RAW 264.7^(LPS))to induce ROS-RNS secretion.To test how these proinflammatory ROS-RNS secreting cells interact with Mg corrosion in vitro,Mg and AZ61 discs were suspended approximately 2 mm above a monolayer of RAW 264.7 cells,either with or without ^(LPS).The surfaces of both materials showed acute(24 h)changes when incubated in the proinflammatory RAW 264.7^(LPS) environment.Mg discs incubated with RAW 264.7^(LPS) macrophages showed greater corrosion pitting,while AZ61 showed morphological and elemental bulk product changes via scanning electron microscopy-energy dispersive X-ray spectroscopy(SEM-EDX).X-ray photoelectron spectroscopy(XPS)analysis showed a reduction in the Ca/P ratio of the surface products for AZ61 disc incubated with RAW 264.7^(LPS),but not the Mg discs.Moreover,RAW 264.7^(LPS) macrophages were found to be more viable in the acute biodegradative environment generated by Mg materials,as demonstrated by calcein-AM and cleaved(active)caspase-3 staining(CC3).^(LPS) stimulation caused an increase in ROS-RNS,and a decrease in antioxidant peroxidase activity.Mg and AZ61 were found to change this ROS-RNS balance,independently of physiological antioxidant mechanisms.The findings highlight the complexity of the cellular driven acute inflammatory responses to different biodegradable Mg,and how it can potentially affect performance of these materials.

Transplantation of neuronal precursors derived from human pluripotent stem cells is a promising therapy for the treatment of neurological disorders associated with neuronal loss,such us neurodegenerative diseases,brain trauma and stroke.The functional integration of grafted neurons differentiated from stem cells into the host injured neuronal circuitry has been a major challenge in cell therapy strategies for brain repair(Palma-Tortosa et al.,2021).Even though other cell types or mechanisms may provide modest clinical improvements,neuronal replacement and reconstruction of the damaged area are crucial for an optimal and long-term recovery.

Physical activity and exercise have several beneficial roles in enhancing both physiological and psychological well-being of an individual.In addition to aiding the regulation of aerobic and anaerobic metabolism,exercise can stimulate the synthesis of exerkine hormones in the circulatory system.Among several exerkines that have been investigated for their therapeutic potential,Brain-derived neurotrophic factor(BDNF)is considered the most promising candidate,especially in the management of neurodegenerative diseases.Owing to the ability of physical activity to enhance BDNF synthesis,several experimental studies conducted so far have validated this hypothesis and produced satisfactory results at the pre-clinical level.This review highlights some of the recent animal model studies that have evaluated the efficiency of exercise in enhancing BDNF synthesis and promoting neuroprotective effects.Further,this review focuses on understanding the therapeutic benefits of exercise-induced exerkine synthesis as a non-pharmacological strategy in Parkinson’s disease(PD).Regarding physical activity and exerkine induction,the neuromuscular electrical stimulation(NMES)strategy could be considered as an alternate treatment modality for patients affected with PD.

Early-photon guided reconstruction method for time-domain fluorescence lifetime tomography

A reconstruction method guided by early-photon fluorescence yield tomography is proposed for time-domain fluorescence lifetime tomography （FLT） in this study. The method employs the early-arriving photons to reconstruct a fluorescence yield map, which is utilized as a priori information to reconstruct the FLT via all the photons along the temporal-point spread functions. Phantom experiments demonstrate that, compared with the method using all the photons for reconstruction of fluorescence yield and lifetime maps, the proposed method can achieve higher spatial resolution and reduced crosstalk between different targets without sacrificing the quantification accuracy of lifetime and contrast between heterogeneous targets.

A mathematic equation derived from host-pathogen interactions elucidates the significance of integrating modern medicine with traditional Chinese medicine to treat infectious diseases

The prognosis of infectious diseases is determined by host-pathogen interactions.Control of pathogens has been the central dogma of treating infectious diseases in modern medicine,but the pathogen-directed medicine is facing significant challenges,including a lack of effective antimicrobials for newly emerging pathogens,pathogen drug resistance,and drug side effects.Here,a mathematic equation(ter-med equation of host-pathogen interactions,HPI-Equation)is developed to dissect the key variables of hostpathogen interactions.It shows that control of pathogens does not necessarily lead to host recov-ery.Instead,a combination of promoting a host’s power of self-healing and balancing immune responses provides the best benefit for host.Moreover,the HPI-Equation elucidates the scientific basis of traditional Chinese medicine(TCM),a host-based medicine that treats infectious diseases by promot-ing self-healing power and balancing immune responses.The importance of self-healing power eluci-dated in the HPIEquation is confirmed by recent studies that the tolerance mechanism,which is discovered in plants and animals and conceptually similar to self-healing power,improves host survival without directly attacking pathogens.In summary,the HPI-Equation describes host-pathogen interac-tions with mathematical logic and precision;it translates the ancient wisdoms of TCM into apprehen-sible modern sciences and opens a new venue for integrating TCM and modern medicine for a future medicine.

Neurodegeneration affects a large number of cell types including neurons,astrocytes or oligodendrocytes,and neural stem cells.Neural stem cells can generate new neuronal populations through proliferation,migration,and differentiation.This neurogenic potential may be a relevant factor to fight neurodegeneration and aging.In the last years,we can find growing evidence suggesting that melatonin may be a potential modulator of adult hippocampal neurogenesis.The lack of therapeutic strategies targeting neurogenesis led researchers to explore new molecules.Numerous preclinical studies with melatonin observed how melatonin can modulate and enhance molecular and signaling pathways involved in neurogenesis.We made a special focus on the connection between these modulation mechanisms and their implication in neurodegeneration,to summarize the current knowledge and highlight the therapeutic potential of melatonin.

Multimodal treatments of right gastroepiploic arterial leiomyosarcoma with hepatic metastasis:A case report and review of the literature

Leiomyosarcoma of an artery is very rare, and cases with hepatic metastasis are even rarer. We describe a case of a 70-year-old man who after follow up due to rectal cancer, presented with an intra-abdominal hypervascular mass and a hepatic mass. After surgical resection, it was diagnosed as a leiomyosarcoma of the right gastroepiploic artery with hepatic metastasis. Multiple metastases had recurred at the liver. He has survived more than 53 mo through multimodal treatments(three surgical resections, radiofrequency ablation, transarterial chemoembolization, chemotherapies, and targeted therapy). Multimodal treatments, including active surgical resection, may behelpful in the treatment of aggressive diseases such as arterial leiomyosarcoma with metastasis.

Choroid plexus tumor necrosis factor receptor 1:a new neuroinflammatory piece of the complex Alzheimer's disease puzzle

Due to the aging of the population and despite the enormous scientific effort,Alzheimer's disease remains one of the biggest medical and pharmaceutical challenges in current medicine.Novel insights highlight the importance of neuroinflammation as an undeniable player in the onset and progression of Alzheimer's disease.Tumor necrosis factor is a master inflammatory cytokine that signals via tumor necrosis factor receptor 1 and tumor necrosis factor receptor 2,but that also regulates several brain functions in health and disease.However,clinical trials investigating drugs that interfere with the tumor necrosis factor pathway in Alzheimer's disease led to inconclusive results,partially because not only the pro-inflammatory tumor necrosis factor/tumor necrosis factor receptor 1,but also the beneficial tumor necrosis factor/tumor necrosis factor receptor 2 signaling was antagonized in these trials.We recently found that tumor necrosis factor is the main upregulated cytokine in the choroid plexus of Alzheimer's disease patients,signaling via tumor necrosis factor receptor 1.In agreement with this,choroidal tumor necrosis factor/tumor necrosis factor receptor 1 signaling was also upregulated in different Alzheimer's disease mouse models.Interestingly,both genetic and nanobody-based pharmacological blockage of tumor necrosis factor receptor 1 signaling was accompanied by favorable effects on Alzheimer's disease-associated inflammation,choroidal morphology and cognitive functioning.Here,we briefly summarize the detrimental effects that can be mediated by tumor necrosis factor/tumor necrosis factor receptor 1 signaling in(early) Alzheimer's disease,and the consequences this might have on the disease progression.As the main hypothesis in Alzheimer's disease clinical trials is still based on the amyloid beta-cascade,the importance of Alzheimer's disease-associated neuroinflammation urge the development of novel therapeutic strategies that might be effective in the early stages of Alzheimer's disease and prevent the irreversi

Deep learning in cortical surface-based neuroimage analysis:a systematic review

Deep learning approaches,especially convolutional neural networks(CNNs),have become the method of choice in the field of medical image analysis over the last few years.This prevalence is attributed to their excellent abilities to learn features in a more effective and efficient manner,not only for 2D/3D images in the Euclidean space,but also for meshes and graphs in non-Euclidean space such as cortical surfaces in neuroimaging analysis field.The brain cerebral cortex is a highly convoluted and thin sheet of gray matter(GM)that is thus typically represented by triangular surface meshes with an intrinsic spherical topology for each hemisphere.Accordingly,novel tailored deep learning methods have been developed for cortical surface-based analysis of neuroimaging data.This paper reviewsed the representative deep learning techniques relevant to cortical surface-based analysis and summarizes recent major contributions to the field.Specifically,we surveyed the use of deep learning techniques for cortical surface reconstruction,registration,parcellation,prediction,and other applications.We concluded by discussing the open challenges,limitations,and potentials of these techniques,and suggested directions for future research.
