Bioaccumulation of metals in human blood in industrially contaminated area

Heavy metals were analyzed in different foods crops, milk, meat and blood samples collected from different age group subjects such as children （1–12 years）, adolescent （12–18 years）, adults （18–45 years） and old age （above 45 and 55 years for males and females, respectively） from polluted and relatively less polluted areas. The results revealed that the consumption of contaminated food crops, meat and milk have significantly increased the concentrations of selected metals in the human blood. Cu, Zn and Mn concentrations were significantly higher （p 0.05） in the blood samples collected from the polluted area as compared to control area. Old people had accumulated high concentrations of metals as compared to the younger ones within the same area. Males accumulated higher concentrations of metals as compared to females.

Medicinal plants have been used by marginal communities to treat various ailments. However, the potential of endophytes within these bio-prospective medicinal plants remains unknown. The present study elucidates the endophytic diversity of medicinal plants（Caralluma acutangula, Rhazya stricta, and Moringa peregrina） and the endophyte role in seed growth and oxidative stress. Various organs of medicinal plants yielded ten endophytes, which were identified as Phoma sp.（6 isolates）, Alternaria sp.（2）, Bipolaris sp.（1）, and Cladosporium sp.（1） based on 18 S rD NA sequencing and phylogenetic analysis. The culture filtrates（CFs; 25%, 50%, and 100% concentrations） from these endophytes were tested against the growth of normal and dwarf mutant rice lines. Endophytic CF exhibited dose-dependent growth stimulation and suppression effects. CF（100%） of Phoma sp. significantly increased rice seed germination and growth compared to controls and other endophytes. This growth-promoting effect was due to the presence of indole acetic acid in endophytic CF. The gas chromatography/mass spectrometry（GC/MS） analysis showed the highest indole acetic acid content（（54.31±0.21） μmol/L） in Bipolaris sp. In addition, the isolate of Bipolaris sp. exhibited significantly higher radical scavenging and anti-lipid peroxidation activity than the other isolates. Bipolaris sp. and Phoma sp. also exhibited significantly higher flavonoid and phenolic contents. The medicinal plants exhibited the presence of bio-prospective endophytic strains, which could be used for the improvement of crop growth and the mitigation of oxidative stresses.

Objective:To screen for Escherichia coli(***)resistant to tetracycline,followed by identification of tet efflux genes by polymerase chain reaction(PCR).In addition,detection of tetracycline residues in chicken livers and kidneys were conducted using high performance liquid chromatography-tandem quadrupole mass spectrometry(HPLC-MS-MS).Methods:Strains of *** were isolated from samples of chicken colon and screened for tetracycline *** genes conferring resistance(Tc^r)were detected by polymerase chain reaction(PCR).Most of the isolates were resistant to tetracycline(97.9%).Results:PCR analysis indicated that Tc^r *** R-plasmids contained tet(A),tet(B)and a combination of both efflux *** of the isolates contained other efflux tet genes tet(C,D,E and Y).High performance liquid chromatography-tandem quadrupole mass spectrometry(HPLC-MS-MS),a sensitive technique,was used to detect residues of chlortetracycline(CTC),oxytetracyeline(OTC),doxveycline(DC)in chicken livers and *** samples containing tetracycline residues were at 0.13-0.65pg/μL ***:Tetracycline and other antibiotics are commonly used in the poultry and meat production industry for prevention of microbial *** antibiotic resistant bacteria in Oman have increased to alarming levels,threatening public health,domestic and may have adverse effect on environment.

Cucurbitaceae family contains important economic and medical crops, they can be divided into two categories according to the worldwide consumption, use, and production. The Cucurbit Popular Crops (CuPoC) are comprised of cucumber, the Cucurbita group (pumpkin and squash), melon and watermelon. On the other hand, Neglected and Underutilized Cucurbit Species (NUCuS) group has been used as food sources, medicinal properties and elements in the elaboration of different types of items. The NUCuS is represented in this review work by bitter gourd, bottle gourd, chayote, ridge gourd, and snake gourd, which are recognized mainly in Asia. The center of origin of the majority of NUCuS was proposed to be in the Old World. In contrast, the origin of chayote or Sechiumedule (Jacq.) Sw. was suggested in the New World, precisely in Mesoamerican region based on linguistic uses and distribution of wild relatives. The environmental factors along with artificial selection, production systems and traditional knowledge have been influenced the evolutionary history of NUCuS, Infraspecific variation of chayote has been reported in Mexico over-described varietal groups. These descriptors were determined based on biochemical and morpho-structural traits;however, cytogenetic analyses are scarce. Specifically, chromosome and nuclear content analyses are important to support botanical groups, analyze artificial selection history, developing breeding and conservation programs. The present review paper discusses agronomic and evolutionary importance based on cytological evidence in NUCuS, mainly in the prominent chayote;with the perspective to prompt breeding, conservation, cytology, structural and functional genomics research for its sustainable utilization.

Tracking Selection Signatures Based on Variation in <i>OsLEA</i>27 within Myanmar Landraces of Upland and Dryland Rice

To track the selection evident along the genome segment of OsLEA27?gene, a member of dehydrin gene family, 2.9-kbp nucleotide sequence containing the promoter 5’ upstream and transcribed region of OsLEA27?was determined for 35 upland and dryland Myanmar landraces from drought-prone areas. Nucleotide diversity, neutrality tests, haplotype network analysis, and linkage disequilibrium (LD) analysis were performed to infer the impact of selection and to investigate nonrandom associations of SNPs within all or part of the entire OsLEA27?region. The evidence for LD, the presence of two distinct haplotype groups across four different geographical regions, and the significant values obtained in a sliding-window analysis of mutation-drift tests all suggest the effects of selection on OsLEA27?in a set of 30 landraces. The neutrality test values for 5’ upstream region of OsLEA27?were significantly negative (p OsLEA27 region was significantly negative in accessions of the northern group, indicating a recent increase in population size or selection pressure. This evidence for selection signatures at OsLEA27 in this study sample provides insight into the roles of selection, crop adaptation, and genetic diversity in establishing present-day variation at the OsLEA27 locus.

Characterization of the reproductive morphology in genus Zingiber is still relevant on classical taxonomic studies because most of the species are phenotypically similar and difficult to distinguish in the absence of the flowering stage. Reports mainly derived based on the herbarium collections are insufficient for the detailed morphological review. Zingiber barbatum (Wall.) belongs to the genus Zingiber and has been neglected for a long time. Existed intraspecific morphological and genetic variations also intricate the description of this species. The objective of this study was to characterize the reproductive morphology of under-exploited Z. barbatum species. The study was done based on the traditional description of morphology and habit of inflorescence and flowers, with a recording of minimum quantitative and qualitative parameters and phenological observation regarding inflorescence life span and duration of blossom. Assessment of reproductive morphology displayed phenotypic variations regarding inflorescences habit during the growth stage and morphological features of the flower. Oman is located in the tropic of Cancer with extremely hot temperatures ranging between 15°C-25°C in winter and 30 - 48 in the summer with low rainfall. Most of the water source is desalinated seawater. Recycling of sewage effluent is a common practice used for farming, public parks and industry. The dried sludge product is used as fertilizer. The highly concentrated sludge with heavy metals is either incinerated or buried in landfills. Heavy metals were analyzed and compared in treated sewage effluent (TSE), slurry sludge, dried sludge, landfill and underground water. Heavy metal concentrations were analyzed using the inductive coupled plasma optical emission spectrometer (ICP-EOS). All samples contained the same heavy metals with different concentrations. The heaviest concentrations were Zn, Cu, Cr and Ni with traces of V, Cd, Pb and Ba. TSE and sludge were the main sources of landfill and underground water contamination. Due to water evaporation, heavy metals concentrations in sludge were higher compared to TSE and underground water. Dried sludge and landfill contained higher concentrations than the others. Based on this investigation, it appears that the source of heavy metals is from TSE originated mainly in industries. If the infiltration of heavy metals to soil and underground water continues, it will be a serious environmental and health problem in the future.

The Effect of Physical Factors on Fecal Coliform Viability Rate in Sewage Sludge

Experiments were conducted on the survival rate of fecal coliform after spreading the sewage sludge for desiccation on sludge beds in an open field throughout the year. The ambient temperature, humidity, sunlight intensity and solar radiation were measured during the period of sludge desiccation. The maximum average temperature, sunlight intensity and solar radiation were recorded throughout the year. During May and August the minimum microbial counts were recorded after four weeks of sludge desiccation. Relative humidity did not have significant effect on survival rate. The minimum microbial counts were taken after four weeks of sludge drying. When the sludge was mixed with three different media, the maximum count of fecal coliform was presented when soil mixed with sludge and irrigated with treated sewage effluent (TSE). When soil mixed with sludge irrigated with well water, the number of fecal coliform count dropped significantly. The minimum count occurred when compost was used instead of sludge. This was a clear indication that the source of contamination was from sludge and was mostly contributed by the TSE irrigation. The use of dried sludge after four weeks of treatment had the lowest fecal coliform count and was safe to be used as fertilizer for farming. Oman is located in a semi-equatorial belt with arid environmental conditions which are ideal for elimination of microbial pathogens. Thus, sludge desiccation in such environmental conditions, specifically during the hot months of the year, is perfect for such practice.
