A fermented rapeseed meal additive:Effects on production performance,nutrient digestibility,colostrum immunoglobulin content and microbial flora in sows

This study was to assess the effect of fermented rapeseed meal(FRSM) in the diet of sows,taking into account the physiological period(pregnancy or lactation) and reproductive cycle(primiparous or multiparous sows),on production performance,nutrient digestibility,colostrum immunoglobulin content,and microbial flora in sows.The experimental material included 30 primiparous gilts and 30 multiparous sows after their second lactation.The animals in the control groups C_G(gilts) and Cs(sows)received a standard diet for pregnant or lactating sows,depending on the reproductive period.Experimental groups E_G and E_S comprised gilts and multiparous sows,respectively,receiving a diet with a 4%share of FRSM in place of soybean meal up to 100 d of gestation.In addition,from 100 d of gestation to7 d of lactation,the sows in experimental groups received a diet with a 9% share of FRSM,and then again a diet with a 4% share of FRSM until the end of lactation.The addition of 4% to 9% share of a FRSM component in feed significantly improves production parameters,mainly in primiparous gilts,leading to an increase in litter size and in litter weight at 28 d of age.It also helps to improve the digestibility of crude protein,fat,and crude fiber,and positively affects the gut microbiota of sows.Fermentation of rapeseed meal is an effective way to reduce anti-nutrients and to increase the level of lactic acid in the diet It also stimulates the immune system,which improves piglet health,reducing the severity of diarrhoea and mortality.

Aphrodisiac activity of aqueous extract of Anthonotha macrophylla P.Beauv.leaves in female Wistar rats

OBJECTIVE： This study investigated the aphrodisiac activity of aqueous extract of Anthonotha macrophylla leaves （AEAML） in female Wistar rats. METHODS： Thirty female rats were assigned into five groups of six animals each, such that rats in groups A, B, C, D and E received orally 1 mL of distilled water only, 5.7 mg/kg body weight （BW） of Exus Ginseng （a polyherbal formulation）, 25, 50 and 100 mg/kg BW of AEAML, respectively. After 1 h of treatment, female sexual behaviour parameters were monitored for 30 min. Luteinizing hormone （LH）, follicle stimulating hormone （FSH）, estradiol （E） and testosterone were also determined using standard methods. RESULTS： Six secondary metabolites were detected in AEAML with alkaloids （13.00 mg/L） being the highest. AEAML at 25 and 50 mg/kg BW significantly （P〈0.05） increased the darting frequency, hopping frequency, lordosis frequency, anogenital grooming, genital grooming and licking behaviour, whereas the darting latency, hopping latency and lordosis latency decreased （P〈0.05） in a manner comparable with Exus Ginseng. The 25 and 50 mg/kg BW of AEAML increased （P〈0.05） the serum concentrations of FSH, LH and E, like Exus Ginseng, whereas testosterone content was not altered （P〉0.05）. The 100 mg/kg BW of AEAML did not alter （P〉0.05） the sexual behaviour parameters, serum FSH, LH and E contents, whereas testosterone content increased significantly. The ovarian histology of the animals treated with Exus Ginseng, AEAML at 25 and 50 mg/kg BW revealed developing and ruptured follicles with numerous corpora lutea in the stroma, whereas the 100 mg/kg BW of AEAML produced fewer follicles and corpora lutea. CONCLUSION： The enhanced proceptive, receptive and orientation components of the female sexual behaviour by the 25 and 50 mg/kg BW of AEAML confers sexual invigorating potential on the plant. This study thus justifies the folkloric claim of the plant as a female aphrodisiac.

Inhibitory potentials of phenolic-rich extracts from Bridelia ferruginea on two key carbohydrate-metabolizing enzymes and Fe^(2＋)-induced pancreatic oxidative stress

Objective： The current study was designed to evaluate the various antioxidant potentials and inhibitory effects of phenolic-rich leaf extracts of Bridelia ferruginea（BF） on the in vitro activities of some key enzymes involved in the metabolism of carbohydrates.Methods： In this study, BF leaf free and bound phenolic-rich extracts were used. We quantified total phenolic and flavonoid contents, and evaluated several antioxidant activities using assays for ferric reducing antioxidant power, total antioxidant activity（phosphomolybdenum reducing ability）, 1,1-diphenyl-2-picrylhydrazyl and thiobarbituric acid reactive species. Also, extracts were tested for their ability to inhibit a-amylase and α-glucosidase activity.Results： The total phenolic and total flavonoid contents in the free phenolic extract of BF were significantly greater than in the bound phenolic extract. Also, all the antioxidant activities considered were significantly greater in the free phenolic extract than in the bound phenolic extract. In the same vein, the free phenolic-rich extract had a significantly higher percentage inhibition against α-glucosidase activity（IC50= 28.5 mg/mL） than the bound phenolic extract（IC50= 340.0 mg/mL）. On the contrary, the free phenolic extract（IC50= 210.0 mg/mL） had significantly lower inhibition against α-amylase than the bound phenolic-rich extract（IC50= 190.0 mg/mL）.Conclusion： The phenolic-rich extracts of BF leaves showed antioxidant potentials and inhibited two key carbohydrate-metabolizing enzymes in vitro.

Diet in irritable bowel syndrome: What to recommend, not what to forbid to patients.

A substantial proportion of patients with irritable bowel syndrome(IBS) associate their symptoms with the ingestion of specific foods. Therefore, in recent years, scientific research has increasingly focused on the role of diet in IBS and dietary management is now considered an important tool in IBS treatment. This article reviews the main dietary approaches in IBS emphasizing evidence from experimental and observational studies and summarizing the main diet and lifestyle recommendations provided by dietary guidelines and scientific literature. Despite the limited evidence for a beneficial role, general advice on healthy eating and lifestyle is recommended as the first-line approach in the dietary management of IBS. Standard recommendations include adhering to a regular meal pattern, reducing intake of insoluble fibers, alcohol, caffeine, spicy foods, and fat, as well as performing regular physical activity and ensuring a good hydration. Second-line dietary approach should be considered where IBS symptoms persist and recommendations include following a low FODMAP diet, to be delivered only by a healthcare professional with expertise in dietary management. The efficacy of this diet is supported by a growing body of evidence. In contrast, the role of lactose or gluten dietary restriction in the treatment of IBS remains subject to ongoing research with a lack of high-quality evidence. Likewise, further clinical trials are needed to conclude the efficacy of probiotics on IBS symptoms.

Parquetina nigrescens leaves： chemical profile and influence on the physical and biochemical indices of sexual activity of male Wistar rats

OBJECTIVE： The leaves of Parquetina nigrescens have been claimed in folk medicine to be useful for managing sexual dysfunction, but there is inadequate scientific evidence for this claim. This investigation was conducted to assess the effects of aqueous leaf extract of Parquetina nigrescens （AEPN） in rats induced with sexual dysfunction. METHODS： Male rats were allocated into various groups after being induced into sexual dysfunction with paroxetine hydrochloride. The groups were treated with distilled water, PowMaxM （reference drug）, and the AEPN at 20, 40 and 80 mg/kg body weight （BW） respectively for 7 d. Frequencies of mount （MF）, intromission （IF）, ejaculation （EF）, as well as latencies of mount （ML）, intromission （IL） and post - ejaculatory interval （PEI） were progressively monitored with receptive female rats on days 1, 3 and 7. The precentage of mounted, intromitted, and ejaculated rats, intromission ratio （IR）, copulatory efficiency （CE） and intercopulatory interval （ICI） were also computed. Standard methods were employed to determine the levels of serum luteinizing and follicle stimulating hormones （LH and FSH）, testosterone, nitric oxide （NO） and the activity of phosphodiesterase V （PDE5）. RESULTS： The plant contained alkaloids, saponins, flavonoids, cardiac glycosides, steroids, tannins, phlobatannins, cardenolides, phenolics, anthraquinones and triterpenes with alkaloids （2.32 mg/g） occurring in greatest quantity while flavonoids, anthraquinones and cardenolides （0.01 mg/g, each） were the least. All the 20 standard amino acids were detected in the plant leaf, with the lowest concentration being 0.30 mg/g for methionine and the highest being 2.12 mg/g for cysteine. Furthermore, P. nigrescens leaves contained Na, K, Fe, Mg, Zn, Ca, Cu, Mn, P, Pb, Cd, Ni, Cr, and Co, with Fe present in the highest and Co the lowest concentrations. Metabolic reprogramming in tumors:Contributions of the tumor microenvironment

The genetic alterations associated with cell transformation are in large measure expressed in the metabolic phenotype as cancer cells proliferate and change their local environment,and prepare for metastasis.Qualitatively,the fundamental biochemistry of cancer cells is generally the same as in the untransformed cells,but the cancer cells produce a local environment,the TME,that is hostile to the stromal cells,and compete for nutrients.In order to proliferate,cells need sufficient nutrients,either those that cannot be made by the cells themselves,or must be made from simpler precursors.However,in solid tumors,the nutrient supply is often limiting given the potential for rapid proliferation,and the poor quality of the vasculature.Thus,cancer cells may employ a variety of strategies to obtain nutrients for survival,growth and metastasis.Although much has been learned using established cell lines in standard culture conditions,it is becoming clear from in vivo metabolic studies that this can also be misleading,and which nutrients are used for energy production versus building blocks for synthesis of macromolecules can vary greatly from tumor to tumor,and even within the same tumor.Here we review the operation of metabolic networks,and how recent understanding of nutrient supply in the TME and utilization are being revealed using stable isotope tracers in vivo as well as in vitro.

Moringa oleifera-based diet protects against nickel-induced hepatotoxicity in rats

Multiple health-promoting effects have been attributed to the consumption of Moringa oleifera leaves,as part of diet without adequate scientific credence.This study evaluated the effect of M.oleifera-based diets on nickel（Ni）-induced hepatotoxicity in rats.Male rats assigned into six groups were given oral administration of 20 mg/kg body weight nickel sulfate in normal saline and either fed normal diet or M.oleifera-based diets for 21 days.All animals were sacrificed under anesthesia 24 hours after the last treatment.Ni exposure elevated the rat plasma activities of alanine transaminase,aspartate transaminase and alkaline phosphatase significantly.Ni exposure also raised the levels of triglyceride,total cholesterol and low-density lipoprotein cholesterol while depleting the high-density lipoprotein cholesterol concentration.Further,Ni exposure raised rat plasma malondialdehyde but depleted reduced glutathione concentrations.The histopathological presentations revealed inflammation and cellular degeneration caused by Ni exposure.We show evidence that M.oleifera-based diets protected against Ni-induced hepatotoxicity by improving the rat liver function indices,lipid profile as well as restoring cellular architecture and integrity.Study lends credence to the health-promoting value of M.oleifera as well as underscores its potential to attenuate hepatic injury.

Histopathological,histochemical and biochemical postmortem changes in induced fatal hypothermia in rats

Reaching a postmortem diagnosis of hypothermia is challenging in forensic practice.Therefore,this study was conducted to detect the histopathological,histochemical and biochemical changes that occur in adult albino rats following exposure to induced fatal hypothermia.Twenty-four adult albino rats were divided into the negative control,moderate hypothermia,severe hypothermia and hypoxia groups.Rats in the control group were euthanized when those in the moderate hypothermic group died.Blood samples were collected via heart puncture,and the cerebrum,heart,suprarenal gland,kidney,liver and skeletal muscle were removed to investigate the biochemical,histochemical and histopathological changes.Postmortem assessment depicted significant changes in lipid peroxidation,represented by increased malondialdehyde levels in the studied organs of the rats in hypothermic and hypoxia groups.Histopathological examination of the rats’organs revealed degeneration and necrosis in the hypothermia and hypoxia groups.Sections taken from the severe hypothermic rats revealed a loss of normal cardiac tissue architecture,necrotic changes in the pyramidal cells in the cerebral cortex,and massive necrosis,mainly in the tubules of the renal cortex and medulla.These findings suggest that histological changes might be used as biochemical markers for postmortem diagnosing of fatal hypothermia,particularly in severe hypothermic conditions.

Male infertility has become a problem worldwide,and recent research has emphasized the development of more effective therapy options.Among natural compounds,rutin has been widely studied for its potential to treat dysfunction related to male infertility,including a reduction in sperm quality,spermatogenesis disruption and structural disruption in the testis.A thorough review of scientific literature published in several databases,including Google Scholar,PubMed/MEDLINE and Scopus,was used to synthesize the present state of research on the role of rutin in male reproductive health.Rutin has been shown to possess antiapoptotic,antioxidant and anti-inflammatory activities,among others,which are crucial in the management of male infertility.Numerous investigations have shown that rutin protects against male infertility and have explored the underlying mechanisms involved.The present review,therefore,assesses the therapeutic mechanisms involved in male infertility treatment using rutin.Rutin was able to mitigate the induced oxidative stress,apoptosis,inflammation,and related physiological processes that can cause testicular dysfunction.

Antifertility effects of 60-day oral gavage of ethanol extract of Spondias mombin leaves in guinea pigs: A biochemical, reproductive and histological study

Objective:To investigate the impact of ethanol extract of Spondias(S.)mombin leaves on the biochemical parameters of testicular function,hormonal profile,sperm profile and reproductive outcomes of guinea pigs.Methods:Sexually matured male[(1000.40±8.12)g]and female[(810.00±7.09)g]guinea pigs were used.In testicular function study,a total of 24 male guinea pigs were randomised into 4 groups of 6 guinea pigs each.Group A(control)was orally administered 1 mL of physiological saline,once daily for 60 days while groups B,C and D were treated like the control group except that they were orally administered 100,250 and 500 mg/kg body weight of ethanol extract of S.mombin leaves.For the fertility study,the same animal groupings and treatments in the testicular function study were adopted.The male guinea pigs were paired with the females(1:1)and afterwards examined for pregnancy outcomes.Results:The ethanol extract of S.mombin leaves contained saponins,alkaloids,flavonoids,tannins,steroids,phenolics,phlobatannins,cardiac glycosides,cardenolides and dienolides with saponins(4.80 mg/mL)occurring the most whilst cardenolides and dienolides(0.08 mg/mL)were the least.The ethanol extract of S.mombin leaves significantly and dose dependently reduced the activities of alkaline phosphatase,glutamate dehydrogenase,3-hydroxy-3-methylglutaryl coenzyme A reductase,malic enzyme,17-β-hydroxysteroid dehydrogenase,lactate dehydrogenase,catalase and superoxide dismutase as well as levels of testosterone,glycogen,total protein and ascorbic acid in the testes when compared with the control group(P<0.05).All the doses of ethanol extract of S.mombin leaves also reduced the levels of sorbitol dehydrogenase,3-β-hydroxysteroid dehydrogenase,glucose-6-phosphate dehydrogenase and sialic acid whereas the levels of testicular acid phosphatase,gamma glutamyl transferase and cholesterol increased dose dependently(P<0.05).The serum luteinising hormone,testosterone and estradiol were reduced after the administration of ethanol extract of S.mombin leaves
