Proteases are enzymes that cleave and hydrolyse the peptide bonds between two specific amino acid residues of target substrate ***-controlled proteolysis plays a key role in the degradation and recycling of proteins,which is essential for various physiological ***,solving the substrate identification problem will have important implications for the precise understanding of functions and physiological roles of proteases,as well as for therapeutic target identification and pharmaceutical ***,there is a great demand for bioinformatics methods that can predict novel substrate cleavage events with high accuracy by utilizing both sequence and structural *** this study,we present Procleave,a novel bioinformatics approach for predicting protease-specific substrates and specific cleavage sites by taking into account both their sequence and 3D structural *** features of known cleavage sites were represented by discrete values using a LOWESS data-smoothing optimization method,which turned out to be critical for the performance of *** optimal approximations of all structural parameter values were encoded in a conditional random field(CRF)computational framework,alongside sequence and chemical group-based ***,we demonstrate the outstanding performance of Procleave through extensive benchmarking and independent *** is capable of correctly identifying most cleavage sites in the case ***,when applied to the human structural proteome encompassing 17,628 protein structures,Procleave suggests a number of potential novel target substrates and their corresponding cleavage sites of different *** is implemented as a webserver and is freely accessible at http://***/.

The biomolecular mechanisms that regulate tooth root development and odontoblast differentiation are poorly *** found that Atp6i deficient mice(Atp6i^(−/−))arrested tooth root formation,indicated by truncated Hertwig’s epithelial root sheath(HERS)***,Atp6i deficiency significantly reduced the proliferation and differentiation of radicular odontogenic cells responsible for root ***6i^(−/−)mice had largely decreased expression of odontoblast differentiation marker gene expression profiles(Col1a1,Nfic,Dspp,and Osx)in the alveolar ***6i^(−/−)mice sample RNA-seq analysis results showed decreased expression levels of odontoblast ***,there was a significant reduction in Smad2/3 activation,inhibiting transforming growth factor-β(TGF-β)signaling in Atp6i^(−/−)*** treating pulp precursor cells with Atp6i^(−/−)or wild-type OC bone resorption-conditioned medium,we found the latter medium to promote odontoblast differentiation,as shown by increased odontoblast differentiation marker genes expression(Nfic,Dspp,Osx,and Runx2).This increased expression was significantly blocked by anti-TGF-β1 antibody neutralization,whereas odontoblast differentiation and Smad2/3 activation were significantly attenuated by Atp6i^(−/−)OC conditioned ***,ectopic TGF-β1 partially rescued root development and root dentin deposition of Atp6i^(−/−)mice tooth germs were transplanted under mouse kidney ***,our novel data shows that the prevention of TGF-β1 release from the alveolar bone matrix due to OC dysfunction may lead to osteopetrosis-associated root formation via impaired radicular odontoblast *** such,this study uncovers TGF-β1/Smad2/3 as a key signaling pathway regulating odontoblast differentiation and tooth root formation and may contribute to future therapeutic approaches to tooth root regeneration.

Regional Heterogeneity in 3D Myocardial Shortening in Hypertensive Left Ventricular Hypertrophy: A Cardiovascular CMR Tagging Substudy to the Life Study

Background: Increased relative wall thickness in hypertensive left ventricular hypertrophy (LVH) has been shown by echocardiography to allow preserved shortening at the endocardium despite depressed LV midwall circumferential shortening (MWCS). Depressed MWCS is an adverse prognostic indicator, but whether this finding reflects reduced global or regional LV myocardial function, as assessed by three-dimensional (3D) myocardial strain, is unknown. Methods and Results: Cardiac Magnetic Resonance (CMR) tissue tagging permits direct evaluation of regional 3D intramyocardial strain, independent of LV geometry. We evaluated 21 hypertensive patients with electrocardiographic LVH in the LIFE study and 8 normal controls using 3D MR tagging and echocardiography. Patients had higher MR LV mass than normals (116 ± 40 versus 63 ± 6 g/m2, P = 0.002). Neither echocardiographic fractional shortening (32 ± 6 versus 33% ± 3%), LVEF (63% versus 64%) or mean end-systolic stress (175 ± 27 versus 146 ± 28 g/cm2) were significantly different, yet global MWCS was decreased by both echocardiography (13.4 ± 2.8 versus 18.2% ± 1.5%, P P P = 0.002) in LVH and greater in lateral and anterior regions versus septal and posterior regions ( P P P 0.60, P = 0.001 for both). Conclusions: In patients with hypertensive LVH, despite normal LV function via echocardiography or CMR, CMR intramyocardial tagging show depressed global MWCS while 3D MR strain revealed marked underlying regional heterogeneity of LV dysfunction.

背景:心血管并发症是糖尿病患者的主要并发症之一。糖尿病状态下,血管内皮细胞发生内皮间质转化（endothelial to mesenchymal transition,EnMT）。EnMT指内皮细胞失去其特异性抗原如VE-cadherin,CD31,VEGFR-1/2,而获得间质细胞抗原如α-Smooth muscle actin （α-SMA）,Ⅰ型和Ⅲ型胶原,同时在损伤部位出现脂质沉积、血小板黏附聚集、释放生长因子,引起血管壁增厚,发生形态学改变。环状RNA是一类新型非编码RNA,研究表明,环状RNA在心血管疾病中发挥了重要作用。mm9irc10056（ID:mmuirc001572）的基因位置是chr7:66496336-66502587,宿主基因是Ube3a,其由Memczak S等人于2015年发现并发表于Nature上。目的:观察mm9irc10056对糖尿病诱导的动脉内皮细胞间质转化中的作用,其机制可能涉及Ube3a。方法:取自发性Ⅱ型糖尿病模型db/db小鼠（n=6）和野生型对照小鼠（n=6）胸主动脉,提取RNA;培养小鼠主动脉内皮细胞（mouse aortic endothelial cells,MAEC）,用重组晚期糖基化终末产物（advanced glycation end products,AGEs）(0,50,100,200μg/mL)分别孵育4h,24h,48h。mm9irc10056过表达腺病毒和siRNA转染MAEC细胞,分别观察mm9irc10056对AGEs诱导的EnMT作用及对Ube3a表达的影响。Ube3a过表达慢病毒转染MAEC细胞,观察Ube3a对AGEs诱导的EnMT的作用。采用qRT-PCR和FISH检测mm9irc10056的表达;通过免疫荧光和Westrenblot检测CD31和α-SMA表达来评价EnMT的变化。结果:与野生型小鼠相比,db/db小鼠中mm9irc10056表达明显下调。AGEs呈浓度和时间依赖性地抑制MAEC细胞中mm9irc10056表达。MAEC细胞经AGEs诱导后,CD31表达明显下调,α-SMA表达明显上调。干扰mm9irc10056能诱导EnMT的发生;而采用腺病毒过表达mm9irc10056能上调Ube3a的mRNA表达水平。外源性的AGEs能抑制MAEC中Ube3a的表达,而过表达Ube3a能抑制AGEs诱导的EnMT的发生。结论:mm9irc10056参与调节糖尿病血管内皮细胞EnMT过程,其作用机制可能由其介导宿主基因Ube3a的表达来进行。

To assess the peripheral refractive changes that occur when eye growth decelerates in *** We measured peripheral refraction and on-axis vitreous chamber depth on a total of 53 marmosets;36 treated monocularly for 12 weeks 9 hrs/day(26 with-5 D contact lenses,10 with+5 D)and 17 untreated *** the 26 marmosets treated with-5 D,10 wore contact lenses 9 hrs/day without interruptions and were mea-sured at the end of 10 weeks of treatment(T10)and after 4 weeks of recovery(R14).Eight had both contact lenses removed for 30 mins twice/day(mid-morning and mid-afternoon)during the first four weeks of treatment(early interruption group)and eight had the lenses removed during the second four weeks of treatment(late interruption group).The two interruption groups were measured after 4,8 and 12 weeks of treatment only(T4,T8,T12).
