To assess the peripheral refractive changes that occur when eye growth decelerates in *** We measured peripheral refraction and on-axis vitreous chamber depth on a total of 53 marmosets;36 treated monocularly for 12 weeks 9 hrs/day(26 with-5 D contact lenses,10 with+5 D)and 17 untreated *** the 26 marmosets treated with-5 D,10 wore contact lenses 9 hrs/day without interruptions and were mea-sured at the end of 10 weeks of treatment(T10)and after 4 weeks of recovery(R14).Eight had both contact lenses removed for 30 mins twice/day(mid-morning and mid-afternoon)during the first four weeks of treatment(early interruption group)and eight had the lenses removed during the second four weeks of treatment(late interruption group).The two interruption groups were measured after 4,8 and 12 weeks of treatment only(T4,T8,T12).

Histone H3.3 glycine 34 to arginine/valine(G34R/V)mutations drive deadly gliomas and show exquisite regional and temporal specificity,suggesting a developmental context permissive to their *** we show that 50%of G34R/V tumors(n=95)bear activating PDGFRA mutations that display strong selection pressure at *** considered gliomas,G34R/V tumors actually arise in GSX2/DLX-expressing interneuron progenitors,where G34R/V mutations impair neuronal *** lineage of origin may facilitate PDGFRA co-option through a chromatin loop connecting PDGFRA to GSX2 regulatory elements,promoting PDGFRA overexpression and *** the single-cell level,G34R/V tumors harbor dual neuronal/astroglial identity and lack oligodendroglial programs,actively repressed by GSX2/DLX-mediated cell fate specification.G34R/V may become dispensable for tumor maintenance,whereas mutant-PDGFRA is potently ***,our results open novel research avenues in deadly tumors.G34R/V gliomas are neuronal malignancies where interneuron progenitors are stalled in differentiation by G34R/V mutations and malignant gliogenesis is promoted by co-option of a potentially targetable pathway,PDGFRA signaling.

血管紧张素转化酶(ACE)抑制剂通过影响肾素-血管紧张素系统,对减缓和抑制高血压具有重要的作用.该研究通过超滤、凝胶过滤色谱、反相高效液相色谱等方法,从钝顶螺旋藻的木瓜蛋白酶水解液中分离、纯化得到一种血管紧张素转化酶(ACE)抑制肽,并利用基质辅助激光解吸电离-飞行时间质谱(MALDI-TOF-MS)和氨基酸测序对纯化肽进行鉴定.此外,对其抑制类型和体外模拟消化环境稳定性也进行了研究.结果表明,分子质量范围为0～3000ku的酶解液ACE抑制活性最高,IC50值为(1.03±0.04)g/L.该部分酶解液通过纯化获得ACE抑制肽,IC50值为(0.0094±0.0002)g/L,相当于(27.36±0.14)μmol/L,序列经鉴定为***-Burk图和Dixon图表明该ACE抑制肽为非竞争性抑制剂,Ki值为(23.59±0.54)μmol/L.体外稳定性实验显示,该抑制肽在胃蛋白酶、胰凝乳蛋白酶、胰蛋白酶等胃肠蛋白酶的消化下能够保持良好的抑制活性,表明螺旋藻源ACE抑制肽可以用于降血压功能食品和药剂方面,具有很好的发展前景.

Chronic hepatitis C virus(HCV) infection can result in steatosis,a condition displaying aberrant accumulation of neutral lipid vesicles,the component of lipid droplets(LDs),which are essential for HCV ***,the interplays between HCV infection and steatosis remain ***,we show that HCV infected cells having higher levels of CD2 associated protein(CD2 AP),which plays two distinct,yet tightly-linked roles in HCV pathogenesis: elevated CD2 AP binding the domain III of NS5 A via its SH3 *** interaction transports NS5 A to LDs via an actin dependent manner to facilitate viral assembly;up-regulated CD2 AP interacting with E3 ligases cbl/cbl-c to degrade insulin receptor substrate 1(IRS1),which in turn disrupts insulin signaling and increases LDs accumulation thru the IRS1-Akt-AMPK-hormone sensitive lipase(HSL) signaling axis to accommodate viral *** HCV infected mouse model and human subjects,CD2 AP expression is up-regulated and the upregulation correlates well with liver *** importantly,hepatocellular carcinoma showed significantly stronger CD2 AP staining than peritumor ***: CD2 AP was identified as a protein upregulated by HCV infection,which in turn stimulated HCV assembly and steatosis by disrupting insulin signaling,targeting CD2 AP may offer an opportunity for alleviating HCV infection and its associated liver pathology.

健康老龄化是指人在维持良好身体功能、保证生活质量前提下的衰老过程。这个过程能否实现取决于机体固有能力,一方面包括心理、生理能力,另一方面包括机体所处环境以及二者的相互作用。在老龄化进程中,骨骼肌肉健康是维持老年人身体功能的重要条件。全球肌少症、骨质疏松症和骨关节炎等老龄化相关的骨骼肌肉疾病及机体失能所致的负担正在增加,因此随着人口老龄化的加剧,防治这类疾病也尤为重要。以此为契机,中华医学会、中华医学会骨质疏松和骨矿盐疾病分会、欧洲骨质疏松和骨关节炎临床经济学会联合开设论坛,共同探讨增龄性肌肉骨骼疾病的现行临床诊治策略。本次会议邀请了中国和欧洲的专家到场,分享这3种疾病的临床诊治经验;双方通过经验交流、讨论异同以取长补短,从而实现对疾病的更佳防治,维持老年人的自身能力、延缓老龄化带来的身体功能退化。展望未来,希望双方经验及最佳临床实践的交流能推进全球战略,以减轻肌肉骨骼疾病的负担,促进符合个体化需求的健康老龄进程。

AIM: To review methods of assessing adherence and strategies to improve adherence to cardiovascular disease(CVD) medications,among South Asian CVD patients. METHODS: We conducted a systematic review of English language studies that examined CVD medication adherence in South Asian populations from 1966 to April 1,2015 in SCOPUS and Pub Med. Working in duplicate,we identified 61 studies. After exclusions,26 studies were selected for full text review. Of these,17 studies were included in the final review. We abstracted data on several factors including study design,study population,method of assessing adherence and adherence rate. RESULTS: These studies were conducted in India(n = 11),Pakistan(n = 3),Bangladesh(n = 1),Nepal(n = 1) and sri Lanka(n = 1). Adherence rates ranged from 32%-95% across studies. Of the 17 total publications included,10 focused on assessing adherence to CVD medications and 7 focused on assessing the impact of interventions on medication adherence. The validated Morisky Medication Adherence Scale(MMAS) was used as the primary method of assessing adherence in five studies. Three studies used validated questionnaires similar to the MMAS,and one study utilized Medication Event Monitoring System caps,with the remainder of the studies utilizing pill count and self-report measures. As expected,studies using non-validated self-report measures described higher rates of adherence than studies using validated scale measurements and pill count. The included intervention studies examined the use of polypill therapy,provider education and patient counseling to improve medication adherence. CONCLUSION: The overall medication adherence rates were low in the region,which suggest a growing need for future interventions to improve adherence.

Background:Genetic testing for breast cancer susceptibility is widely used,but for many genes,evidence of an association with breast cancer is weak,underlying risk estimates are imprecise,and reliable subtype-specific risk estimates are ***:We used a panel of 34 putative susceptibility genes to perform sequencing on samples from 60,466 women with breast cancer and 53,461 *** separate analyses for protein-truncating variants and rare missense variants in these genes,we estimated odds ratios for breast cancer overall and tumor *** evaluated missense-variant associations according to domain and classification of pathogenicity.
